Assembly and function of a bacterial genotoxin

The tripartite cytolethal distending toxin (CDT) induces cell cycle arrest and apoptosis in eukaryotic cells. The subunits CdtA and CdtC associate with the nuclease CdtB to form a holotoxin that translocates CdtB into the host cell, where it acts as a genotoxin by creating DNA lesions. Here we show that the crystal structure of the holotoxin from Haemophilus ducreyi reveals that CDT consists of an enzyme of the DNase-I family, bound to two ricin-like lectin domains. CdtA, CdtB and CdtC form a ternary complex with three interdependent molecular interfaces, characterized by globular, as well as extensive non-globular, interactions. The lectin subunits form a deeply grooved, highly aromatic surface that we show to be critical for toxicity. The holotoxin possesses a steric block of the CdtB active site by means of a non-globular extension of the CdtC subunit, and we identify putative DNA binding residues in CdtB that are essential for toxin activity.     

甘蓝型杂交油菜"蜀杂9号"种子纯度的RAPD鉴定

通过50个随机引物对“蜀杂9号”亲本进行RAPD扩增,选出了两个能在父、母本间产生多态性扩增的随机引物S18和S31,在杂种F,代中验证了它们的特异性和稳定性,证明随机引物S18和S31能分别在F1代中稳定扩增出父本和母本的特异标记片段S18750与S31550．对该引物的RAPD-PCR的反应体系优化后,对“蜀杂9号”父、母本和杂种F1代的单株进行了随机检测,结果表明这两个RAPD标记配合使用能稳定可靠地鉴定甘蓝型杂交油菜“蜀杂9号”种子的纯度．     

Analysis of components of conserved “backbone sequences”among genomes of Shigella spp. strains

Difference in the genomic compositions of prokaryotes is the basis of the diversity in their biological characters. However, besides their flora-or strain-specific genes,those floras with closer relationship in the evolution also have conserved “backbone sequences”, which reveal the marks of their origin and evolution, and these “backbone sequences”are just the basis of their elementary living abilities and common biological properties. Shigella is very closely related to E. coli in the origin and evolution, and may turn out to belong to the same genus. In this study, a microarray containing E. coli K-12 whole genome and Sf301 specific ORFs is used to investigate the genomic components of four Shigella strains. The results indicate that 16%-22% K-12 ORFs sequences are not detected in the genome of Shigella strains while the genome of Shigella contains at least 2800 conserved ORFs, which compose the common “backbone sequences”.Advanced analysis indicated that the “backbone sequences”are the essential components in maintaining the normal physiological activities of intestinal bacteria. Furthermore,only 20% Sf301-specific ORFs exist in other strains simultaneously, which demonstrate the great genome heterogeneity and the genetic diversity among the strains.

基于元评估的思想,利用肯德尔和谐系数、克朗巴赫系数、内容效度比等分析方法对政府绩效评估的主体一致性信度、评估指标体系信度与效度、评估数据测量信度、评估结果效度进行检测,为控制政府绩效评估偏差,提高评估活动的权威性、科学性和有效性提供了一个可行的途径。     

Integrated semantic similarity model based on ontology

To solve the problem of the inadequacy of semantic processing in the intelligent question answering system. an integrated semantic similarity model which calculates the semantic similarity using the geometric distance and informarion content is presented in this paper. With the help of interrelationship between concepts, the information content ofconcepts and the strength of the edges in the ontology network. we can calculate the semantic similarity between two concepts and provide information for the further calculation of the semantic similarity between user‘s question and answers in knowlegdge base. The results of the experiments on the prototype have shown that the semantic problem in natural language processing can also be solved with the help of the knowledge and the abundant semantic information in ontology. More than 90% accuracy with less than 50 ms average searching time in the intelligent question answering prototype system based on ontology has been reached. The result is vety satisfied.

经济增长的复杂性与“J”结构

从现代经济增长的事实和复杂性角度讨论了经济增长的原因和机制 .“J”结构是我们提出的一种经济增长复杂性的重要模式 ,这种具有潜在后发性优势但又需要先期投入的经济现象普遍存在于国际贸易、金融、教育、人力资源、生态环境等不同领域 .由于增长的复杂性 ,“J”结构及与之相对应的“J”过程或“J”效应往往被波动所掩盖 .本文进一步从物理、数学的角度指出“J”过程的本质是系统存在多重均衡时的非平衡相变过程 .最后 ,讨论了宏观经济系统中宏观变量之间的相互关系 ,“J”结构体现了这些宏观变量之间非线性相互作用机制.     

一种适用于软件雷达系统的数据流驱动机制

针对雷达信号处理各功能模块之间显著的流水性特征 ,结合一个通用的雷达信号处理系统———由清华大学研制的通用高速实时雷达信号处理系统的实验样机DSM(dataflow ,sharedmemoryandmultiplebusinterconnec tion) ,在研究现有静态、动态数据流驱动机制特点的基础上 ,提出了一种新型的适用于软件雷达系统的进程间准动态数据流驱动机制 ,并针对一个雷达信号处理的范例程序说明了编译器数据流驱动表的生成和系统数据流驱动的过程。     

新一代开放系统及互操作性研究

经历 2 0世纪 90年代的持续实践 ,采用“开放系统”的思想来集成大型的计算机系统已成为技术界不争的事实。然而 ,技术的发展使得传统的开放系统面临严峻的挑战 ,需要新的标准、技术来使开放系统体现高度的智能协作性及移动性。基于完善的开放系统的实际形成有赖于“互操作性”的实现这一原因 ,以互操作性为研究对象 ,以移动agent为研究工具 ,探讨了新一代开放系统实现机制以及未来发展的前景。
