Cuticular hydrocarbons (CHs) contain complex mixtures of components that cover the surface of all insects to restrict water loss, and in many species function in chemical communication. The complex mixtures have been used as a chemotaxonomic tool since the 1970s. The majority of CH components can be identified by gas chromatography-mass spectrometry (GC-MS), allowing hydrocarbon profiles to be used as chemotaxonomic characters, which can differentiate insect species, including cryptic species. The present study used GC-MS for the identification of CHs of males and females of three species of calliphorids: Cochliomyia macellaria (Fabricius), Hemilucilia segmentaria (Fabricius) and Lucilia cuprina (Wiedmann), A comparison of the chromatographic profiles from the three species allows them to be readily separated. Hydrocarbons identified from the three species contain 23 to 37 carbons and include n-alkanes, methyl-branched alkanes and alkenes. The most abundant compound for both sexes of C. macellaria was n-heptacosane, while for H. segmentaria it was 2-methyltriacontane; for L. cuprina, however, the most abundant compound was hentriacontene for females and nonacosene for males. The results obtained in this study are important for the establishment of a database to be used in chemotaxonomy of sanitarily and forensically important insects.  相似文献

Sphingosine 1-phosphate (S1P) is a bioactive lipid that acts through a family of G-protein-coupled receptors. Herein, we report evidence of a novel redox-based cross-talk between S1P and insulin signaling pathways. In skeletal muscle cells S1P, through engagement of its S1P2 receptor, is found to produce a transient burst of reactive oxygen species through a calcium-dependent activation of the small GTPase Rac1. S1P-induced redox-signaling is sensed by protein tyrosine phosphatase-1B, the main negative regulator of insulin receptor phosphorylation, which undergoes oxidation and enzymatic inhibition. This redox-based inhibition of the phosphatase provokes a ligand-independent trans-phosphorylation of insulin receptor and a strong increase in glucose uptake. Our results propose a new role of S1P, recognizing the lipid as an insulin-mimetic cue and pointing at reactive oxygen species as critical regulators of the cross-talk between S1P and insulin pathways. Any possible implication of S1P-directed insulin signaling in diabetes and insulin resistance remains to be established.  相似文献

Resetting epigenetic signatures to induce somatic cell reprogramming   总被引:1,自引:0,他引:1  
Somatic cell reprogramming consists of the induction of a complex sequence of events that results in the modification of the developmental state of the cell. It is now routinely possible to reprogram fully differentiated cells back to pluripotent cells, and to transdifferentiate cells of a given type in cells of a totally different lineage origin. However, whether there are key initiating factors that are distinct from those that control stem-cell renewal and that can initiate the reprogramming process remains unknown. In contrast, what is clear is that, by modifying the epigenetic status of a cell, its reprogramming can be initiated. Here, we review the current literature that shows how the plasticity of a cell can be modulated by modifying its epigenetic status, and we discuss how epigenetic barriers can be removed, to induce an efficient reprogramming process.  相似文献

Aldehyde oxidases (AOXs) and xanthine dehydrogenases (XDHs) belong to the family of molybdo-flavoenzymes. Although AOXs are not identifiable in fungi, these enzymes are represented in certain protists and the majority of plants and vertebrates. The physiological functions and substrates of AOXs are unknown. Nevertheless, AOXs are major drug metabolizing enzymes, oxidizing a wide range of aromatic aldehydes and heterocyclic compounds of medical/toxicological importance. Using genome sequencing data, we predict the structures of AOX genes and pseudogenes, reconstructing their evolution. Fishes are the most primitive organisms with an AOX gene (AOXα), originating from the duplication of an ancestral XDH. Further evolution of fishes resulted in the duplication of AOXα into AOXβ and successive pseudogenization of AOXα. AOXβ is maintained in amphibians and it is the likely precursors of reptilian, avian, and mammalian AOX1. Amphibian AOXγ is a duplication of AOXβ and the likely ancestor of reptilian and avian AOX2, which, in turn, gave rise to mammalian AOX3L1. Subsequent gene duplications generated the two mammalian genes, AOX3 and AOX4. The evolution of mammalian AOX genes is dominated by pseudogenization and deletion events. Our analysis is relevant from a structural point of view, as it provides information on the residues characterizing the three domains of each mammalian AOX isoenzyme. We cloned the cDNAs encoding the AOX proteins of guinea pig and cynomolgus monkeys, two unique species as to the evolution of this enzyme family. We identify chimeric RNAs from the human AOX3 and AOX3L1 pseudogenes with potential to encode a novel microRNA.  相似文献

This paper deals with the function of farm land. The objective is to demonstrate that the socio-environmental function of land ownership has a different and broader role than mere administration. According to the current Brazilian Constitution, owning land gives a right to property that is bound by the social and environmental requirements prescribed by the law. Therefore, the nature of rights over land property has changed. The traditional capitalist ownership has been socialised, having as its organising and limiting principle the well-being of present and future generations. Although participatory supply chain analysis (PSCA) is widely used to trigger action in development aid, there is very little published describing the method, its benefits and limitations. The theoretical background of participatory processes related to supply chain analysis and the related benefits and limitations found while implementing the method in Laos are presented. It is concluded that joint problem-solving in supply chains can be facilitated through PSCA as the method allows participants to evaluate their initial assumptions, investigate institutional barriers, gather new information and realise some of the interdependencies among supply chain participants. On the other hand, the lack of legitimate representatives, the failure to convince important stakeholders to participate, distances between participants, the length and breadth of the supply chain were factors limiting joint action. This method is recommended to study supply chains of niche products and local/regional products locally processed.  相似文献
