Neuroendocrine disruption of water balance in insect larvae was evaluated as the basis for a new approach to pest insect control. Effects on water balance and food consumption were measured for larvae ofHeliothis virescens (cotton budworm) treated with syntheticManduca sexta diuretic neurohormone (MasDH). Synthetic MasDH (50–250 nM) caused dose-dependent increases in fluid secretion by larval Malpighian tubules in vitro; higher concentrations resulted in lower fluid secretion. Last instar larvae injected with 20–100 pmol MasDH exhibited increasing weight loss. Larvae injected with 500 pmol MasDH dramatically reduced both water excretion and food consumption by 70%. These latter results indicate that high titers of diuretic hormone can suppress feeding damage by larval insects by depressing water excretion and food consumption. 相似文献

Umbellula encrinus Linnaeus, 1758 and Anthoptilum grandiflorum (Verrill, 1879) are two species of deep-water sea pens (Cnidaria: Octocorallia) commonly found in the Northwest Atlantic. These sea pens are frequently caught as fishing bycatch, but little is known about their biology. Here we characterise the internal skeleton (axis) of these two morphologically contrasting species, in terms of axis carbonate composition and proportion, morphology and metrics (i.e. cross-sectional area and weight) through gradients along their lengths. Using the conspicuous growth rings present in the axis of U. encrinus we also estimated intra-colonial variation in radial growth rates for this species. X-ray diffraction (XRD) analyses revealed that the carbonate portion of the axis is composed of magnesian calcite (Ca, Mg)CO3 in both species. On average, the axis is composed of 71% carbonate material in U. encrinus, and 65% carbonate in A. grandiflorum. The carbonate proportion was greatly reduced from base (71%) to tip (33%) in the axis of U. encrinus, while in A. grandiflorum changes were less abrupt (~6% difference from base to tip). In U. encrinus, the axis is quadrangular in shape through its whole length, twisting longitudinally. Twisting can occur in both clockwise and anti-clockwise directions. The axis of A. grandiflorum varied from elliptical near the base to cylindrical through most of the remaining length, with no twisting observed. Axes’ cross-sectional areas also decreased with distance from the base in both species, being the largest near the base. Estimated radial growth rates in U. encrinus were variable within a single colony, ranging between 0.04 and 0.08 mm.yr?1. This is the first study to describe changes along the axes of sea pens, and to characterise the axes in the studied species. Accurate prediction of electrostatic effects on catalytic activity is an essential component of protein design. Site-directed mutagenesis of charged groups in subtilisin of Bacillus amyloliquefaciens has provided experimental measurements of electrostatic interactions which may be used to test such theoretical methods. The pKa of the histidine of the active site has been perturbed by +0.08 to -1.0 units by modifying one or two residues. Electrostatic effects in proteins can be modelled by the algorithm of Warwicker and Watson, which uses classical electrostatics and considers both the charge position and the shape of the molecule. Height is a classic polygenic trait, reflecting the combined influence of multiple as-yet-undiscovered genetic factors. We carried out a meta-analysis of genome-wide association study data of height from 15,821 individuals at 2.2 million SNPs, and followed up the strongest findings in >10,000 subjects. Ten newly identified and two previously reported loci were strongly associated with variation in height (P values from 4 x 10(-7) to 8 x 10(-22)). Together, these 12 loci account for approximately 2% of the population variation in height. Individuals with < or =8 height-increasing alleles and > or =16 height-increasing alleles differ in height by approximately 3.5 cm. The newly identified loci, along with several additional loci with strongly suggestive associations, encompass both strong biological candidates and unexpected genes, and highlight several pathways (let-7 targets, chromatin remodeling proteins and Hedgehog signaling) as important regulators of human stature. These results expand the picture of the biological regulation of human height and of the genetic architecture of this classical complex trait. 相似文献

Syk is a protein tyrosine kinase that is widely expressed in haematopoietic cells. It is involved in coupling activated immunoreceptors to downstream signalling events that mediate diverse cellular responses including proliferation, differentiation and phagocytosis. Syk expression has been reported in cell lines of epithelial origin, but its function in these cells remains unknown. Here we show that Syk is commonly expressed in normal human breast tissue, benign breast lesions and low-tumorigenic breast cancer cell lines. Syk messenger RNA and protein, however, are low or undetectable in invasive breast carcinoma tissue and cell lines. Transfection of wild-type Syk into a Syk-negative breast cancer cell line markedly inhibited its tumour growth and metastasis formation in athymic mice. Conversely, overexpression of a kinase-deficient Syk in a Syk-positive breast cancer cell line significantly increased its tumour incidence and growth. Suppression of tumour growth by the reintroduction of Syk appeared to be the result of aberrant mitosis and cytokinesis. We propose that Syk is a potent modulator of epithelial cell growth and a potential tumour suppressor in human breast carcinomas. 相似文献
