Analysis of a human DNA excision repair gene involved in group A xeroderma pigmentosum and containing a zinc-finger domain

Xeroderma pigmentosum (XP) is an autosomal recessive disease, characterized by a high incidence of sunlight-induced skin cancer. Cells from people with this condition are hypersensitive to ultraviolet because of a defect in DNA repair. There are nine genetic complementation groups of XP, groups A-H and a variant. We have cloned the mouse DNA repair gene that complements the defect of group A, the XPAC gene. Here we report molecular cloning of human and mouse XPAC complementary DNAs. Expression of XPAC cDNA confers ultraviolet-resistance on several group A cell lines, but not on lines of other XP groups. Almost all group A lines tested showed abnormality or absence of XPAC messenger RNAs. These results indicate that a defective XPAC gene causes group A XP. The human and mouse XPAC genes are located on chromosome 9q34.1 and chromosome 4C2, respectively. Human XPAC cDNA encodes a protein of 273 amino acids with a zinc-finger motif.     

Construction of a thin film PO 2-electrode using gold paste

Summary A thin film gold electrode for biological oxygen measurement was constructed using gold paste. This electrode is easy to construct, has high sensitivity and is disturbed only slightly by bubbling of the surrounding fluid. As an application, the electrode was used to measure oxygen consumption of the skin and yielded a reasonable value.With leave from the department of gynecology, School of Medicine, Hokkaido University.Glass laboratory.     

The effect of glutamate,cysteate and related amino acids on insect muscle

Summary Application of Glu, CySO₂ or CySO₃ to blowfly larvae caused paralysis and increased the membrane potential of larval muscle. The contents of Glu and CySO₃ in larval muscle containing motor nerve terminals were markedly decreased after the perfusion of high K⁺ saline solution.     

Juvenile hormone I is the principal juvenile hormone in a hemipteran insect,Riptortus clavatus

Juvenile hormone I (JH I) was identified by combined gas chromatography/mass spectrometry as the predominant JH in the hemolymph of female adults of the bean bug,Riptortus clavatus (Thunberg) (Hemiptera: Alydidae). Among JH I, II, and III, JH I was the most effective hormone for inducing the synthesis of yolk proteins in diapause adults.     

Intensification of the 5.9-nm actin layer line in contracting muscle

According to the cross-bridge model of muscle contraction, an interaction of myosin heads with interdigitating actin filaments produces tension. Although X-ray equatorial diffraction patterns of active (contracting) muscle show that the heads are in the vicinity of the actin filaments, structural proof of actual attachment of heads to actin during contraction has been elusive. We show here that during contraction of frog skeletal muscle, the 5.9-nm layer line arising from the genetic helix of actin is intensified by as much as 56% of the change which occurs when muscle enters rigor, using a two-dimensional X-ray detector. This provides strong structural evidence that myosin heads do in fact attach during contraction.     

Genome sequence of the ultrasmall unicellular red alga Cyanidioschyzon merolae 10D

Small, compact genomes of ultrasmall unicellular algae provide information on the basic and essential genes that support the lives of photosynthetic eukaryotes, including higher plants. Here we report the 16,520,305-base-pair sequence of the 20 chromosomes of the unicellular red alga Cyanidioschyzon merolae 10D as the first complete algal genome. We identified 5,331 genes in total, of which at least 86.3% were expressed. Unique characteristics of this genomic structure include: a lack of introns in all but 26 genes; only three copies of ribosomal DNA units that maintain the nucleolus; and two dynamin genes that are involved only in the division of mitochondria and plastids. The conserved mosaic origin of Calvin cycle enzymes in this red alga and in green plants supports the hypothesis of the existence of single primary plastid endosymbiosis. The lack of a myosin gene, in addition to the unexpressed actin gene, suggests a simpler system of cytokinesis. These results indicate that the C. merolae genome provides a model system with a simple gene composition for studying the origin, evolution and fundamental mechanisms of eukaryotic cells.
