A question of long-standing interest to philosophers, psychologists and neuroscientists is how the brain selects which signals enter consciousness. Binocular rivalry and attention both involve selection of visual stimuli, but affect perception quite differently. During binocular rivalry, awareness alternates between two different stimuli presented to the two eyes. In contrast, attending to one of two different stimuli impairs discrimination of the ignored stimulus, but without causing it to disappear from consciousness. Here we show that despite this difference, attention and rivalry rely on shared object-based selection mechanisms. We cued attention to one of two superimposed transparent surfaces and then deleted the image of one surface from each eye, resulting in rivalry. Observers usually reported seeing only the cued surface. They were also less accurate in judging unpredictable changes in the features of the uncued surface. Our design ensured that selection of the cued surface could not have resulted from spatial, ocular or feature-based mechanisms. Rather, attention was drawn to one surface, and this caused the other surface to be perceptually suppressed during rivalry. These results raise the question of how object representations compete during these two forms of perceptual selection, even as the features of those objects change unpredictably over time.  相似文献

The concept of an electrical current encircling the Earth at high altitudes was first proposed in 1917 to explain the depression of the horizontal component of the Earth's magnetic field during geomagnetic storms. In situ measurements of the extent and composition of this current were made some 50 years later and an image was obtained in 2001 (ref. 6). Ring currents of a different nature were observed at Jupiter and their presence inferred at Saturn. Here we report images of the ring current at Saturn, together with a day-night pressure asymmetry and tilt of the planet's plasma sheet, based on measurements using the magnetospheric imaging instrument (MIMI) on board Cassini. The ring current can be highly variable with strong longitudinal asymmetries that corotate nearly rigidly with the planet. This contrasts with the Earth's ring current, where there is no rotational modulation and initial asymmetries are organized by local time effects.  相似文献

Because only a small fraction of asbestos-exposed individuals develop malignant mesothelioma, and because mesothelioma clustering is observed in some families, we searched for genetic predisposing factors. We discovered germline mutations in the gene encoding BRCA1 associated protein-1 (BAP1) in two families with a high incidence of mesothelioma, and we observed somatic alterations affecting BAP1 in familial mesotheliomas, indicating biallelic inactivation. In addition to mesothelioma, some BAP1 mutation carriers developed uveal melanoma. We also found germline BAP1 mutations in 2 of 26 sporadic mesotheliomas; both individuals with mutant BAP1 were previously diagnosed with uveal melanoma. We also observed somatic truncating BAP1 mutations and aberrant BAP1 expression in sporadic mesotheliomas without germline mutations. These results identify a BAP1-related cancer syndrome that is characterized by mesothelioma and uveal melanoma. We hypothesize that other cancers may also be involved and that mesothelioma predominates upon asbestos exposure. These findings will help to identify individuals at high risk of mesothelioma who could be targeted for early intervention.  相似文献

We sequenced eight melanoma exomes to identify new somatic mutations in metastatic melanoma. Focusing on the mitogen-activated protein (MAP) kinase kinase kinase (MAP3K) family, we found that 24% of melanoma cell lines have mutations in the protein-coding regions of either MAP3K5 or MAP3K9. Structural modeling predicted that mutations in the kinase domain may affect the activity and regulation of these protein kinases. The position of the mutations and the loss of heterozygosity of MAP3K5 and MAP3K9 in 85% and 67% of melanoma samples, respectively, together suggest that the mutations are likely to be inactivating. In in vitro kinase assays, MAP3K5 I780F and MAP3K9 W333* variants had reduced kinase activity. Overexpression of MAP3K5 or MAP3K9 mutants in HEK293T cells reduced the phosphorylation of downstream MAP kinases. Attenuation of MAP3K9 function in melanoma cells using siRNA led to increased cell viability after temozolomide treatment, suggesting that decreased MAP3K pathway activity can lead to chemoresistance in melanoma.  相似文献

Anthocyanins are red and violet pigments that color flowers, fruits and epidermal tissues in virtually all flowering plants. A single order, Caryophyllales, contains families in which an unrelated family of pigments, the betalains, color tissues normally pigmented by anthocyanins. Here we show that CYP76AD1 encoding a novel cytochrome P450 is required to produce the red betacyanin pigments in beets. Gene silencing of CYP76AD1 results in loss of red pigment and production of only yellow betaxanthin pigment. Yellow betalain mutants are complemented by transgenic expression of CYP76AD1, and an insertion in CYP76AD1 maps to the R locus that is responsible for yellow versus red pigmentation. Finally, expression of CYP76AD1 in yeast verifies its position in the betalain biosynthetic pathway. Thus, this cytochrome P450 performs the biosynthetic step that provides the cyclo-DOPA moiety of all red betacyanins. This discovery will contribute to our ability to engineer this simple, nutritionally valuable pathway into heterologous species.  相似文献

This paper explores the legacy of the great French experimental physicist Victor Regnault through the example of Gabriel Lippmann, whose engagement with electrical standardization during the early 1880s was guided by Regnault's methodological precept to measure ‘directly’. Lippmann's education reveals that the theoretical practice of ‘direct’ measurement entailed eliminating extraneous physical effects through the experimental design, rather than, like physicists in Britain and Germany, making numerical ‘corrections’ to measured values. It also provides, paradoxically, exemplars of the qualitative theoretical practices that sustained Regnault's misguided ambition to avoid theory. By considering the largely negative reactions to Lippmann's proposals for selecting suitable electrical units and methods of measuring the ohm, this paper associates these theoretical practices with the ineffectual rhetorical strategies employed by Lippmann to promote his work, and thereby indicates that the practice of direct measurement was limited to French experimental physics. Whilst this result aligns readily with the existence of divergent nineteenth century British and German cultures of precision, it emerges from a very different disciplinary context in which the practice of precision electrical measurement developed independently of submarine telegraphy. This is because, as this paper illustrates, telegraphic engineering and experimental physics remained separate professions in France.  相似文献

The comparison of related genomes has emerged as a powerful lens for genome interpretation. Here we report the sequencing and comparative analysis of 29 eutherian genomes. We confirm that at least 5.5% of the human genome has undergone purifying selection, and locate constrained elements covering ~4.2% of the genome. We use evolutionary signatures and comparisons with experimental data sets to suggest candidate functions for ~60% of constrained bases. These elements reveal a small number of new coding exons, candidate stop codon readthrough events and over 10,000 regions of overlapping synonymous constraint within protein-coding exons. We find 220 candidate RNA structural families, and nearly a million elements overlapping potential promoter, enhancer and insulator regions. We report specific amino acid residues that have undergone positive selection, 280,000 non-coding elements exapted from mobile elements and more than 1,000 primate- and human-accelerated elements. Overlap with disease-associated variants indicates that our findings will be relevant for studies of human biology, health and disease.  相似文献
