Local hydrogen-clearance and 257-01257-01257-01- measurements in micro-areas of the rat brain

Résumé La pression partielle de l'oxygène et la «clearance» de l'hydrogène ont été mesurées dans des régions circonscrites. Les deux valeurs sont enregistrées par la même microélectrode de platine (diamètre de la pointe 10). Cette méthode nouvelle est appliquée dans des expériences faites sur le cerveau de Rats narcotisés et on est capable d'évaluer les inhomogénités locales de l'apport d'oxygène dépendant du flux du sang.     

The branching programme of mouse lung development

Mammalian lungs are branched networks containing thousands to millions of airways arrayed in intricate patterns that are crucial for respiration. How such trees are generated during development, and how the developmental patterning information is encoded, have long fascinated biologists and mathematicians. However, models have been limited by a lack of information on the normal sequence and pattern of branching events. Here we present the complete three-dimensional branching pattern and lineage of the mouse bronchial tree, reconstructed from an analysis of hundreds of developmental intermediates. The branching process is remarkably stereotyped and elegant: the tree is generated by three geometrically simple local modes of branching used in three different orders throughout the lung. We propose that each mode of branching is controlled by a genetically encoded subroutine, a series of local patterning and morphogenesis operations, which are themselves controlled by a more global master routine. We show that this hierarchical and modular programme is genetically tractable, and it is ideally suited to encoding and evolving the complex networks of the lung and other branched organs.     

磁性隧道结中自旋相关输运的势垒影响

为了研究势垒对铁磁/绝缘层/半导体/绝缘层/铁磁(FM/I/SM/I/FM)双隧道结中自旋相关电子输运特性的影响,提出了在半导体层厚度合适的情况下,非对称势垒对于提高平行结构磁性双隧道结的自旋注入效率SIE(spin injection efficiency)更具优势.数值计算结果表明,当两势垒强度的比率达到合适数值时双结的SIE和隧穿磁电阻TMR(tunneling magnetore resistance)都将达到最大,这给提高从铁磁到半导体的SIE带来新选择.研究还表明,非对称势垒结构磁性隧道结中增大铁磁交换能对提高SIE和TMR都是有益的,而且铁磁交换能的增加对SIE的提高要比对TMR的提高更显著.     

MAP and kinesin-dependent nuclear positioning is required for skeletal muscle function

The basic unit of skeletal muscle in all metazoans is the multinucleate myofibre, within which individual nuclei are regularly positioned. The molecular machinery responsible for myonuclear positioning is not known. Improperly positioned nuclei are a hallmark of numerous diseases of muscle, including centronuclear myopathies, but it is unclear whether correct nuclear positioning is necessary for muscle function. Here we identify the microtubule-associated protein ensconsin (Ens)/microtubule-associated protein 7 (MAP7) and kinesin heavy chain (Khc)/Kif5b as essential, evolutionarily conserved regulators of myonuclear positioning in Drosophila and cultured mammalian myotubes. We find that these proteins interact physically and that expression of the Kif5b motor domain fused to the MAP7 microtubule-binding domain rescues nuclear positioning defects in MAP7-depleted cells. This suggests that MAP7 links Kif5b to the microtubule cytoskeleton to promote nuclear positioning. Finally, we show that myonuclear positioning is physiologically important. Drosophila ens mutant larvae have decreased locomotion and incorrect myonuclear positioning, and these phenotypes are rescued by muscle-specific expression of Ens. We conclude that improper nuclear positioning contributes to muscle dysfunction in a cell-autonomous fashion.     

关于加强青年农民工的思想道德建设的思考

青年农民工是我国工业化、城市化进程中的一支重要力量,他们在为我国经济社会发展做出巨大贡献的同时,却遭受着不公正的待遇,不能取得与拥有城镇户口身份的劳动者平等的地位并享受相应权益等等,这些问题在一定程度上造成青年农民工在思想和道德素质上表现出种种不足,给社会和谐稳定带来了隐患。为此,加强青年农民工的思想道德建设,成为和谐社会发展中的一个重要课题。     

Postischemic ATP levels predict hepatic function 24 hours following ischemia in the rat

Hepatic function was assessed by the aminopyrine breath test (ABT) in male Sprague Dawley rats 24 h after partial hepatic ischemia. ABT decreased progressively to 26.3 (p less than 0.05) and 19.7% of dose (p less than 0.05) after 90 and 120 min of ischemia, respectively. ABT at 24 h after injury was correlated to the concentration of ATP in the ischemic lobes 1 h after the onset of reperfusion (r2 = 0.971) but not to ALT activity in plasma at 1 h (r2 = 0.391). We conclude that postischemic ATP levels are a better index of subsequent hepatic function than ALT.     

Adenovirus-mediated in vivo gene transfer and expression in normal rat liver.

Replication deficient, recombinant adenovirus (Ad) vectors do not require target cell replication for transfer and expression of exogenous genes and thus may be useful for in vivo gene therapy in hepatocytes. In vitro, primary cultures of rat hepatocytes infected with a recombinant Ad containing a human alpha 1-antitrypsin cDNA (Ad-alpha 1AT) synthesized and secreted human alpha 1AT for 4 weeks. In rats, in vivo intraportal administration of a recombinant Ad containing the E. coli lacZ gene, was followed by expression of beta-galactosidase in hepatocytes 3 days after infection. Intraportal infusion of Ad-alpha 1AT produced detectable serum levels of human alpha 1AT for 4 weeks. Thus, targeted gene expression has been achieved in the liver, albeit at low levels, suggesting that adenovirus vectors may be a useful means for in vivo gene therapy in liver disorders.
