Biological invasions are one of the greatest threats to native species in natural ecological systems. One of the most successful invasive species is Bromus tectorum L. (cheatgrass), which is having marked impacts on native plant communities and ecosystem processes. However, we know little about the effects of this invasion on native animal species in the Intermountain West. Because ants have been used to detect ecological change associated with anthropogenic land use, they seem well suited for a preliminary evaluation of the consequences of cheatgrass-driven habitat conversion. In our study, we used pitfall traps to assess ant community assemblages in intact sagebrush and nearby cheatgrass-dominated vegetation. Ant abundance was about 10-fold greater in cheatgrass-dominated plots than in sagebrush plots. We also noted differences in diversity and evenness between habitat types at both the species and the functional-group levels of organization. At the species level, Shannon’s diversity index was greater in sagebrush plots than in cheatgrass-dominated plots. However, at the functional-group level, Simpson’s and Shannon’s diversity indices and the Brillouin evenness index were greater in cheatgrass-dominated plots than in sagebrush plots. Further, common species / functional groups tended to be more abundant while less common species / functional groups tended to be less abundant in cheatgrass-dominated plots compared to intact sagebrush plots. Patterns appear to be at least partially related to resource availabilities. This initial survey of ant communities from intact-native and altered vegetation types may be indicative of similar trends of biodiversity shifts throughout the Intermountain West where cheatgrass has successfully replaced native species. We also discuss the implications of ant communities on land management activities, specifically in the context of aridland restoration. 相似文献

To identify the genetic bases for nine metabolic traits, we conducted a meta-analysis combining Korean genome-wide association results from the KARE project (n = 8,842) and the HEXA shared control study (n = 3,703). We verified the associations of the loci selected from the discovery meta-analysis in the replication stage (30,395 individuals from the BioBank Japan genome-wide association study and individuals comprising the Health2 and Shanghai Jiao Tong University Diabetes cohorts). We identified ten genome-wide significant signals newly associated with traits from an overall meta-analysis. The most compelling associations involved 12q24.11 (near MYL2) and 12q24.13 (in C12orf51) for high-density lipoprotein cholesterol, 2p21 (near SIX2-SIX3) for fasting plasma glucose, 19q13.33 (in RPS11) and 6q22.33 (in RSPO3) for renal traits, and 12q24.11 (near MYL2), 12q24.13 (in C12orf51 and near OAS1), 4q31.22 (in ZNF827) and 7q11.23 (near TBL2-BCL7B) for hepatic traits. Cancer stem cells have been hypothesized to drive the growth and metastasis of tumors. Because they need to be targeted for cancer treatment, they have been isolated from many solid cancers. However, cancer stem cells from primary human gastric cancer tissues have not been isolated as yet. For the isolation, we used two cell surface markers: the epithelial cell adhesion molecule (EpCAM) and CD44. When analyzed by flow cytometry, the EpCAM+/CD44+ population accounts for 4.5% of tumor cells. EpCAM+/CD44+ gastric cancer cells formed tumors in immunocompromised mice; however, EpCAM?/CD44?, EpCAM+/CD44? and EpCAM?/CD44+ cells failed to do so. Xenografts of EpCAM+/CD44+ gastric cancer cells maintained a differentiated phenotype and reproduced the morphological and phenotypical heterogeneity of the original gastric tumor tissues. The tumorigenic subpopulation was serially passaged for several generations without significant phenotypic alterations. Moreover, EpCAM+/CD44+, but not EpCAM?/CD44?, EpCAM+/CD44? or EpCAM?/CD44+ cells grew exponentially in vitro as cancer spheres in serum-free medium, maintaining the tumorigenicity. Interestingly, a single cancer stem cell generated a cancer sphere that contained various differentiated cells, supporting multi-potency and self-renewal of a cancer stem cell. EpCAM+/CD44+ cells had greater resistance to anti-cancer drugs than other subpopulation cells. The above in vivo and in vitro results suggest that cancer stem cells, which are enriched in the EpCAM+/CD44+ subpopulation of gastric cancer cells, provide an ideal model system for cancer stem cell research. 相似文献

Membrane protrusions, like lamellipodia, and cell movement are dependent on actin dynamics, which are regulated by a variety of actin-binding proteins acting cooperatively to reorganize actin filaments. Here, we provide evidence that Swiprosin-1, a newly identified actin-binding protein, modulates lamellipodial dynamics by regulating the accessibility of F-actin to cofilin. Overexpression of Swiprosin-1 increased lamellipodia formation in B16F10 melanoma cells, whereas knockdown of Swiprosin-1 inhibited EGF-induced lamellipodia formation, and led to a loss of actin stress fibers at the leading edges of cells but not in the cell cortex. Swiprosin-1 strongly facilitated the formation of entangled or clustered F-actin, which remodeled the structural organization of actin filaments making them inaccessible to cofilin. EGF-induced phosphorylation of Swiprosin-1 at Ser183, a phosphorylation site newly identified using mass spectrometry, effectively inhibited clustering of actin filaments and permitted cofilin access to F-actin, resulting in actin depolymerization. Cells overexpressing a Swiprosin-1 phosphorylation-mimicking mutant or a phosphorylation-deficient mutant exhibited irregular membrane dynamics during the protrusion and retraction cycles of lamellipodia. Taken together, these findings suggest that dynamic exchange of Swiprosin-1 phosphorylation and dephosphorylation is a novel mechanism that regulates actin dynamics by modulating the pattern of cofilin activity at the leading edges of cells. 相似文献

This paper proposed a design and implementation procedure of the Time Synchronization Module (TSM) for the Automatic Identification System (AIS). The proposed TSM module uses a Temperature Compensated Crystal Oscillator (TCXO) as a local reference clock, and consists of a Digitally Controlled Oscillator (DCO), a divider, a phase discriminator, and register blocks. The TSM measures time difference between the 1 PPS from the Global Navigation Satellite System (GNSS) receiver and the generated transmitter clock. The measured time difference is compensated by controlling the DCO and the transmit clock is synchronized to the Universal Time Coordinated (UTC). The designed TSM can also be synchronized to the reference time derived from the received message. The proposed module is tested using the experimental AIS transponder set. The experimental results show that the proposed module satisfies the functional and timing specification of the AIS technical standard, ITU-R M.1371. 相似文献

Papaya, a fruit crop cultivated in tropical and subtropical regions, is known for its nutritional benefits and medicinal applications. Here we report a 3x draft genome sequence of 'SunUp' papaya, the first commercial virus-resistant transgenic fruit tree to be sequenced. The papaya genome is three times the size of the Arabidopsis genome, but contains fewer genes, including significantly fewer disease-resistance gene analogues. Comparison of the five sequenced genomes suggests a minimal angiosperm gene set of 13,311. A lack of recent genome duplication, atypical of other angiosperm genomes sequenced so far, may account for the smaller papaya gene number in most functional groups. Nonetheless, striking amplifications in gene number within particular functional groups suggest roles in the evolution of tree-like habit, deposition and remobilization of starch reserves, attraction of seed dispersal agents, and adaptation to tropical daylengths. Transgenesis at three locations is closely associated with chloroplast insertions into the nuclear genome, and with topoisomerase I recognition sites. Papaya offers numerous advantages as a system for fruit-tree functional genomics, and this draft genome sequence provides the foundation for revealing the basis of Carica's distinguishing morpho-physiological, medicinal and nutritional properties. 相似文献

Viruses are obligate parasites of Eukarya, Archaea and Bacteria. Acanthamoeba polyphaga mimivirus (APMV) is the largest known virus; it grows only in amoeba and is visible under the optical microscope. Mimivirus possesses a 1,185-kilobase double-stranded linear chromosome whose coding capacity is greater than that of numerous bacteria and archaea1, 2, 3. Here we describe an icosahedral small virus, Sputnik, 50 nm in size, found associated with a new strain of APMV. Sputnik cannot multiply in Acanthamoeba castellanii but grows rapidly, after an eclipse phase, in the giant virus factory found in amoebae co-infected with APMV4. Sputnik growth is deleterious to APMV and results in the production of abortive forms and abnormal capsid assembly of the host virus. The Sputnik genome is an 18.343-kilobase circular double-stranded DNA and contains genes that are linked to viruses infecting each of the three domains of life Eukarya, Archaea and Bacteria. Of the 21 predicted protein-coding genes, eight encode proteins with detectable homologues, including three proteins apparently derived from APMV, a homologue of an archaeal virus integrase, a predicted primase-helicase, a packaging ATPase with homologues in bacteriophages and eukaryotic viruses, a distant homologue of bacterial insertion sequence transposase DNA-binding subunit, and a Zn-ribbon protein. The closest homologues of the last four of these proteins were detected in the Global Ocean Survey environmental data set5, suggesting that Sputnik represents a currently unknown family of viruses. Considering its functional analogy with bacteriophages, we classify this virus as a virophage. The virophage could be a vehicle mediating lateral gene transfer between giant viruses. 相似文献

The stability of the Antarctic ice shelves in a warming climate has long been discussed, and the recent collapse of a significant part, over 12,500 km2 in area, of the Larsen ice shelf off the Antarctic Peninsula has led to a refocus toward the implications of ice shelf decay for the stability of Antarctica's grounded ice. Some smaller Antarctic ice shelves have undergone periodic growth and decay over the past 11,000 yr (refs 7-11), but these ice shelves are at the climatic limit of ice shelf viability and are therefore expected to respond rapidly to natural climate variability at century to millennial scales. Here we use records of diatoms, detrital material and geochemical parameters from six marine sediment cores in the vicinity of the Larsen ice shelf to demonstrate that the recent collapse of the Larsen B ice shelf is unprecedented during the Holocene. We infer from our oxygen isotope measurements in planktonic foraminifera that the Larsen B ice shelf has been thinning throughout the Holocene, and we suggest that the recent prolonged period of warming in the Antarctic Peninsula region, in combination with the long-term thinning, has led to collapse of the ice shelf. 相似文献

Several mammals--including sheep, mice, cows, goats, pigs, rabbits, cats, a mule, a horse and a litter of three rats--have been cloned by transfer of a nucleus from a somatic cell into an egg cell (oocyte) that has had its nucleus removed. This technology has not so far been successful in dogs because of the difficulty of maturing canine oocytes in vitro. Here we describe the cloning of two Afghan hounds by nuclear transfer from adult skin cells into oocytes that had matured in vivo. Together with detailed sequence information generated by the canine-genome project, the ability to clone dogs by somatic-cell nuclear transfer should help to determine genetic and environmental contributions to the diverse biological and behavioural traits associated with the many different canine breeds. 相似文献
