The development of nineteenth-century geodetic measurement challenges the dominant coherentist account of metric success. Coherentists argue that measurements of a parameter are successful if their numerical outcomes convergence across varying contextual constraints. Aiming at numerical convergence, in turn, offers an operational aim for scientists to solve problems of coordination. Geodesists faced such a problem of coordination between two indicators of the earth's polar flattening, which were both based on imperfect ellipsoid models. While not achieving numerical convergence, their measurements produced novel data that grounded valuable theoretical hypotheses. Consequently, they ought to be regarded as epistemically successful. This insight warrants a dynamic revision of coherentism, which allows to judge the success of a metric based on both its coherence and fruitfulness. On that view, scientific measurement aims to coordinate theoretical definitions and produce novel data and theoretical insights.  相似文献

The low regeneration potential of the central nervous system (CNS) represents a challenge for the development of new therapeutic strategies. Mesenchymal stem cells (MSCs) have been proposed as a possible therapeutic tool for CNS disorders. In addition to their differentiation potential, it is well accepted nowadays that their beneficial actions can also be mediated by their secretome. Indeed, it was already demonstrated, both in vitro and in vivo, that MSCs are able to secrete a broad range of neuroregulatory factors that promote an increase in neurogenesis, inhibition of apoptosis and glial scar formation, immunomodulation, angiogenesis, neuronal and glial cell survival, as well as relevant neuroprotective actions on different pathophysiological contexts. Considering their protective action in lesioned sites, MSCs’ secretome might also improve the integration of local progenitor cells in neuroregeneration processes, opening a door for their future use as therapeutical strategies in human clinical trials. Thus, in this review we analyze the current understanding of MSCs secretome as a new paradigm for the treatment of CNS neurodegenerative diseases.  相似文献

Plant immune responses are usually accompanied by the production of extracellular superoxide at and surrounding infection sites. Extracellular reactive oxygen intermediates (ROIs) in plants were proposed to drive programmed cell death correlated with disease resistance (the hypersensitive response). ROIs derived from this oxidative burst are generated by plasma membrane NADPH oxidases, anchored by gp91(phox) proteins related to those responsible for the respiratory oxidative burst activated in mammalian neutrophils during infection. Mutation of Arabidopsis thaliana respiratory burst oxidase (Atrboh) genes eliminated pathogen-induced ROI production but had only a modest effect on the hypersensitive response. We show that Atrboh function can be activated by exogenous ROIs. Unexpectedly, the subsequent oxidative burst can suppress cell death in cells surrounding sites of NADPH oxidase activation. This cell death requires salicylic acid, a plant immune system activator. Thus, ROIs generated by Atrboh proteins can antagonize salicylic acid-dependent pro-death signals. These results have implications for understanding how salicylic acid activates defense signaling in cells spatially removed from infection sites without causing cell death.  相似文献

ABSTRACT

Few studies have focused on the diverse fauna of southwestern Brazilian Amazonia. This region, spared from large-scale human occupation until the second half of the twentieth century, has been threatened by expanding agriculture, logging, and mining. Here, we describe a new nurse frog (Allobates, Aromobatidae) from the open highland habitats of Serra dos Pacaás Novos, one of the few large remnants of relatively intact native vegetation in the Brazilian state of Rondônia. The new species is diagnosed by its metallic orange dorsal colouration, weakly expanded discs on hands, and presence of two subarticular tubercles on Finger IV. A phylogenetic analysis based on DNA sequence data recovered the new species as more closely related to nurse frogs from the Madeira and Tapajós River basins; this pattern may reflect a history of dispersal uphill from a lowland ancestor. Our results also indicate that the generic assignment of Allobates alessandroi (Grant and Rodriguez, 2001), a high-elevation species from the Andes of Peru, needs to be reassessed; we discuss the phylogenetic affinities of this species based on morphological attributes.

对松材线虫与其在葡萄牙和欧洲的媒介昆虫樟子松墨天牛(Monochamus galloprovincialis)进行了首次研究。在病死木中发现,松材线虫在天牛刚羽化时聚集在蛹室中,线虫主要集中在天牛的后胸部。天牛羽化后6周内,松材线虫主要在天牛成虫补充营养时进入新寄主树,其次在天牛产卵时也会进入寄主树。总的看来,松材线虫在欧洲与其携带天牛之间互作与其在亚洲与松墨天牛及在北美与卡来罗纳墨天牛之间的关系十分相似。经研究,影响线虫与天牛携带关系的因子有内源和外源两类因子,如化学气味和各种信号。但是,其他多数因子尚未了解。研究影响线虫与天牛携带关系的因子将有助于了解两种生物密切关系的内在机制,并有助于研发出新的防控方法。  相似文献

The hippocampus is necessary for the acquisition and retrieval of declarative memories. The best-characterized sensory input to the hippocampus is the perforant path projection from layer II of entorhinal cortex (EC) to the dentate gyrus. Signals are then processed sequentially in the hippocampal CA fields before returning to the cortex via CA1 pyramidal neuron spikes. There is another EC input-the temporoammonic (TA) pathway-consisting of axons from layer III EC neurons that make synaptic contacts on the distal dendrites of CA1 neurons. Here we show that this pathway modulates both the plasticity and the output of the rat hippocampal formation. Bursts of TA activity can, depending on their timing, either increase or decrease the probability of Schaffer-collateral (SC)-evoked CA1 spikes. TA bursts can also significantly reduce the magnitude of synaptic potentiation at SC-CA1 synapses. The TA-CA1 synapse itself exhibits both long-term depression (LTD) and long-term potentiation (LTP). This capacity for bi-directional plasticity can, in turn, regulate the TA modulation of CA1 activity: LTP or LTD of the TA pathway either enhances or diminishes the gating of CA1 spikes and plasticity inhibition, respectively.  相似文献

在韧性断裂中微观孔洞演化机制的基础上,提出了一个基于孔洞演化机制的非耦合型韧性断裂预测模型.模型充分考虑了两种典型的孔洞演化机制:孔洞的长大机制和孔洞的拉长扭转机制.该模型引入了三个具有不同物理意义的材料参数:材料对不同孔洞演化机制的敏感度、应力状态敏感度系数和材料的损伤阈值,并使用等效塑性应变增量表征其对韧性损伤累积过程的驱动作用.为了使模型可以更好地反映三维应力状态对材料韧性断裂性能的影响,将该模型从主应力空间转换到由应力三轴度、罗德参数和临界断裂应变构成的三维空间,得到了由模型确定的三维韧性断裂曲面,并研究了相关参数对三维韧性断裂曲面及平面应力二维韧性断裂曲线的影响.利用5083-O铝合金、TRIP690钢和Docol 600DL双相钢三个典型的轻质高强板材的韧性断裂数据验证了该模型对不同材料和不同应力状态的适用性和准确性.  相似文献

Female multiple mating and alternative mating systems can decrease the opportunity for sexual selection. Sperm competition is often the outcome of females mating with multiple males and has been observed in many animals, and alternative reproductive systems are widespread among species with external fertilization and parental care. Multiple paternity without associated complex behaviour related to mating or parental care is also seen in simultaneously spawning amphibians and fishes that release gametes into water. Here we report 'clutch piracy' in a montane population of the common frog Rana temporaria, a reproductive behaviour previously unknown in vertebrates with external fertilization. Males of this species clasp the females and the pair deposits one spherical clutch of eggs. No parental care is provided. 'Pirate' males search for freshly laid clutches, clasp them as they would do a female and fertilize the eggs that were left unfertilized by the 'parental' male. This behaviour does not seem to be size-dependent, and some males mate with a female and perform clutch piracy in the same season. Piracy affected 84% of the clutches and in some cases increased the proportion of eggs fertilized, providing direct fitness benefits both for the pirate males and the females. Sexual selection--probably caused by a strong male-biased sex ratio--occurs in this population, as indicated by size-assortative mating; however, clutch piracy may reduce its impact. This provides a good model to explore how alternative mating strategies can affect the intensity of sexual selection.  相似文献
