Similarities between metallothionein and low molecular weight testicular cadmium-binding protein

The incorporation of 35S-cysteine and 3H-glutamic acid was studied in mouse hepatic and renal metallothionein and in testicular cadmium-binding protein of similar molecular weight. Preferential incorporation of 35S-cysteine over 3H-glutamic acid was observed not only in hepatic and renal metallothionein, but also in testicular cadmium-binding protein. When the antigenic reactivity of these proteins was compared, all three proteins reacted with the metallothionein antibody. These similarities suggest that the low molecular weight testicular cadmium-binding protein is apparently metallothionein.     

Torus shaped bands at the 2R telomere region and at the region 68 of the salivary gland chromosomes ofDrosophila auraria

Summary The telomere of the 2R arm of the salivary gland chromosomes ofD. auraria exhibits a definite toroidal structure in routine squashed preparations, stained either by propionic orcein-carmine or by fluorescent dyes. There is evidence that a band (or bands) of region 68 (possibly homologous to that ofD. melanogaster) of the 3L chromosome arm also exhibits a toroidal structure. These toroids are associated with heterochromatin, but it is not certain that they are themselves heterochromatic.Acknowledgments. Role for mouse macrophage IgG Fc receptor as ligand-dependent ion channel

The interaction of ligands with the mouse macrophage Fc receptor which binds IgG2b and IgG1 immune complexes (FcR gamma 2b/gamma 1) triggers phagocytosis and secretion of various mediators of inflammation. FcR gamma 2b/gamma 1 has been purified using a monoclonal anti-FcR antibody, 2.4G2, and seems to be an integral membrane glycoprotein of molecular weight (Mr) 47,000-60,000 (ref. 6). Monoclonal antibody 2.4G2 is suitable as a tool for functional studies of FcR because it binds to a functional site of the receptor and induces cellular responses that are normally associated with the occupied receptor. We reported previously that binding of ligands to the macrophage FcR resulted in Na+/K+ ion fluxes through the plasma membrane, and that similar ion fluxes were observed in proteoliposomes containing reconstituted FcR. We have now incorporated FcR into planar lipid bilayers and report here that FcR gamma 2b/gamma 1 forms ligand-dependent cation-selective ion channels, with a conductance of 60 pS in 1 M KCl and an average open channel lifetime of 250 ms. The conductance decays to baseline levels within a few minutes. These results suggest a receptor-ionophore model for the signalling of phagocytosis and inflammatory responses.     

High-resolution (1.5 A) crystal structure of phospholipase C from Bacillus cereus

Both the phosphatidylinositol-hydrolysing and the phosphatidylcholine-hydrolysing phospholipases C have been implicated in the generation of second messengers in mammalian cells. The phosphatidylcholine-hydrolysing phospholipase C (PLC) from Bacillus cereus, a monomeric protein containing 245 amino-acid residues, is similar to some of the corresponding mammalian proteins. This, together with the fact that the bacterial enzyme can mimic the action of mammalian PLC in causing, for example, enhanced prostaglandin biosynthesis, suggests that B. cereus PLC can be used as a model for the hitherto poorly characterized mammalian PLCs. We report here the three-dimensional structure of B. cereus PLC at 1.5 A resolution. The enzyme is an all-helix protein belonging to a novel structural class and contains, at least in the crystalline state, three Zn2+ in the active site. We also present preliminary results from a study at 1.9 A resolution of the complex between PLC and inorganic phosphate (Pi) which indicate that the substrate binds directly to the metal ions.     

Novel and sensitive noncompetitive enzyme immunoassay for kassinin in rat plasma

A novel and sensitive noncompetitive enzyme immunoassay for kassinin is described. Kassinin was biotinylated using sulfosuccinimidyl-6-(biotinamido)hexanoate. The biotinylated kassinin was trapped on anti-kassinin IgG-coated polystyrene balls and, after washing to eliminate other biotinylated substances, was eluted with HCl. The biotinylated kassinin eluted was reacted with anti-kassinin Fab'-peroxidase conjugate and trapped onto streptavidin-coated polystyrene balls. Peroxidase activity bound to the polystyrene balls was assayed by fluorimetry. The detection limit of kassinin was 0.13 pg (0.1 fmol)/tube or 0.065 microgram/l of rat plasma, which was 750-fold or 15-fold lower than that for competitive radioimmunoassay.

Tendon shortening in striated muscle.

Tendon shortening in the digastric muscle of the rabbit resulted in a short term increase in gross muscle length and sarcomere length. Subsequently, muscle and sarcomere lengths decreased to less than control values. Long-term measurements suggested that a return to control sarcomere length may have been achieved by a reduction in sarcomere number.
