Spinocerebellar ataxia type 10 (SCA10; MIM 603516; refs 1,2) is an autosomal dominant disorder characterized by cerebellar ataxia and seizures. The gene SCA10 maps to a 3.8-cM interval on human chromosome 22q13-qter (refs 1,2). Because several other SCA subtypes show trinucleotide repeat expansions, we examined microsatellites in this region. We found an expansion of a pentanucleotide (ATTCT) repeat in intron 9 of SCA10 in all patients in five Mexican SCA10 families. There was an inverse correlation between the expansion size, up to 22.5 kb larger than the normal allele, and the age of onset (r2=0.34, P=0.018). Analysis of 562 chromosomes from unaffected individuals of various ethnic origins (including 242 chromosomes from Mexican persons) showed a range of 10 to 22 ATTCT repeats with no evidence of expansions. Our data indicate that the new SCA10 intronic ATTCT pentanucleotide repeat in SCA10 patients is unstable and represents the largest microsatellite expansion found so far in the human genome. 相似文献

We show here that Mater, a mouse oocyte protein dependent on the maternal genome, is essential for embryonic development beyond the two-cell stage. Genome-wide analysis of single-nucleotide polymorphisms in human expressed sequences 总被引:16,自引:0,他引:16
Single-nucleotide polymorphisms (SNPs) have been explored as a high-resolution marker set for accelerating the mapping of disease genes. Here we report 48,196 candidate SNPs detected by statistical analysis of human expressed sequence tags (ESTs), associated primarily with coding regions of genes. We used Bayesian inference to weigh evidence for true polymorphism versus sequencing error, misalignment or ambiguity, misclustering or chimaeric EST sequences, assessing data such as raw chromatogram height, sharpness, overlap and spacing, sequencing error rates, context-sensitivity and cDNA library origin. Three separate validations-comparison with 54 genes screened for SNPs independently, verification of HLA-A polymorphisms and restriction fragment length polymorphism (RFLP) testing-verified 70%, 89% and 71% of our predicted SNPs, respectively. Our method detects tenfold more true HLA-A SNPs than previous analyses of the EST data. We found SNPs in a large fraction of known disease genes, including some disease-causing mutations (for example, the HbS sickle-cell mutation). Our comprehensive analysis of human coding region polymorphism provides a public resource for mapping of disease genes (available at http://www.bioinformatics.ucla.edu/snp). 相似文献

Social responsibility can be seen as a condition of efficient change management and even as a tool in crisis solving, mainly if associated with innovative social thinking and action. Change/Innovation management is the principal topic of the study, because it is seen as the main way to grow social effectiveness. New management fields are signaled; among them, crisis management and strategic management are accented and concisely presented. A system of effectiveness principles and its internal structure is described and explained. How to innovate is one of questions that our work tries to answer, because a well managed change offers the best formula, both fast and durable, of social problem solving. Can we, finally, design a science of socially responsible change, applied by change/innovation management? Crisis and lack or excess of effectiveness are strongly connected, as showed by our findings. Responsibility-ruled social effectiveness increasing is hence a central theme of the presented study. Economic and global effectiveness without abuse as aspects of social effectiveness are pointed out and a system of criteria for socially responsible social effectiveness evaluating and increasing is sketched. 相似文献

Geometric frustration is a broad phenomenon that results from an intrinsic incompatibility between some fundamental interactions and the underlying lattice geometry. Geometric frustration gives rise to new fundamental phenomena and is known to yield intriguing effects such as the formation of exotic states like spin ice, spin liquids and spin glasses. It has also led to interesting findings of fractional charge quantization and magnetic monopoles. Mechanisms related to geometric frustration have been proposed to understand the origins of relaxor and multiferroic behaviour, colossal magnetocapacitive coupling, and unusual and novel mechanisms of high-transition-temperature superconductivity. Although geometric frustration has been particularly well studied in magnetic systems in the past 20 years or so, its manifestation in the important class formed by ferroelectric materials (which are compounds with electric rather than magnetic dipoles) is basically unknown. Here we show, using a technique based on first principles, that compositionally graded ferroelectrics possess the characteristic 'fingerprints' associated with geometric frustration. These systems have a highly degenerate energy surface and display critical phenomena. They further reveal exotic orderings with novel stripe phases involving complex spatial organization. These stripes display spiral states, topological defects and curvature. Compositionally graded ferroelectrics can thus be considered the 'missing link' that brings ferroelectrics into the broad category of materials able to exhibit geometric frustration. Our ab initio calculations allow deep microscopic insight into this novel geometrically frustrated system. 相似文献

The acquired immunodeficiency syndrome (AIDS)-causing lentiviruses human immunodeficiency virus (HIV) and simian immunodeficiency virus (SIV) effectively evade host immunity and, once established, infections with these viruses are only rarely controlled by immunological mechanisms. However, the initial establishment of infection in the first few days after mucosal exposure, before viral dissemination and massive replication, may be more vulnerable to immune control. Here we report that SIV vaccines that include rhesus cytomegalovirus (RhCMV) vectors establish indefinitely persistent, high-frequency, SIV-specific effector memory T-cell (T(EM)) responses at potential sites of SIV replication in rhesus macaques and stringently control highly pathogenic SIV(MAC239) infection early after mucosal challenge. Thirteen of twenty-four rhesus macaques receiving either RhCMV vectors alone or RhCMV vectors followed by adenovirus 5 (Ad5) vectors (versus 0 of 9 DNA/Ad5-vaccinated rhesus macaques) manifested early complete control of SIV (undetectable plasma virus), and in twelve of these thirteen animals we observed long-term (≥1 year) protection. This was characterized by: occasional blips of plasma viraemia that ultimately waned; predominantly undetectable cell-associated viral load in blood and lymph node mononuclear cells; no depletion of effector-site CD4(+) memory T cells; no induction or boosting of SIV Env-specific antibodies; and induction and then loss of T-cell responses to an SIV protein (Vif) not included in the RhCMV vectors. Protection correlated with the magnitude of the peak SIV-specific CD8(+) T-cell responses in the vaccine phase, and occurred without anamnestic T-cell responses. Remarkably, long-term RhCMV vector-associated SIV control was insensitive to either CD8(+) or CD4(+) lymphocyte depletion and, at necropsy, cell-associated SIV was only occasionally measurable at the limit of detection with ultrasensitive assays, observations that indicate the possibility of eventual viral clearance. Thus, persistent vectors such as CMV and their associated T(EM) responses might significantly contribute to an efficacious HIV/AIDS vaccine. 相似文献
