Vegetation phenology is an important indicator of climate change impacts on the seasonal dynamics of the biosphere. However, little is known about the influence of elevation on spring phenological sensitivity to temperature in an alpine ecosystem. Based on remotely sensed land surface phenology and temperature data from 2001 to 2010, this study investigated the rate of spring phenological change of the Tibetan Plateau (TP) grasslands in response to interannual temperature variations at different elevations. Results suggest that spring phenology in the TP grasslands exhibits a stronger response to changes in temperature at higher elevations than at lower ones. In particular, spring phenology advanced by 1-2 days in response to a 1 ℃ increase in May average temperature at elevations from 3,000 to 3,500 m, while the rate was up to 8-9 days/℃ at 5,000-5,500 m. Analysis using accumulated growing degree days (AGDD) from January 1 through May 31 showed the same general trend with increased elevation associated with increased sensitivity (as measured by phenological change per unit of AGDD change). Such temperature sensitivity gradients in the TP grasslands could be partly explained by the growth efficiency hypothesis which suggests that vegetation adapted to colder climates likely requires less heat energy for the onset of growing season and vice versa in warmer climates. Furthermore, accumulated growing degree days from January 1 to the greenup date were found to decrease with increasing elevations, which provided evidence to support the applicability of the growth efficiency hypothesis in an alpine grassland ecosystem.  相似文献

In this paper it is argued that human interpretation is an inherently paradoxical and complex mechanism. Human interpretation is underpinned by values, preferences and contrasts, and assumptions, and surfaced through an idiosyncratic combination of personal choice and logic (Pinker, The blank slate: the modern denial of human nature, 2003). In order to find ways through interpretive diversity, Janusian thinking is a conscious and purposeful mechanism (Rothenberg, Creat Res J 9(2–3):207–231, 1996) that allows each one to think paradoxically. Coping with paradoxes is not only a cognitive challenge in trying to resolve the irresolvable but also an emotional one, as emotion might distort the paradox. Janusian attitudinal mapping allows individuals to face the true paradox and to review the assumptions behind it. Such review may modify or even abolish certain assumptions altogether. However, Janusian attitudinal mapping is an emotional undertaking that should follow the three elements involving social reform for advancing and fostering knowledge: shock, open communication and experimentation, and paradox leadership (Lewis, Acad Manage Rev 25(4):760–786, 2000).

The hydrogen atom is one of the most important and influential model systems in modern physics. Attempts to understand its spectrum are inextricably linked to the early history and development of quantum mechanics. The hydrogen atom's stature lies in its simplicity and in the accuracy with which its spectrum can be measured and compared to theory. Today its spectrum remains a valuable tool for determining the values of fundamental constants and for challenging the limits of modern physics, including the validity of quantum electrodynamics and--by comparison with measurements on its antimatter counterpart, antihydrogen--the validity of CPT (charge conjugation, parity and time reversal) symmetry. Here we report spectroscopy of a pure antimatter atom, demonstrating resonant quantum transitions in antihydrogen. We have manipulated the internal spin state of antihydrogen atoms so as to induce magnetic resonance transitions between hyperfine levels of the positronic ground state. We used resonant microwave radiation to flip the spin of the positron in antihydrogen atoms that were magnetically trapped in the ALPHA apparatus. The spin flip causes trapped anti-atoms to be ejected from the trap. We look for evidence of resonant interaction by comparing the survival rate of trapped atoms irradiated with microwaves on-resonance to that of atoms subjected to microwaves that are off-resonance. In one variant of the experiment, we detect 23 atoms that survive in 110 trapping attempts with microwaves off-resonance (0.21 per attempt), and only two atoms that survive in 103 attempts with microwaves on-resonance (0.02 per attempt). We also describe the direct detection of the annihilation of antihydrogen atoms ejected by the microwaves.  相似文献

Mutations in the IDH1 and IDH2 genes encoding isocitrate dehydrogenases are frequently found in human glioblastomas and cytogenetically normal acute myeloid leukaemias (AML). These alterations are gain-of-function mutations in that they drive the synthesis of the ‘oncometabolite’ R-2-hydroxyglutarate (2HG). It remains unclear how IDH1 and IDH2 mutations modify myeloid cell development and promote leukaemogenesis. Here we report the characterization of conditional knock-in (KI) mice in which the most common IDH1 mutation, IDH1(R132H), is inserted into the endogenous murine Idh1 locus and is expressed in all haematopoietic cells (Vav-KI mice) or specifically in cells of the myeloid lineage (LysM-KI mice). These mutants show increased numbers of early haematopoietic progenitors and develop splenomegaly and anaemia with extramedullary haematopoiesis, suggesting a dysfunctional bone marrow niche. Furthermore, LysM-KI cells have hypermethylated histones and changes to DNA methylation similar to those observed in human IDH1- or IDH2-mutant AML. To our knowledge, our study is the first to describe the generation and characterization of conditional IDH1(R132H)-KI mice, and also the first report to demonstrate the induction of a leukaemic DNA methylation signature in a mouse model. Our report thus sheds light on the mechanistic links between IDH1 mutation and human AML.  相似文献

Cases of convergent evolution that involve changes in the same developmental pathway, called parallelism, provide evidence that a limited number of developmental changes are available to evolve a particular phenotype. To our knowledge, in no case are the genetic changes underlying morphological convergence understood. However, morphological convergence is not generally assumed to imply developmental parallelism. Here we investigate a case of convergence of larval morphology in insects and show that the loss of particular trichomes, observed in one species of the Drosophila melanogaster species group, has independently evolved multiple times in the distantly related D. virilis species group. We present genetic and gene expression data showing that regulatory changes of the shavenbaby/ovo (svb/ovo) gene underlie all independent cases of this morphological convergence. Our results indicate that some developmental regulators might preferentially accumulate evolutionary changes and that morphological parallelism might therefore be more common than previously appreciated.  相似文献

The laminin-binding integrin α6β4 plays key roles in both normal epithelial and endothelial cells and during tumor cell progression, metastasis, and angiogenesis. Previous cysteine mutagenesis studies have suggested that palmitoylation of α6β4 protein supports a few integrin-dependent functions and molecular associations. Here we took another approach and obtained strikingly different results. We used overexpression and RNAi knockdown in multiple cell types to identify protein acyl transferase DHHC3 as the enzyme responsible for integrin β4 and α6 palmitoylation. Ablation of DHHC3 markedly diminished integrin-dependent cellular cable formation on Matrigel, integrin signaling through Src, and β4 phosphorylation on key diagnostic amino acids (S1356 and 1424). However, unexpectedly, and in sharp contrast to prior α6β4 mutagenesis results, knockdown of DHHC3 accelerated the degradation of α6β4, likely due to an increase in endosomal exposure to cathepsin D. When proteolytic degradation was inhibited (by Pepstatin A), rescued α6β4 accumulated intracellularly, but was unable to reach the cell surface. DHHC3 ablation effects were strongly selective for α6β4. Cell-surface levels of ~10 other proteins (including α3β1) were not diminished, and the appearance of hundreds of other palmitoylated proteins was not altered. Results obtained here demonstrate a new substrate for the DHHC3 enzyme and provide novel opportunities for modulating α6β4 expression, distribution, and function.  相似文献

We developed a computational method to characterize aneuploidy in tumor samples based on coordinated aberrations in expression of genes localized to each chromosomal region. We summarized the total level of chromosomal aberration in a given tumor in a univariate measure termed total functional aneuploidy. We identified a signature of chromosomal instability from specific genes whose expression was consistently correlated with total functional aneuploidy in several cancer types. Net overexpression of this signature was predictive of poor clinical outcome in 12 cancer data sets representing six cancer types. Also, the signature of chromosomal instability was higher in metastasis samples than in primary tumors and was able to stratify grade 1 and grade 2 breast tumors according to clinical outcome. These results provide a means to assess the potential role of chromosomal instability in determining malignant potential over a broad range of tumors.  相似文献

This paper examines the implications of the Doctrine of Specific Nerve Energies for contemporary philosophy and psychology. Part I analyzed Johannes Peter Müller’s canonical formulation of the Doctrine, arguing that it follows from empirical results combined with methodological principles. Here, I argue that these methodological principles remain valid in psychology today, consequently, any naturalistic philosophy of perception must accept the Doctrine’s skeptical conclusion, that the qualities of our perceptual experience are not determined by, and thus do not reveal the nature of, their causes in the world. Nevertheless, this does not mean that we must be global skeptics; rather, I argue that contemporary epistemology of perception should embrace Müller’s own response to the Doctrine: epistemic structural realism. As articulated by Müller’s student, Helmholtz, structural realism follows from the Doctrine once we recognize that active exploration constitutes part of the mechanism that determines perceptual experience, a view congenial to contemporary theories of embodied perception in cognitive science. Structural realists in philosophy of science should likewise heed the lessons of the Doctrine, as it played a critical part in the early history of their view, and may still serve a constructive role as exemplar today.  相似文献
