松江鲈MRF4基因克隆与组织表达分析

MRF4(myf6或herculin)基因属于MyoD家族成员之一,为成肌细胞分化成肌管的重要调控因子.本实验以松江鲈(Trachidermus fasciatus)肌肉总RNA为模板,采用RT-PCR结合RACE的方法,获得1 049 bp全长cDNA序列,包含了1个含有720个核苷酸的开放性阅读框(ORF),共编码239个氨基酸.其编码区序列与石斑鱼同源性最高、为91.81%,其次是东方红鳍鲀、为83.40%,与哺乳类、鸟类的同源性在65%左右,与爪蟾同源性最低仅为59.65%.预测松江鲈MRF4蛋白具有保守的HLH结构域,bHLH结构域形成剪刀状α-螺旋二聚体.用荧光定量PCR方法检测发现,MRF4基因的mRNA在松江鲈6种组织中均有表达,但其在肌肉组织中表达量极显著高于其他组织,提示其在肌肉的生长发育过程中起着重要作用.     

Mutations in IHH, encoding Indian hedgehog, cause brachydactyly type A-1.

Brachydactyly type A-1 (BDA-1; MIM 112500) is characterized by shortening or missing of the middle phalanges (Fig. 1a). It was first identified by Farabee in 1903 (ref. 2), is the first recorded example of a human anomaly with Mendelian autosomal-dominant inheritance and, as such, is cited in most genetic and biological textbooks. Here we show that mutations in IHH, which encodes Indian hedgehog, cause BDA-1. We have identified three heterozygous missense mutations in the region encoding the amino-terminal signaling domain in all affected members of three large, unrelated families. The three mutant amino acids, which are conserved across all vertebrates and invertebrates studied so far, are predicted to be adjacent on the surface of IHH.

Mechanism of oxidative damage to DNA by Fe-loaded MCM-41 irradiated with visible light

The mechanism of oxidative damage to deoxyribonucleic acid (DNA) by iron-containing mesoporous molecular sieves (MCM-41) irradiated with visible light was elucidated. Fe-loaded MCM-41 (Fe/MCM-41) was used as a photocatalyst and the damage to calf thymus DNA caused by hydrogen peroxide (H2O2) was studied. The damage and extent of oxidation of DNA were measured by high-performance liquid chromatography (HPLC) and intermediate products were detected by HPLC/electrospray ionization tandem mass spectrometry. Electron spin resonance was used to detect changes in reactive oxygen species and peroxidase catalytic spectrophotometry was used to determine the concentration of H2O2. The results indicated that Fe/MCM-41 efficiently activated H2O2 in solution at pH 4.0-8.0 under irradiation with visible light. The photocatalytic system degraded DNA most effectively at pH 5.0-6.0 but also operated at pH 8.0. At pH 4.2, the degree of DNA damage reached 25.65% after 5 h and the kinetic constant was 5.89×10 2 min 1. Damage to DNA was predominantly caused by hydroxyl radicals generated in the system. The mechanism of DNA damage is of potential concern to human health because it can occur in neutral solutions irradiated by visible light.

A nonmonotone line search filter method with reduced Hessian updating for nonlinear optimization

This paper proposes a nonmonotone line search filter method with reduced Hessian updating for solving nonlinear equality constrained optimization. In order to deal with large scale problems, a reduced Hessian matrix is approximated by BFGS updates. The new method assures global convergence without using a merit function. By Lagrangian function in the filter and nonmonotone scheme, the authors prove that the method can overcome Maratos effect without using second order correction step so that the locally superlinear convergence is achieved. The primary numerical experiments are reported to show effectiveness of the proposed algorithm.
