DNA is often modelled as an isotropic rod, but its chiral structure suggests the possible importance of anisotropic mechanical properties, including coupling between twisting and stretching degrees of freedom. Simple physical intuition predicts that DNA should unwind under tension, as it is pulled towards a denatured structure. We used rotor bead tracking to directly measure twist-stretch coupling in single DNA molecules. Here we show that for small distortions, contrary to intuition, DNA overwinds under tension, reaching a maximum twist at a tension of approximately 30 pN. As tension is increased above this critical value, the DNA begins to unwind. The observed twist-stretch coupling predicts that DNA should also lengthen when overwound under constant tension, an effect that we quantitatively confirm. We present a simple model that explains these unusual mechanical properties, and also suggests a possible origin for the anomalously large torsional rigidity of DNA. Our results have implications for the action of DNA-binding proteins that must stretch and twist DNA to compensate for variability in the lengths of their binding sites. The requisite coupled DNA distortions are favoured by the intrinsic mechanical properties of the double helix reported here.  相似文献

We examined the evidence supporting the hypothesis that jaguars (Panthera onca) have morphological and behavioural adaptations to facilitate reptile predation. Jaguars’ head and bite features show adaptations to durophagy (consumption of hard-integumented prey) that are very unusual within the genus Panthera. These include: thick canines, well-developed head muscles and a fatal bite directed to braincase or nape. These characteristics have been previously considered an adaptation for the consumption of reptilian prey, whose thick integument poses a challenge to predation. Although causation of any trait as result of natural selection is hard to demonstrate with ecological evidence, its consequences can be suggested and predictions made. Here, through a review of the literature on jaguar predatory habits, we tallied the evidence for saurophagy against environmental characteristics correlated with jaguar predation on reptiles. We offer a new explanation for the presence of those traits, based on the selection patterns, prey abundances and main predation habits over the geographic range of the jaguar. We believe that these features allow jaguars to overcome dangerous and/or armoured prey by dispatching them rapidly through a bite to the head or nape. Reptile consumption is restricted to areas of high reptile abundance while dangerous or armoured prey is consistently preyed upon throughout the jaguar’s distribution, and selected even when their abundance is low. This questions the validity of the saurophagy hypothesis. We believe that these results are relevant to jaguar conservation and management, because some of the main prey species are severely threatened over their range, and because threatened reptiles are also consumed as prey.  相似文献

Basidiomes of polypore fungi host many insects. Yet systematic information about insect assemblages from most fungal species is lacking. We studied the insect community associated with the wood-decaying fungus Inonotus radiatus (Sowerby: Fr.) P. Karst. (Hymenochaetales). More specifically, we studied the effect of successional stage and weight of basidiomes, as well as shore exposition (north or south), on species richness and composition, as well as occurrence and abundance of the most abundant fungivores. Basidiomes were collected from riparian forests at five lakes in Sweden. Insects were reared out from the basidiomes in the laboratory. A total of 5645 adult insect individuals of 117 taxa were obtained. Among these, 2782 specimens of 36 taxa use Inonotus radiatus basidiomes as breeding habitat. Eight species of parasitic wasps were new to Sweden. The most abundant fungivore was Ennearthron cornutum (Ciidae), which is a generalist breeding in many polypore species. Based on our material and literature, the melandryid beetles Abdera affinis and A. flexuosa seem to be specialists on the order Hymenochaetales. Other frequent fungivores were Dorcatoma dresdensis (Ptinidae), and the lepidopterans Archinemapogon yildizae, Nemapogon cloacellus and N. picarellus (Tineidae). The parasitoid community associated with the tineid moths was similar to the community associated with the other polypore species in the order Polyporales. In contrast, the parasitoids associated with Dorcatoma (and/or Abdera) were different from the other Polyporales species, suggesting that the fungal host species is more important for these parasitoid species than the beetle host species itself. The most abundant and frequent parasitoids were the braconids Diospilus dispar and Colastes fritzeni, which both parasitise Dorcatoma. Species richness was significantly smaller in fresh than in more decayed basidiomes, but species composition did not differ. There was no difference in species composition or richness between north and south shorelines.  相似文献

Enterotypes of the human gut microbiome   总被引:6,自引:0,他引:6  
Our knowledge of species and functional composition of the human gut microbiome is rapidly increasing, but it is still based on very few cohorts and little is known about variation across the world. By combining 22 newly sequenced faecal metagenomes of individuals from four countries with previously published data sets, here we identify three robust clusters (referred to as enterotypes hereafter) that are not nation or continent specific. We also confirmed the enterotypes in two published, larger cohorts, indicating that intestinal microbiota variation is generally stratified, not continuous. This indicates further the existence of a limited number of well-balanced host-microbial symbiotic states that might respond differently to diet and drug intake. The enterotypes are mostly driven by species composition, but abundant molecular functions are not necessarily provided by abundant species, highlighting the importance of a functional analysis to understand microbial communities. Although individual host properties such as body mass index, age, or gender cannot explain the observed enterotypes, data-driven marker genes or functional modules can be identified for each of these host properties. For example, twelve genes significantly correlate with age and three functional modules with the body mass index, hinting at a diagnostic potential of microbial markers.  相似文献

Potent virus-specific cytotoxic T lymphocyte (CTL) responses elicited by candidate AIDS vaccines have recently been shown to control viral replication and prevent clinical disease progression after pathogenic viral challenges in rhesus monkeys. Here we show that viral escape from CTL recognition can result in the eventual failure of this partial immune protection. Viral mutations that escape from CTL recognition have been previously described in humans infected with human immunodeficiency virus (HIV) and monkeys infected with simian immunodeficiency virus (SIV). In a cohort of rhesus monkeys that were vaccinated and subsequently infected with a pathogenic hybrid simian-human immunodeficiency virus (SHIV), the frequency of viral sequence mutations within CTL epitopes correlated with the level of viral replication. A single nucleotide mutation within an immunodominant Gag CTL epitope in an animal with undetectable plasma viral RNA resulted in viral escape from CTLs, a burst of viral replication, clinical disease progression, and death from AIDS-related complications. These data indicate that viral escape from CTL recognition may be a major limitation of the CTL-based AIDS vaccines that are likely to be administered to large human populations over the next several years.  相似文献

Huntingtin interacting protein 1 (HIP1) is an accessory protein of the clathrin-mediated endocytosis (CME) pathway, yet its precise role and the step at which it becomes involved are unclear. We employed live-cell imaging techniques to focus on the early steps of CME and characterize HIP1 dynamics. We show that HIP1 is highly colocalized with clathrin at the plasma membrane and shares similar dynamics with a subpopulation of clathrin assemblies. Employing transferrin receptor fused to pHluorin, we distinguished between open pits to which HIP1 localizes and newly internalized vesicles that are devoid of HIP1. Moreover, shRNA knockdown of clathrin compromised HIP1 membranal localization, unlike the reported behavior of Sla2p. HIP1 fragment, lacking its ANTH and Talin-like domains, inhibits internalization of transferrin, but retains colocalization with membranal clathrin assemblies. These data demonstrate HIP1’s role in pits maturation and formation of the coated vesicle, and its strong dependence on clathrin for membranal localization. Working memory represents the ability of the brain to hold externally or internally driven information for relatively short periods of time. Persistent neuronal activity is the elementary process underlying working memory but its cellular basis remains unknown. The most widely accepted hypothesis is that persistent activity is based on synaptic reverberations in recurrent circuits. The entorhinal cortex in the parahippocampal region is crucially involved in the acquisition, consolidation and retrieval of long-term memory traces for which working memory operations are essential. Here we show that individual neurons from layer V of the entorhinal cortex-which link the hippocampus to extensive cortical regions-respond to consecutive stimuli with graded changes in firing frequency that remain stable after each stimulus presentation. In addition, the sustained levels of firing frequency can be either increased or decreased in an input-specific manner. This firing behaviour displays robustness to distractors; it is linked to cholinergic muscarinic receptor activation, and relies on activity-dependent changes of a Ca2+-sensitive cationic current. Such an intrinsic neuronal ability to generate graded persistent activity constitutes an elementary mechanism for working memory.  相似文献

In photosynthetic cells, chloroplasts migrate towards illuminated sites to optimize photosynthesis and move away from excessively illuminated areas to protect the photosynthetic machinery. Although this movement of chloroplasts in response to light has been known for over a century, the photoreceptor mediating this process has not been identified. The Arabidopsis gene NPL1 (ref. 2) is a paralogue of the NPH1 gene, which encodes phototropin, a photoreceptor for phototropic bending. Here we show that NPL1 is required for chloroplast relocation induced by blue light. A loss-of-function npl1 mutant showed no chloroplast avoidance response in strong blue light, whereas the accumulation of chloroplasts in weak light was normal. These results indicate that NPL1 may function as a photoreceptor mediating chloroplast relocation.  相似文献
