The DNA replication checkpoint response stabilizes stalled replication forks   总被引:62,自引:0,他引:62  
In response to DNA damage and blocks to replication, eukaryotes activate the checkpoint pathways that prevent genomic instability and cancer by coordinating cell cycle progression with DNA repair. In budding yeast, the checkpoint response requires the Mec1-dependent activation of the Rad53 protein kinase. Active Rad53 slows DNA synthesis when DNA is damaged and prevents firing of late origins of replication. Further, rad53 mutants are unable to recover from a replication block. Mec1 and Rad53 also modulate the phosphorylation state of different DNA replication and repair enzymes. Little is known of the mechanisms by which checkpoint pathways interact with the replication apparatus when DNA is damaged or replication blocked. We used the two-dimensional gel technique to examine replication intermediates in response to hydroxyurea-induced replication blocks. Here we show that hydroxyurea-treated rad53 mutants accumulate unusual DNA structures at replication forks. The persistence of these abnormal molecules during recovery from the hydroxyurea block correlates with the inability to dephosphorylate Rad53. Further, Rad53 is required to properly maintain stable replication forks during the block. We propose that Rad53 prevents collapse of the fork when replication pauses.  相似文献

Mammalian mitochondrial DNA (mtDNA) is inherited principally down the maternal line, but the mechanisms involved are not fully understood. Females harboring a mixture of mutant and wild-type mtDNA (heteroplasmy) transmit a varying proportion of mutant mtDNA to their offspring. In humans with mtDNA disorders, the proportion of mutated mtDNA inherited from the mother correlates with disease severity. Rapid changes in allele frequency can occur in a single generation. This could be due to a marked reduction in the number of mtDNA molecules being transmitted from mother to offspring (the mitochondrial genetic bottleneck), to the partitioning of mtDNA into homoplasmic segregating units, or to the selection of a group of mtDNA molecules to re-populate the next generation. Here we show that the partitioning of mtDNA molecules into different cells before and after implantation, followed by the segregation of replicating mtDNA between proliferating primordial germ cells, is responsible for the different levels of heteroplasmy seen in the offspring of heteroplasmic female mice.  相似文献

Cassini's Titan Radar Mapper imaged the surface of Saturn's moon Titan on its February 2005 fly-by (denoted T3), collecting high-resolution synthetic-aperture radar and larger-scale radiometry and scatterometry data. These data provide the first definitive identification of impact craters on the surface of Titan, networks of fluvial channels and surficial dark streaks that may be longitudinal dunes. Here we describe this great diversity of landforms. We conclude that much of the surface thus far imaged by radar of the haze-shrouded Titan is very young, with persistent geologic activity.  相似文献

Self-assembly is emerging as an elegant, 'bottom-up' method for fabricating nanostructured materials. This approach becomes particularly powerful when the ease and control offered by the self-assembly of organic components is combined with the electronic, magnetic or photonic properties of inorganic components. Here we demonstrate a versatile hierarchical approach for the assembly of organic-inorganic, copolymer-metal nanostructures in which one level of self-assembly guides the next. In a first step, ultrathin diblock copolymer films form a regular scaffold of highly anisotropic, stripe-like domains. During a second assembly step, differential wetting guides diffusing metal atoms to aggregate selectively along the scaffold, producing highly organized metal nanostructures. We find that, in contrast to the usual requirement of near-equilibrium conditions for ordering, the metal arranged on the copolymer scaffold produces the most highly ordered configurations when the system is far from equilibrium. We delineate two distinct assembly modes of the metal component-chains of separate nanoparticles and continuous wires-each characterized by different ordering kinetics and strikingly different current-voltage characteristics. These results therefore demonstrate the possibility of guided, large-scale assembly of laterally nanostructured systems.  相似文献

Summary A voltage transient is described which is found during proximal tubular perfusion with impermeant cation or anion salt solutions in the rat. It was shown that the magnitude of transepithelial diffusion potentials depended on luminal hydrostatic pressure, suggesting that the observed transients might be the consequence of the enlargement of ionic pathways by tubular dilatation. Thus, when reporting PD values, care should be taken to define the pressure levels at which measurements were performed. After apoptosis, phagocytes prevent inflammation and tissue damage by the uptake and removal of dead cells. In addition, apoptotic cells evoke an anti-inflammatory response through macrophages. We have previously shown that there is intense lymphocyte apoptosis in an experimental model of Chagas' disease, a debilitating cardiac illness caused by the protozoan Trypanosoma cruzi. Here we show that the interaction of apoptotic, but not necrotic T lymphocytes with macrophages infected with T. cruzi fuels parasite growth in a manner dependent on prostaglandins, transforming growth factor-beta (TGF-beta) and polyamine biosynthesis. We show that the vitronectin receptor is critical, in both apoptotic-cell cytoadherence and the induction of prostaglandin E2/TGF-beta release and ornithine decarboxylase activity in macrophages. A single injection of apoptotic cells in infected mice increases parasitaemia, whereas treatment with cyclooxygenase inhibitors almost completely ablates it in vivo. These results suggest that continual lymphocyte apoptosis and phagocytosis of apoptotic cells by macrophages have a role in parasite persistence in the host, and that cyclooxygenase inhibitors have potential therapeutic application in the control of parasite replication and spread in Chagas' disease.  相似文献
