Testing the existence of unit root and/or level change is necessary in order to understand the underlying processes of time series. In many studies carried out so far, the focus was only on a single aspect of unit root and level change, therefore limiting a full assessment of the given problems. Our study aims to find a solution to the given problems by testing the two hypotheses simultaneously. We derive the likelihood ratio test statistic based on the state space model, and their distributions are created by the simulation method. The performance of the proposed method is validated by simulated time series and also applied to two Korean macroeconomic time series to confirm its practical application. Actin plays a fundamental role in the regulation of spine morphology (both shrinkage and enlargement) upon synaptic activation. In particular, actin depolymerization is crucial for the spine shrinkage in NMDAR-mediated synaptic depression. Here, we define the role of SPIN90 phosphorylation/dephosphorylation in regulating actin depolymerization via modulation of cofilin activity. When neurons were treated with NMDA, SPIN90 was dephosphorylated by STEP61 (striatal-enriched protein tyrosine phosphatase) and translocated from the spines to the dendritic shafts. In addition, phosphorylated SPIN90 bound cofilin and then inhibited cofilin activity, suggesting that SPIN90 dephosphorylation is a prerequisite step for releasing cofilin so that cofilin can adequately sever actin filaments into monomeric form. We found that SPIN90 YE, a phosphomimetic mutant, remained in the spines after NMDAR activation where it bound cofilin, thereby effectively preventing actin depolymerization. This led to inhibition of the activity-dependent redistribution of cortactin and drebrin A, as well as of the morphological changes in the spines that underlie synaptic plasticity. These findings indicate that NMDA-induced SPIN90 dephosphorylation and translocation initiates cofilin-mediated actin dynamics and spine shrinkage within dendritic spines, thereby modulating synaptic activity. 相似文献

The pygmy rabbit ( Brachylagus idahoensis ) is a secretive, obligate sagebrush-steppe resident of the Intermountain West and is one of two rabbits in North America that digs its own burrows. Although the pygmy rabbit has a recorded home range of 0.21–67.9 ha in relatively high sagebrush cover (21%–36%), they spend much of their time within 30–100 m of a burrow system. Due to big sagebrush cover in preferred habitat and the secretive behavior of pygmy rabbits, it is often difficult to study this leporid through direct observation. We used remote cameras to document pygmy rabbit activity at burrow systems in south central Utah from 2006 to 2008. We analyzed photographs from remote cameras for daily and seasonal patterns of activity. Our results suggested that time of day and season were important influences on activity level, while year and site were less so. Pygmy rabbits were active during all time periods of the day, but the greatest activity occurred in the morning, except during winter. Numerous other species were recorded by remote cameras, including other leporids, birds, rodents, reptiles, and terrestrial predators. Remote cameras are a valuable tool in understanding pygmy rabbit behavior, in addition to confirming rabbit presence in areas of interest. 相似文献

Psychiatric illness, such as affective disorders, anxiety disorders and schizophrenia, exerts exceptional personal burden on affected individuals. Although not physically noticeable, these disorders cost enormously on ones’ family and society. Currently pharma-ceutical and psychological treatments are generally accepted as effective for psychiatric disorders, while the exact mechanisms underlying the treatment efficacy, etiology and neurobiology of the disorders remain elusive. In the past decade, "neurogenic hy-pothesis" emerged as an attempt to explain the nature of psychiatric illness. The origination of the hypothesis is based on several pre-clinical and clinical observations. First, stress, which is a common risk factor of the disorders, was found to suppress neurogenesis; second, treatment for the illnesses like antidepressants and antipsychotics were shown to improve neurogenesis and behavioral deficits simultaneously; and third, the therapeutic effect of antidepressants was abolished in animal models when neuro-genesis was blocked. Increasing efforts were invested to determine whether neurogenesis is a key to the understanding and treatment of psychiatric disorders, although contrasting results are also found and thus the importance of neurogenesis remains a matter of debate. The present chapter will discuss the recent findings about the involvement of neurogenesis in major depression, anxiety disorders and schizophrenia, and whether neurogenesis would be a potential target for development of the treatment in the future. 相似文献

In response to continued low population numbers of Northern Pintail ( Anas acuta ) in North America and to increase knowledge of the geographic variation in pintail survival rates, we estimated 126-day (27 October–2 March) survival for male and female pintails wintering in the Middle Rio Grande Valley, New Mexico, during 2001–2002 (SY1) and 2002–2003 (SY2). Sixty-nine adult male and female pintails were marked with radio-transmitters and tracked throughout the study period. Weekly relocation data in relation to study year, sex, time (week), body condition at capture, and hunting seasons were modeled using the known-fate procedure in Program MARK. Year, sex, time, and body condition covariates did not improve model performance in estimating survival, so we used the most parsimonious model to produce an overall winter survival estimate of 0.597 ± 0.077 (95% C.I. = 0.442–0.735). Weekly survival estimates did not differ between hunting and nonhunting seasons. Male and female point estimates did not differ (χ 1 2 = 0.209, P = 0.65). Our adult female survival estimate of 0.639 ± 0.117 (95% C.I. = 0.396–0.827) was 5.5%–28.6% lower than published estimates for adult female pintails in 5 other geographic regions. No winter survival estimates for males in other geographic regions were available for direct comparison with our study. Although relatively small sample sizes may have contributed to the lack of statistical differences in weekly survival between years, sexes, and hunting seasons, as well as to the lack of influence of body condition, relatively abundant water and food resources and absence of hunting in a refuge setting contributed to consistent survival probabilities. Because our low survival estimates, relative to other geographic regions, cannot be attributed to hunting, we postulate that natural causes of mortality play a larger role in pintail survival in our study region than in other wintering regions. 相似文献

We consider the problem of combining multiple dissimilarity representations via the Cartesian product of their embeddings. For concreteness, we choose the inferential task at hand to be classification. The high dimensionality of this Cartesian product space implies the necessity of dimensionality reduction before training a classifier. We propose a supervised dimensionality reduction method, which utilizes the class label information, to help achieve a favorable combination. The simulation and real data results show that our approach can improve classification accuracy compared to the alternatives of principal components analysis and no dimensionality reduction at all. 相似文献

Missense mutations in the CIAS1 gene cause three autoinflammatory disorders: familial cold autoinflammatory syndrome, Muckle-Wells syndrome and neonatal-onset multiple-system inflammatory disease. Cryopyrin (also called Nalp3), the product of CIAS1, is a member of the NOD-LRR protein family that has been linked to the activation of intracellular host defence signalling pathways. Cryopyrin forms a multi-protein complex termed 'the inflammasome', which contains the apoptosis-associated speck-like protein (ASC) and caspase-1, and promotes caspase-1 activation and processing of pro-interleukin (IL)-1beta (ref. 4). Here we show the effect of cryopyrin deficiency on inflammasome function and immune responses. Cryopyrin and ASC are essential for caspase-1 activation and IL-1beta and IL-18 production in response to bacterial RNA and the imidazoquinoline compounds R837 and R848. In contrast, secretion of tumour-necrosis factor-alpha and IL-6, as well as activation of NF-kappaB and mitogen-activated protein kinases (MAPKs) were unaffected by cryopyrin deficiency. Furthermore, we show that Toll-like receptors and cryopyrin control the secretion of IL-1beta and IL-18 through different intracellular pathways. These results reveal a critical role for cryopyrin in host defence through bacterial RNA-mediated activation of caspase-1, and provide insights regarding the pathogenesis of autoinflammatory syndromes. 相似文献

Formation of electron pairs is essential to superconductivity. For conventional superconductors, tunnelling spectroscopy has established that pairing is mediated by bosonic modes (phonons); a peak in the second derivative of tunnel current d2I/dV2 corresponds to each phonon mode. For high-transition-temperature (high-T(c)) superconductivity, however, no boson mediating electron pairing has been identified. One explanation could be that electron pair formation and related electron-boson interactions are heterogeneous at the atomic scale and therefore challenging to characterize. However, with the latest advances in d2I/dV2 spectroscopy using scanning tunnelling microscopy, it has become possible to study bosonic modes directly at the atomic scale. Here we report d2I/dV2 imaging studies of the high-T(c) superconductor Bi2Sr2CaCu2O8+delta. We find intense disorder of electron-boson interaction energies at the nanometre scale, along with the expected modulations in d2I/dV2 (refs 9, 10). Changing the density of holes has minimal effects on both the average mode energies and the modulations, indicating that the bosonic modes are unrelated to electronic or magnetic structure. Instead, the modes appear to be local lattice vibrations, as substitution of 18O for 16O throughout the material reduces the average mode energy by approximately 6 per cent--the expected effect of this isotope substitution on lattice vibration frequencies. Significantly, the mode energies are always spatially anticorrelated with the superconducting pairing-gap energies, suggesting an interplay between these lattice vibration modes and the superconductivity. 相似文献
