Indian ricegrass ( Achnatherum hymenoides [Roem. & Schult.] Barkworth) is a rangeland species native to western North America with populations that may exhibit distinct seed-size morphs borne on separate plants. These morphs are often associated with very different levels of seed dormancy. We compared RAPD profiles of the relatively infrequent and highly dormant jumbo seed morph to its more germinable small or large (non-jumbo) companion seed morphs collected from 9 sites where they were sympatric. Jumbo seed accessions from 9 additional sites that did not exhibit seed polymorphism were included to better sample this seed morph across the species' range. Sympatric seed morphs were consistently distinguishable by their RAPD profiles, and sympatric seed morphs were no more likely to cluster together than nonsympatric seed morphs. The ability of sympatric seed morphs to maintain their genetic identity is compatible with the hypothesis of a predominately autogamous breeding system. Furthermore, these data support the hypothesis that local polymorphic populations result from overlapping waves of migration with distinct geographical origins. The jumbo seed morphs exhibited moderate correlation between genetic distance and geographic distance, but the non-jumbo seed morphs exhibited no such correlation. Jumbo seed morphs that were collected at or east of the Continental Divide in New Mexico and Colorado are more genetically homogeneous than those west of the Continental Divide, and non-jumbo seed morphs from the east are more genetically variable than jumbo seed morphs from the same locations. This suggests that gene flow eastward across the Rocky Mountains has been more frequent for the non-jumbo seed morphs than for the jumbo seed morphs.  相似文献

This compilation, containing published and previously unpublished data, shows these ponderosa-pine-dominated stands to be near the lower end of the biomass range of commercial forest types. The two stands averaged approximately 75 metric tons/ha of plant biomass. Consumers made up less than 0.01 percent of the forest biomass. About 9/10 of the measured consumer biomass consisted of domestic and native ruminants.  相似文献

Niche conservatism--the tendency for closely related species to be ecologically similar--is widespread. However, most studies compare closely related taxa that occur in allopatry; in sympatry, the stabilizing forces that promote niche conservatism, and thus inhibit niche shifts, may be countered by natural selection favouring ecological divergence to minimize the intensity of interspecific interactions. Consequently, the relative importance of niche conservatism versus niche divergence in determining community structure has received little attention. Here, we examine a tropical lizard community in which species have a long evolutionary history of ecological interaction. We find that evolutionary divergence overcomes niche conservatism: closely related species are no more ecologically similar than expected by random divergence and some distantly related species are ecologically similar, leading to a community in which the relationship between ecological similarity and phylogenetic relatedness is very weak. Despite this lack of niche conservatism, the ecological structuring of the community has a phylogenetic component: niche complementarity only occurs among distantly related species, which suggests that the strength of ecological interactions among species may be related to phylogeny, but it is not necessarily the most closely related species that interact most strongly.  相似文献

Circular dichroism and 31P-NMR on synthetic oligomers of (dC-dG) inserted within DNA restriction fragments indicate that the right-handed B-structure can exist in close proximity to the left-handed Z-structure. Also, this salt-induced transition to Z-form in a small (dC-dG) segment (1.3%) of a recombinant plasmid markedly influenced the supercoil of the plasmid. These observations have implications for the postulated role of naturally occurring related simple sequences in the regulation of gene activity.  相似文献

The solar wind blows an immense magnetic bubble, the heliosphere, in the local interstellar medium (mostly neutral gas) flowing by the Sun. Recent measurements by Voyager 2 across the termination shock, where the solar wind is slowed to subsonic speeds before entering the heliosheath, found that the shocked solar wind plasma contains only approximately 20 per cent of the energy released by the termination shock, whereas energetic particles above approximately 28 keV contain only approximately 10 per cent; approximately 70 per cent of the energy is unaccounted for, leading to speculation that the unmeasured pickup ions or energetic particles below 28 keV contain the missing energy. Here we report the detection and mapping of heliosheath energetic ( approximately 4-20 keV) neutral atoms produced by charge exchange of suprathermal ions with interstellar neutral atoms. The energetic neutral atoms come from a source approximately 60 degrees wide in longitude straddling the direction of the local interstellar medium. Their energy spectra resemble those of solar wind pickup ions, but with a knee at approximately 11 keV instead of approximately 4 keV, indicating that their parent ions are pickup ions energized by the termination shock. These termination-shock-energized pickup ions contain the missing approximately 70 per cent of the energy dissipated in the termination shock, and they dominate the pressure in the heliosheath.  相似文献

The human heat-shock factor (HSF) regulates heat-shock genes in response to elevated temperature. When human cells are heated to 43 degrees C, HSF is modified post-translationally from a form that does not bind DNA to a form that binds to a specific sequence (the heat-shock element, HSE) found upstream of heat-shock genes. To investigate the transduction of the heat signal to HSF, and more generally, how mammalian cells respond at the molecular level to environmental stimuli, we have developed a cell-free system that exhibits heat-induced activation of human HSF in vitro. Comparison of HSF activation in vitro and in intact cells suggests that the response of human cells to heat shock involves at least two steps. First, an ATP-independent, heat-induced alteration of HSF allows it to bind the HSE; the temperature at which activation occurs in vitro implies that a human factor directly senses temperature. Second, HSF is phosphorylated. It is possible that similar multi-step activation mechanisms play a role in the response of eukaryotic cells to a variety of environmental stimuli, and that these mechanisms evolved to increase the range and flexibility of the response.  相似文献

Crystal structures of Fab antibody fragments determined by X-ray diffraction characteristically feature four-domain, beta-barrel arrangements. A human antibody Fc fragment has also been found to have four beta-barrel domains. The structures of a few intact antibodies have been solved: in two myeloma proteins, the flexible hinge regions that connect the Fc to the Fab segments were deleted so the molecules were non-functional, structurally restrained, T-shaped antibodies; a third antibody, Kol, had no hinge residues missing but the Fc region was sufficiently disordered that it was not possible to relate its disposition accurately with respect to the Fab components. Here we report the structure at 3.5 A resolution of an IgG2a antitumour monoclonal antibody which contains an intact hinge region and was solved in a triclinic crystal by molecular replacement using known Fc and Fab fragments. The antibody is asymmetric, reflecting its dynamic character. There are two local, apparently independent, dyads in the molecule. One relates the heavy chains in the Fc, the other relates the constant domains of the Fabs. The variable domains are not related by this 2-fold axis because of the different Fab elbow angles of 159 degrees and 143 degrees. The Fc has assumed an asymmetric, oblique orientation with respect to loosely tethered yet almost collinear Fabs. Our study enables the two antigen-binding segments as well as the Fc portion of a functional molecule to be visualized and illustrates the flexibility of these immune response proteins.  相似文献

Fibrosis is a pathological form of aberrant tissue repair, the complications of which account for nearly half of all deaths in the industrialized world. All tissues are susceptible to fibrosis under particular pathological sets of conditions. Though each type of fibrosis has characteristics and hallmarks specific to that particular condition, there appear to be common factors underlying fibrotic diseases. One of these ubiquitous factors is the paradigm of the activated myofibroblast in the promotion of fibrotic phenotypes. Recent research has implicated metabolic byproducts of the amino acid tryptophan, namely serotonin and kynurenines, in the pathology or potential pharmacologic therapy of fibrosis, in part through their effects on development of myofibroblast phenotypes. Here, we review literature underlying what is known mechanistically about the effects of these compounds and their respective pathways on fibrosis. Pharmacologic administration of kynurenine improves scarring outcomes in vivo likely not only through its well-characterized immunosuppressive properties but also via its demonstrated antagonism of fibroblast activation and of collagen deposition. In contrast, serotonin directly promotes activation of fibroblasts via activation of canonical TGF-β signaling, and overstimulation with serotonin leads to fibrotic outcomes in vivo. Recently discovered feedback inhibition between serotonin and kynurenine pathways also reveals more information about the cellular physiology of tryptophan metabolism and may also underlie possible paradigms for anti-fibrotic therapy. Together, understanding of the effects of tryptophan metabolism on modulation of fibrosis may lead to the development of new therapeutic avenues for treatment through exploitation of these effects.  相似文献

Glaciers often erode, transport and deposit sediment much more rapidly than nonglacial environments, with implications for the evolution of glaciated mountain belts and their associated sedimentary basins. But modelling such glacial processes is difficult, partly because stabilizing feedbacks similar to those operating in rivers have not been identified for glacial landscapes. Here we combine new and existing data of glacier morphology and the processes governing glacier evolution from diverse settings to reveal such stabilizing feedbacks. We find that the long profiles of beds of highly erosive glaciers tend towards steady-state angles opposed to and slightly more than 50 per cent steeper than the overlying ice-air surface slopes, and that additional subglacial deepening must be enabled by non-glacial processes. Climatic or glaciological perturbations of the ice-air surface slope can have large transient effects on glaciofluvial sediment flux and apparent glacial erosion rate.  相似文献
