Mutations in KERA, encoding keratocan, cause cornea plana

Specialized collagens and small leucine-rich proteoglycans (SLRPs) interact to produce the transparent corneal structure. In cornea plana, the forward convex curvature is flattened, leading to a decrease in refraction. A more severe, recessively inherited form (CNA2; MIM 217300) and a milder, dominantly inherited form (CNA1; MIM 121400) exist. CNA2 is a rare disorder with a worldwide distribution, but a high prevalence in the Finnish population. The gene mutated in CNA2 was assigned by linkage analysis to 12q (refs 4, 5), where there is a cluster of several SLRP genes. We cloned two additional SLRP genes highly expressed in cornea: KERA (encoding keratocan) in 12q and OGN (encoding osteoglycin) in 9q. Here we report mutations in KERA in 47 CNA2 patients: 46 Finnish patients are homozygous for a founder missense mutation, leading to the substitution of a highly conserved amino acid; and one American patient is homozygous for a mutation leading to a premature stop codon that truncates the KERA protein. Our data establish that mutations in KERA cause CNA2. CNA1 patients had no mutations in these proteoglycan genes.

Zusammenfassung Basierend auf dem speziellen Betragen der isolierten Noradrenalingranula der inneren männlichen Geschlechtsdrüsen, die von kurzen adrenergischen Neuronen innerviert sind, werden Definitionen für 4 «übergangsformen» der peripheren katecholamin-enthaltenden Zellen diskutiert: chromaffine Zellen der Adrenomedulla, kleine verzweigte, nicht adrenale chromaffine Zellen,kurze adrenergische Neurone undlange adrenergische Neurone.     

Viscero-somatic reflexes following distension of urinary bladder in cats: Role of supraspinal neuraxis

Summary Viscero-somatic reflexes have been studied by recording monosynaptic reflexes following distension of the urinary bladder in intact, decerebrate and spinal animals. It was observed that the viscero-somatic responses following bladder distension are inhibitory in nature and this inhibition was highest in decerebrates and least in spinal animals. The site of viscero-somatic interaction probably lies in the bulbar area (supraspinal) and spinal cord.Acknowledgment. Monoamine pathways to the cerebellum and cerebral cortex

Zusammenfassung In kombinierter histochemischer und biochemischer Untersuchung werden Dopamin-, Noradrenalin-und 5-Hydroxytryptamin-Neurone nach Entfernung von Cortex cerebri und Cerebellum studiert. Es ergibt sich, dass die Noradrenalin-Nerventerminale, welche Cortex cerebri und Cerebellum innervieren, vermutlich von feinen Axonen her stammen, deren Zellkörper mindestens zum Teil in der Formatio reticularis der Medulla oblongata gelegen sind. Die Noradrenalin-und 5-Hydroxytryptamin-Axone, die nach dem Cortex cerebri ziehen, passieren vermutlich zur Hauptsache das Cingulum.     

A rapid micro radial electrophoretic method of protein separation on cellulose acetate membranes.

A new rapid micromethod for protein separation under a radial electric field is described. As many as 12 rabbit serum samples could be separated in 4--6 min.     

Comparative evaluation of 7 helminth antigens in the enzyme-linked immunosorbent assay (E.L.I.S.A.).

112 sera from Europeans with parasitologically proven helminthiasis were tested in the enzyme-linked immunosorbent assay (E.L.I.S.A.) against 6 crude extracts of various helminths (2 of adult worms: Dipetalonema viteae, Fasciola hepatica; 3 of eggs: Ascaris suum, Toxocara canis, Schistosoma mansoni; and of Echinococcus granulosus scolices) and against bovine hydatid fluid. Each serum was tested simultaneously at a fixed dilution of 1:160 against all antigens. Extensive cross-reactions were observed, leading to the conclusion that non-purified helminth antigens, even in combination, are of limited value for reliable serodiagnosis in E.L.I.S.A.     

Suppression of immunization to tuberculosis and diphtheria by an extract of lymphoid tissue

Zusammenfassung Dank einer neuen Anwendung von Lymphknotenextrakten (statt Röntgenextrakten, Corticosteroiden, Antilymphozytenserum usw.) wird eine erhebliche Reduktion der immunologischen Reaktion erzielt, wenn die Tiere mit diesen Extrakten vorbehandelt und gleichzeitig mit Antigenen behandelt werden.
