Physiological state of submitochondrial particles and their susceptibility to Triton X-100

Summary The solubilizing effect of Triton X-100 on beef heart submitochondrial particles (ETP_H) has been studied under various physiological conditions. Coupled, uncoupled and azide-inhibited ETP_H particles have been studied. Quantitative and qualitative differences are found in the proteins solubilized by the detergent from ETP_H particles under the various conditions tested.Acknowledgments: This work was supported in part by a grant from a Spanish Comisión Asesora para la Investigación Cientifica y Técnica. Isolation and partial characterization of cuticular collagen from the parasitic nematodeGaigeria pachyscelis

Summary The cuticle from adultGaigeria pachyscelis was isolated by solubilizing the internal tissues with sodium dodecyl sulphate (SDS) at 37°C. Cuticular protein was extracted with guanidine-HCl and -mercaptoethanol and purified by ammonium sulphate fractionation and DEAE-cellulose chromatography. SDS-polyacrylamide gel electrophoresis of purified protein revealed 2 polypeptides with apparent mol. wts of 58,000 and 74,000. As judged from their hydroxyproline content both of them are collagenous in nature. Results of gel filtration indicate that cuticular collagen exists in two forms, a non-associated form at low concentration and an associated form at high concentration.Acknowledgments. Effect of hormones and cyclic AMP on γ-glutamyltranspeptidase activity of rat mammary gland explants

Summary -Glutamyl transpeptidase activity was assayed in midpregnant rat mammary gland explants at 14, 22 and 38 h, in the presence and absence of insulin, prolactin and corticosterone. With these 3 hormones the explants attained the characteristics of a secretory gland after 22 h of tissue culture, at which time the enzyme exhibited its maximal activity. The addition of dibutyryl cyclic AMP in the presence of the 3 hormones produced a significant increase in enzyme activity, which was maximal with a 1 mM concentration of the cyclic nucleotide. A similar effect was observed when theophylline or theophylline plus dibutyryl cyclic AMP were added to the culture medium.This work was supported by grant B1138-8333 from the Departamento de Desarrollo de la Investigación, Universidad de Chile.     

Estrogen and accessory sex gland lipids

Summary Administration of estradiol-17 elicited differential responses on accessory sex glands of rats. In caput epididymis, the estrogen treatment led to an accumulation of glycerides and phospholipids while in cauda epididymis, there was no significant change. However, in seminal vesicles, depletion of phospholipids was observed In prostate, the treatment, resulted in an accumulation of glycerides.Part of Ph.D. Thesis. Umapathy, E., Thesis. Action ofβ-(4-chlorophenyl-GABA) on uptake and metabolism of GABA in different subcellular fractions of rat brain

Summary -(4-chlorophenyl)-GABA, a GABA mimetic compound, acts as an inhibitor of GABA metabolism in both synaptosomal and extrasynaptosomal compartments. It has no significant action on GABA or Glu uptake by synaptosomes.     

Microbiological transformation of biflavone

Summary Incubation of 5,5-7,7-tetramethoxy-8,8-biflavone withAspergillus niger results in the formation of 4,4-dihydroxy-5,5-7,7-tetramethoxy-8,8-biflavan.Part of this work was done at Allahabad University, Chemical Laboratories.     

Effect of dibutyryl cyclic AMP and analogs on the rate of contractions of myocytes in culture

Summary ²O,⁶N-butyryl, 3, 5-cyclic monophosphate (dibu cAMP) when added to fetal rat heart cells in culture inhibits myocyte contraction. This inhibition is 100, 84 and 51% complete when the dibu cAMP concentration used is 2, 0.2 and 0.02 mM, respectively. The potency of dibu cAMP derivatives in myocyte contraction inhibition follows the order, dibu cAMP> ⁶N-bu cAMP> ²O-bu cAMP=AMP>butyrate. The inhibition caused by the first three chemicals is greater than 70%.The author was a Moss Heart Fellow and acknowledges the technical assistance of Ms Martha Peet. Action of glutamic acid and of some glutamate analogues on the molluscan central neurones

Summary The effects of L-glutamic acid and of some glutamate analogues have been studied on the central nervous system of the snailHeobania vermiculata, using conventional electrophysiological techniques. The glutamate H-response had the mean equilibrium value of-(57±4) mV and was associated with a Cl^– conductance change. The D-response to glutamate application showed an involvement of sodium ions. Aspartate was agonist of glutamate action and displayed similar equilibrium value of the H-response, whereas quisqualate H-response was non-invertible.The authors wish to express their thanks to Prof. A. New toxins from Aspergillus fumigatus Fresenius]

A strain of Aspergillus fumigatus Fres., isolated from sugar-beet draffs, synthesizes in vitro four toxic metabolites which have not yet been described in these fungal species. Toxic effects of the most abundant of these metabolites "fumitoxin A" have been studed on chick embryo. Artemia salina larvae, cell cultures, and on mice.
