Drosophila Toll is activated by Gram-positive bacteria through a circulating peptidoglycan recognition protein. 总被引:19,自引:0,他引:19
Microbial infection activates two distinct intracellular signalling cascades in the immune-responsive fat body of Drosophila. Gram-positive bacteria and fungi predominantly induce the Toll signalling pathway, whereas Gram-negative bacteria activate the Imd pathway. Loss-of-function mutants in either pathway reduce the resistance to corresponding infections. Genetic screens have identified a range of genes involved in these intracellular signalling cascades, but how they are activated by microbial infection is largely unknown. Activation of the transmembrane receptor Toll requires a proteolytically cleaved form of an extracellular cytokine-like polypeptide, Sp?tzle, suggesting that Toll does not itself function as a bona fide recognition receptor of microbial patterns. This is in apparent contrast with the mammalian Toll-like receptors and raises the question of which host molecules actually recognize microbial patterns to activate Toll through Sp?tzle. Here we present a mutation that blocks Toll activation by Gram-positive bacteria and significantly decreases resistance to this type of infection. The mutation semmelweis (seml) inactivates the gene encoding a peptidoglycan recognition protein (PGRP-SA). Interestingly, seml does not affect Toll activation by fungal infection, indicating the existence of a distinct recognition system for fungi to activate the Toll pathway. 相似文献

The inventory of water and carbon dioxide reservoirs on Mars are important clues for understanding the geological, climatic and potentially exobiological evolution of the planet. From the early mapping observation of the permanent ice caps on the martian poles, the northern cap was believed to be mainly composed of water ice, whereas the southern cap was thought to be constituted of carbon dioxide ice. However, recent missions (NASA missions Mars Global Surveyor and Odyssey) have revealed surface structures, altimetry profiles, underlying buried hydrogen, and temperatures of the south polar regions that are thermodynamically consistent with a mixture of surface water ice and carbon dioxide. Here we present the first direct identification and mapping of both carbon dioxide and water ice in the martian high southern latitudes, at a resolution of 2 km, during the local summer, when the extent of the polar ice is at its minimum. We observe that this south polar cap contains perennial water ice in extended areas: as a small admixture to carbon dioxide in the bright regions; associated with dust, without carbon dioxide, at the edges of this bright cap; and, unexpectedly, in large areas tens of kilometres away from the bright cap. 相似文献

Stars are spheres of hot gas whose interiors transmit acoustic waves very efficiently. Geologists learn about the interior structure of Earth by monitoring how seismic waves propagate through it and, in a similar way, the interior of a star can be probed using the periodic motions on the surface that arise from such waves. Matthews et al. claim that the star Procyon does not have acoustic surface oscillations of the strength predicted. The relationship between species diversity and ecosystem functioning is a central topic in ecology today. Classical approaches to studying ecosystem responses to nutrient enrichment have considered linear food chains. To what extent ecosystem structure, that is, the network of species interactions, affects such responses is currently unknown. This severely limits our ability to predict which species or functional groups will benefit or suffer from nutrient enrichment and to understand the underlying mechanisms. Here our approach takes ecosystem complexity into account by considering functional diversity at each trophic level. We conducted a mesocosm experiment to test the effects of nutrient enrichment in a lake ecosystem. We developed a model of intermediate complexity, which separates trophic levels into functional groups according to size and diet. This model successfully predicted the experimental results, whereas linear food-chain models did not. Our model shows the importance of functional diversity and indirect interactions in the response of ecosystems to perturbations, and indicates that new approaches are needed for the management of freshwater ecosystems subject to eutrophication. 相似文献

Recurrent chromosomal translocations underlie both haematopoietic and solid tumours. Their origin has been ascribed to selection of random rearrangements, targeted DNA damage, or frequent nuclear interactions between translocation partners; however, the relative contribution of each of these elements has not been measured directly or on a large scale. Here we examine the role of nuclear architecture and frequency of DNA damage in the genesis of chromosomal translocations by measuring these parameters simultaneously in cultured mouse B lymphocytes. In the absence of recurrent DNA damage, translocations between Igh or Myc and all other genes are directly related to their contact frequency. Conversely, translocations associated with recurrent site-directed DNA damage are proportional to the rate of DNA break formation, as measured by replication protein A accumulation at the site of damage. Thus, non-targeted rearrangements reflect nuclear organization whereas DNA break formation governs the location and frequency of recurrent translocations, including those driving B-cell malignancies. 相似文献
