Neuronal action potentials are generated through voltage-gated sodium channels, which are tethered by ankyrinG at the membrane of the axon initial segment (AIS). Despite the importance of the AIS in the control of neuronal excitability, the cellular and molecular mechanisms regulating sodium channel expression at the AIS remain elusive. Our results show that GSK3α/β and β-catenin phosphorylated by GSK3 (S33/37/T41) are localized at the AIS and are new components of this essential neuronal domain. Pharmacological inhibition of GSK3 or β-catenin knockdown with shRNAs decreased the levels of phosphorylated-β-catenin, ankyrinG, and voltage-gated sodium channels at the AIS, both “in vitro” and “in vivo”, therefore diminishing neuronal excitability as evaluated via sodium current amplitude and action potential number. Thus, our results suggest a mechanism for the modulation of neuronal excitability through the control of sodium channel density by GSK3 and β-catenin at the AIS. 相似文献

The diamondback moth ( Plutella xylostella [L.]), is one of the most economically important pests of cruciferous plants throughout the world, causing economically significant damage in broccoli, cabbage, canola, collards, and mustard. In Canada, P. xylostella is an important economic pest of canola ( Brassica napus L. and Brassica rapa L.). In North America, the diamondback moth has been collected as far north as Edmonton, Alberta, Canada. Despite its wide distribution and presence at economically damaging levels in Canada, there have been no published reports of P. xylostella from Alaska. This note documents the 1st collection of this pest species in interior Alaska. With continued moderation of the climate due to global climate change, this species has the potential to become an increasingly important agricultural pest in the state. 相似文献

The main focus of this paper is to model the daily series of banknotes in circulation. The series of banknotes in circulation displays very marked seasonal patterns. To the best of our knowledge the empirical performance of two competing approaches to model seasonality in daily time series, namely the ARIMA‐based approach and the Structural Time Series approach, has never been put to the test. The application presented in this paper provides valid intuition on the merits of each approach. Palmitate activates the NF-κB pathway, and induces accumulation of lipid metabolites and insulin resistance in skeletal muscle
cells. Little information is available whether and how these processes are causally related. Therefore, the objectives were
to investigate whether intra-cellular lipid metabolites are involved in FA-induced NF-κB activation and/or insulin resistance
in skeletal muscle and to investigate whether FA-induced insulin resistance and NF-κB activation are causally related. Inhibiting
DGAT or CPT-1 by using, respectively, amidepsine or etomoxir increased DAG accumulation and sensitized myotubes to palmitate-induced
insulin resistance. While co-incubation of palmitate with etomoxir increased NF-κB transactivation, co-incubation with amidepsine
did not, indicating that DAG accumulation is associated with insulin resistance but not with NF-κB activation. Furthermore,
pharmacological or genetic inhibition of the NF-κB pathway could not prevent palmitate-induced insulin resistance. In conclusion,
we have demonstrated that activation of the NF-κB pathway is not required for palmitate-induced insulin resistance in skeletal

The oceanic overturning circulation has a central role in the Earth's climate system and in biogeochemical cycling, as it transports heat, carbon and nutrients around the globe and regulates their storage in the deep ocean. Mixing processes in the Antarctic Circumpolar Current are key to this circulation, because they control the rate at which water sinking at high latitudes returns to the surface in the Southern Ocean. Yet estimates of the rates of these processes and of the upwelling that they induce are poorly constrained by observations. Here we take advantage of a natural tracer-release experiment-an injection of mantle helium from hydrothermal vents into the Circumpolar Current near Drake Passage-to measure the rates of mixing and upwelling in the current's intermediate layers over a sector that spans nearly one-tenth of its circumpolar path. Dispersion of the tracer reveals rapid upwelling along density surfaces and intense mixing across density surfaces, both occurring at rates that are an order of magnitude greater than rates implicit in models of the average Southern Ocean overturning. These findings support the view that deep-water pathways along and across density surfaces intensify and intertwine as the Antarctic Circumpolar Current flows over complex ocean-floor topography, giving rise to a short circuit of the overturning circulation in these regions. 相似文献

P systems have been used many times to face with computationally difficult problems, such as NP-complete decision problems and NP-hard optimization problems. In this paper we focus our attention on another computationally intractable problem: factorization. In particular, we first propose a simple method to encode binary numbers using multisets. The beneficiation methods for Ethiopian Kenticha pegmatite-spodumene ores were assessed through mineralogical and quantitative analyses with X-ray diffraction (XRD) and energy-dispersive X-ray fluorescence (EDXRF). The tantalite in the upper zone of the Kenticha pegmatite-spodumene deposit is 58.7wt% higher than that in the inner zone. XRD analysis revealed that the upper zone is dominated by manganocolumbite, whereas the inner zone is predominantly tantalite-Mn. Repeated cleaning and beneficiation of the upper-zone ore resulted in concentrate compositions of 57.34wt% of Ta2O5 and 5.41wt% of Nb2O5. Washing the tantalite concentrates using 1vol% KOH and 1 M H2SO4 led to the removal of thorium and uranium radioactive oxides from the concentrate. The findings of this study suggest that the beneficiation and alkaline washing of Kenticha pegmatite-spodumene ores produce a high-grade export-quality tantalite concentrate with negligible radioactive oxides. 相似文献

The paper reviews the development of management science/operational Research (MS/OR) in management education and explore how far the syllabuses of undergraduate business courses and MBA courses in the UK, are giving space to the teaching of management sciences methodologies (MSM). The academic phase of early MS/OR is outlined and a time-based framework to map the evolution of four MS/OR discourses informing a set of MSMs currently in use is advanced. At an undergraduate level the review is based on a sample taken from Universities and Colleges Admissions Services (UCAS) of business and management studies degrees and at a postgraduate level the review considers a sample of MBA programmes offered by six UK universities. Results indicate that most of the MBA courses include a core unit in quantitative methods in the first year and that universities are still at large teaching the ??hard?? end of the MSM spectrum, the type of problem solving methods developed in the 1950s and 1960s. Business courses incorporating units containing systemic thinking and management sciences methodologies developed from the 1970s to now (e.g.: soft systems methodology, viable systems model, cognitive mapping), are rarely taught at undergraduate (UG) level; and at post-graduate (PG) level they have been confined to a handful of institutions in the UK. The paper highlights the fact that MSMs associated with the soft and critical end of the spectrum are still lacking of credibility amongst the designers of management education at both undergraduate and graduate management education. A Soft Systems Methodology (SSM) Based Framework for Evaluating Managed Learning Environments 总被引:1,自引:1,他引:0
Managed Learning Environments (MLEs) in higher education institutions (HEIs) are relatively new to the arena of higher education, even though there are over 90% of institutions in the higher and further education sector who are currently engaged in some kind of MLE development activity (University of Brighton 2003). However, when it comes to the task of assessing the performance of an MLE there are no universally recognisable frameworks for evaluating MLEs in HEIs currently discussed within the literature. The paper advances a general systemic framework for evaluating MLEs based on Checkland’s SSM and reports on the first stages of our attempt to evaluate the MLE at Manchester Metropolitan University involving the team developing the system and the stakeholders concerned. Two of three iterations of this research have been completed and, whilst outside the scope of this paper, have found that SSM has coped with the criteria demanded of the evaluation framework within its context. After completing a stakeholder analysis, the criteria for evaluating an MLE, based on the stakeholders’ requirements, emerged. These iterations have tentatively concluded that by contextualising SSM to the evaluation requirements of an MLE in a UK HEI, the measures of performance suggested by SSM need to be adjusted.
