Radiation damage to proteins is a topic of intense interest to those involved in radiation effects in biology, and also to those involved in radiotherapy. Although it has been widely studied, fundamental processes in protein damage are very hard to specify because of the complexity of the final damage products. But the non-invasive technique of electron-spin resonance is ideally suited to the task of detecting and identifying the primary and secondary products as these are expected to contains unpaired electrons (that is, free-radicals) and such species are uniquely detected by this sensitive form of spectroscopy. Our present study shows that a major radical species formed by electron loss in a range of proteins is the backbone amido radical, -N.(CO)-, characterized by hyperfine coupling to one 14N nucleus. These centres are efficiently trapped in proteins at low temperatures. In contrast, the expected backbone electron-capture centres, -NH(CO.-)-, are not readily trapped and electron transfer occurs until the ejected electron is trapped by some electrophilic centre. Such electron mobility was in fact established in our previous work on oxyhaemoglobin (FeO2----FeO2-), superoxide dismutase (Cu(II)----Cu(I] haemocyanin (Cu(II)O2Cu(I)----Cu(I)O2Cu(II] and various proteins containing S-S bonds (-S-S-)----(-S.-S-) (refs 1-4 respectively). This is strongly supported by our observation that such electrons are captured by DNA molecules, giving T.- centres, when nucleohistones are irradiated, and that Fe(CN)3-(6) ions readily scavenge such electrons from proteins which are devoid of highly electrophilic centres.  相似文献

Résumé Un cas avancé de tumeur canine spontanée et transmissible par voie vénérienne a été examinée au microscope électronique. Ce sarcome est caractérisé par la présence dans les cellules tumérales de rouleaux de rédiculum endoplasmique et de nombreuses inclusions cytoplasmiques. Summary The male antifertility agent -chlorohydrin (I) is metabolized by rat and boar sperm to -chloroactaldehyde (III), -chlorolactate (IV) and Cl and not to the proposed active metabolite, -chlorohydrin-1-phosphate (III). It is proposed that -chlorolactaldehyde is produced intracellularly by a specific enzyme and that this is the metabolite responsible for the species-specific antifertility activity of -chlorohydrin.This work was supported by The National Health and Medical Research Council of Australia.  相似文献

Previous research on the Antikythera Mechanism established a highly complex ancient Greek geared mechanism with front and back output dials. The upper back dial is a 19-year calendar, based on the Metonic cycle, arranged as a five-turn spiral. The lower back dial is a Saros eclipse-prediction dial, arranged as a four-turn spiral of 223 lunar months, with glyphs indicating eclipse predictions. Here we add surprising findings concerning these back dials. Though no month names on the Metonic calendar were previously known, we have now identified all 12 months, which are unexpectedly of Corinthian origin. The Corinthian colonies of northwestern Greece or Syracuse in Sicily are leading contenders-the latter suggesting a heritage going back to Archimedes. Calendars with excluded days to regulate month lengths, described in a first century bc source, have hitherto been dismissed as implausible. We demonstrate their existence in the Antikythera calendar, and in the process establish why the Metonic dial has five turns. The upper subsidiary dial is not a 76-year Callippic dial as previously thought, but follows the four-year cycle of the Olympiad and its associated Panhellenic Games. Newly identified index letters in each glyph on the Saros dial show that a previous reconstruction needs modification. We explore models for generating the unusual glyph distribution, and show how the eclipse times appear to be contradictory. We explain the four turns of the Saros dial in terms of the full moon cycle and the Exeligmos dial as indicating a necessary correction to the predicted eclipse times. The new results on the Metonic calendar, Olympiad dial and eclipse prediction link the cycles of human institutions with the celestial cycles embedded in the Mechanism's gearwork.  相似文献
