Comparative analysis of proteomes using 5-fluorouracil (5-FU)-resistant human colon cancer cell line revealed that decreased
galectin-3 expression was significantly associated with retarded proliferation. However, in the presence of 5-FU proliferation
rate of cells with suppressed galectin-3 expression did not differ from that of cells with normal galectin-3 expression, even
galectin-3 suppression augmented apoptosis. Mechanism by which galectin-3 regulates cancer cell proliferation has been identified
in immunoprecipitates of the anti-galectin-3 antibody. Heterogeneous nuclear ribonucleoprotein Q (hnRNP Q) was identified
as a protein interacting with galectin-3. Interestingly, while galectin-3 protein was not affected by the hnRNP Q level, its
suppression was accompanied by a decrease in hnRNP Q expression. The present study demonstrates that galectin-3 stabilizes
hnRNP Q via complex formation, and reduction in the hnRNP Q level leads to slow proliferation and less susceptibility to 5-FU.

Class 0 protostars, the youngest type of young stellar objects, show many signs of rapid development from their initial, spheroidal configurations, and therefore are studied intensively for details of the formation of protoplanetary disks within protostellar envelopes. At millimetre wavelengths, kinematic signatures of collapse have been observed in several such protostars, through observations of molecular lines that probe their outer envelopes. It has been suggested that one or more components of the proto-multiple system NGC 1333-IRAS 4 (refs 1, 2) may display signs of an embedded region that is warmer and denser than the bulk of the envelope. Here we report observations that reveal details of the core on Solar System dimensions. We detect in NGC 1333-IRAS 4B a rich emission spectrum of H2O, at wavelengths 20-37 microm, which indicates an origin in extremely dense, warm gas. We can model the emission as infall from a protostellar envelope onto the surface of a deeply embedded, dense disk, and therefore see the development of a protoplanetary disk. This is the only example of mid-infrared water emission from a sample of 30 class 0 objects, perhaps arising from a favourable orientation; alternatively, this may be an early and short-lived stage in the evolution of a protoplanetary disk. 相似文献

Telomere dysfunction may result in chromosomal abnormalities, DNA damage responses, and even cancer. Early studies in lower organisms have helped to establish the crucial role of telomerase and telomeric proteins in maintaining telomere length and protecting telomere ends. In Oxytricha nova, telomere G-overhangs are protected by the TEBP-alpha/beta heterodimer. Human telomeres contain duplex telomeric repeats with 3' single-stranded G-overhangs, and may fold into a t-loop structure that helps to shield them from being recognized as DNA breaks. Additionally, the TEBP-alpha homologue, POT1, which binds telomeric single-stranded DNA (ssDNA), associates with multiple telomeric proteins (for example, TPP1, TIN2, TRF1, TRF2 and RAP1) to form the six-protein telosome/shelterin and other subcomplexes. These telomeric protein complexes in turn interact with diverse pathways to form the telomere interactome for telomere maintenance. However, the mechanisms by which the POT1-containing telosome communicates with telomerase to regulate telomeres remain to be elucidated. Here we demonstrate that TPP1 is a putative mammalian homologue of TEBP-beta and contains a predicted amino-terminal oligonucleotide/oligosaccharide binding (OB) fold. TPP1-POT1 association enhanced POT1 affinity for telomeric ssDNA. In addition, the TPP1 OB fold, as well as POT1-TPP1 binding, seemed critical for POT1-mediated telomere-length control and telomere-end protection in human cells. Disruption of POT1-TPP1 interaction by dominant negative TPP1 expression or RNA interference (RNAi) resulted in telomere-length alteration and DNA damage responses. Furthermore, we offer evidence that TPP1 associates with the telomerase in a TPP1-OB-fold-dependent manner, providing a physical link between telomerase and the telosome/shelterin complex. Our findings highlight the critical role of TPP1 in telomere maintenance, and support a yin-yang model in which TPP1 and POT1 function as a unit to protect human telomeres, by both positively and negatively regulating telomerase access to telomere DNA. 相似文献

By adopting majority rule within a renormalization group approach, we can show that the strength of inter-group interaction in a social system either grows or shrinks monotonically with increasing group size, depending on the initial coupling strength of individuals in the group. This contrasts with the findings of previous studies in which the strength of the interaction grows with group size regardless of the initial strength. Our approach clearly demonstrates that the phenomena of crowd psychology, such as an extreme anti-reaction between different ethnic or ideological groups, could be a consequence of the many-particle nature of social systems when the initial strength of the interaction is larger than a critical value. The effect of neutral opinion holders is critically examined using the spin-one Ising model. The critical size of the population of neutral opinion holders, that can prevent crowd polarization, depends on the block spin rule. 相似文献

One of the most commonly found transforming ras oncogenes in human tumours has a valine codon replacing the glycine codon at position 12 of the normal c-Ha-ras gene. To understand the structural reasons behind cell transformation arising from this single amino acid substitution, we have determined the crystal structure of the GDP-bound form of the mutant protein, p21(Val-12), encoded by this oncogene. We report here the overall structure of p21(Val-12) at 2.2 A resolution and compare it with the structure of the normal c-Ha-ras protein. One of the major differences is that the loop of the transforming ras protein that binds the beta-phosphate of the guanine nucleotide is enlarged. Such a change in the 'catalytic site' conformation could explain the reduced GTPase activity of the mutant, which keeps the protein in the GTP bound 'signal on' state for a prolonged period time, ultimately causing cell transformation. 相似文献

The balloon mania between 1783 and 1785 put an extraordinary strain on the Paris Academy of Sciences, threatening its status as the highest tribunal of European science. Faced with repeated royal directives and public frenzy, the Academy manoeuvred carefully to steer the research toward the hydrogen balloon and thereby to maintain its scientific superiority. Antoine-Laurent Lavoisier seized this moment when the promise of ‘the empire of airs’ brought science to the centre of public attention to push his theoretical reform for chemistry. He utilized the established protocol of the Academy and the resources of the Republic of Letters to broadly publicize his first decomposition of water in April 1784. Once he attained the publicity and the balloon fever began to cool, he staged a much more elaborate experiment for the experts in February 1785. Although this second, large-scale experiment cost much expense and effort, Lavoisier never published a full account. The differential care given to the two experiments can only be understood in their divergent rhetorical contexts. The written accounts in 1784 addressed the court of public opinion, while the exclusive performance in 1785 aimed at the court of experts to inaugurate the collaborative phase of the Chemical Revolution. 相似文献
