螺旋搅拌桨是新型工业结晶技术与设备的关键部件之一。基于CAD/CAM技术,研究了螺旋搅拌桨的设计与数控加工技术,利用VC++和UG/OPENGRIP技术开发螺旋桨三维造型模块,快速生成螺旋桨CAD数据模型。根据螺旋桨叶面曲率变化划分出不同的数控加工区域,给出了五轴数控加工的刀轴倾角的确定方法。在DMC2600五轴数控加工中心对中1300螺旋搅拌桨实现了数控加工,达到了产品技术要求。  相似文献

Cyclin-dependent kinases (CDKs) drive major cell cycle events including the initiation of chromosomal DNA replication. We identified two S phase CDK (S-CDK) phosphorylation sites in the budding yeast Sld3 protein that, together, are essential for DNA replication. Here we show that, when phosphorylated, these sites bind to the amino-terminal BRCT repeats of Dpb11. An Sld3-Dpb11 fusion construct bypasses the requirement for both Sld3 phosphorylation and the N-terminal BRCT repeats of Dpb11. Co-expression of this fusion with a phospho-mimicking mutant in a second essential CDK substrate, Sld2, promotes DNA replication in the absence of S-CDK. Therefore, Sld2 and Sld3 are the minimal set of S-CDK targets required for DNA replication. DNA replication in cells lacking G1 phase CDK (G1-CDK) required expression of the Cdc7 kinase regulatory subunit, Dbf4, as well as Sld2 and Sld3 bypass. Our results help to explain how G1- and S-CDKs promote DNA replication in yeast.  相似文献

The dense granule protein 4 (GRA4) is a granular protein from Toxoplasma gondii, and is a candidate for vaccination against this parasite. In this study, the plasmid pcDNA3.1-GRA4 (pGRA4), encoding for the GRA4 antigen, was incorporated by the dehydration-rehydration method into liposomes composed of 16 mmol/L egg phosphatidylcholine (PC), 8 mmol/L dioleoyl phosphatidylethanolamine (DOPE), and 4 mmol/L 1,2-diodeoyl-3-(trimethylammonium) propane (DOTAP). C57BL/6 mice and BALB/c mice were immunized intramuscularly three times with liposome-encapsulated pGRA4 to determine whether DNA immunization could elicit a protective immune response to T. gondii. Enzyme-linked immunosorbent assay (ELISA) of sera from immunized mice showed that liposome-encapsulated pGRA4 generated high levels of IgG antibodies to GRA4. Production of primary interferon (IFN)-γ and interleukin (IL)-2 in GRA4-stimulated splenocytes from vaccinated mice suggested a modulated Th1-type response. 72.7% of C57BL/6 mice immunized with liposome-encapsulated pGRA4 survived the challenge with 80 tissue cysts of ME49 strain, whereas C57BL/6 mice immunized with pGRA4 had only a survival rate of 54.5%. When immunized BALB/c mice were intraperitoneally challenged with 103 tachyzoites of the highly virulent RH strain, the survival time of mice immunized with liposome-encapsulated pGRA4 was markedly longer than that of other groups. Our observations show that liposome-encapsulated pGRA4 enhanced the protective effect against infection of T. gondii.  相似文献

以顶部带有2层钢框架的北京机场控制塔台的振动台试验模型为计算对象,采用基于振型叠加的结构地震响应时程分析法,计算了该高耸塔台模型在地震作用下各个振型的时程反应.计算中采用了Rayleigh阻尼,由于结构是钢一钢筋混凝土结构,振型阻尼比不易确定,在本次计算中振型阻尼比取振动台试验测出的第1阶和第2阶振型阻尼比.结果表明高振型对塔台结构的最大位移的影响不大,但高阶振型对塔台结构地震剪力的影响却不容忽视.将计算结果与试验值进行了对比分析,验证了所得结论的正确性.  相似文献

Long-duration gamma-ray bursts (GRBs) release copious amounts of energy across the entire electromagnetic spectrum, and so provide a window into the process of black hole formation from the collapse of massive stars. Previous early optical observations of even the most exceptional GRBs (990123 and 030329) lacked both the temporal resolution to probe the optical flash in detail and the accuracy needed to trace the transition from the prompt emission within the outflow to external shocks caused by interaction with the progenitor environment. Here we report observations of the extraordinarily bright prompt optical and gamma-ray emission of GRB 080319B that provide diagnostics within seconds of its formation, followed by broadband observations of the afterglow decay that continued for weeks. We show that the prompt emission stems from a single physical region, implying an extremely relativistic outflow that propagates within the narrow inner core of a two-component jet.  相似文献

The NAD-dependent protein deacetylase Sir2 (silent information regulator 2) regulates lifespan in several organisms. SIRT1, the mammalian orthologue of yeast Sir2, participates in various cellular functions and possibly tumorigenesis. Whereas the cellular functions of SIRT1 have been extensively investigated, less is known about the regulation of SIRT1 activity. Here we show that Deleted in Breast Cancer-1 (DBC1), initially cloned from a region (8p21) homozygously deleted in breast cancers, forms a stable complex with SIRT1. DBC1 directly interacts with SIRT1 and inhibits SIRT1 activity in vitro and in vivo. Downregulation of DBC1 expression potentiates SIRT1-dependent inhibition of apoptosis induced by genotoxic stress. Our results shed new light on the regulation of SIRT1 and have important implications in understanding the molecular mechanism of ageing and cancer.  相似文献

The magnetic field pervading our Galaxy is a crucial constituent of the interstellar medium: it mediates the dynamics of interstellar clouds, the energy density of cosmic rays, and the formation of stars. The field associated with ionized interstellar gas has been determined through observations of pulsars in our Galaxy. Radio-frequency measurements of pulse dispersion and the rotation of the plane of linear polarization, that is, Faraday rotation, yield an average value for the magnetic field of B approximately 3 microG (ref. 2). The possible detection of Faraday rotation of linearly polarized photons emitted by high-redshift quasars suggests similar magnetic fields are present in foreground galaxies with redshifts z > 1. As Faraday rotation alone, however, determines neither the magnitude nor the redshift of the magnetic field, the strength of galactic magnetic fields at redshifts z > 0 remains uncertain. Here we report a measurement of a magnetic field of B approximately 84 microG in a galaxy at z = 0.692, using the same Zeeman-splitting technique that revealed an average value of B = 6 microG in the neutral interstellar gas of our Galaxy. This is unexpected, as the leading theory of magnetic field generation, the mean-field dynamo model, predicts large-scale magnetic fields to be weaker in the past rather than stronger.  相似文献

研究了改性木质素系外加剂GCL1-M对水泥砂浆物理力学性能和微观结构的影响,结果表明,GCL1-M砂浆减水率达到30%,掺砂浆28天抗压强度比达到了120%。掺GCL1-M砂浆的渗透压力比达到330%,比掺CLS的砂浆提高了1倍多。氮吸附测试和电子扫描显微观测等结果表明,GCL1-M减慢了水泥水化反应放热速度,降低砂浆的总孔容积,阻塞毛细管通道,提高砂浆的抗渗能力。吸附性能、Zeta电位及环境扫描电镜测试结果表明,GCL1-M在水泥颗粒表面形成网状结构吸附,增加了颗粒间的静电斥力位能,增强了对水泥砂浆的分散和保水作用,有利于提高水泥砂浆的抗渗性能和抗压强度。  相似文献
