The comparison of related genomes has emerged as a powerful lens for genome interpretation. Here we report the sequencing and comparative analysis of 29 eutherian genomes. We confirm that at least 5.5% of the human genome has undergone purifying selection, and locate constrained elements covering ~4.2% of the genome. We use evolutionary signatures and comparisons with experimental data sets to suggest candidate functions for ~60% of constrained bases. These elements reveal a small number of new coding exons, candidate stop codon readthrough events and over 10,000 regions of overlapping synonymous constraint within protein-coding exons. We find 220 candidate RNA structural families, and nearly a million elements overlapping potential promoter, enhancer and insulator regions. We report specific amino acid residues that have undergone positive selection, 280,000 non-coding elements exapted from mobile elements and more than 1,000 primate- and human-accelerated elements. Overlap with disease-associated variants indicates that our findings will be relevant for studies of human biology, health and disease.  相似文献

This paper presents an open-ended, exploratory study of formal and informal communication and control subprocesses during information systems development (ISD) over an extended period in a large bureaucracy. Using a participant action research strategy, the case study explores the continuous unintended disruptions and barriers to the effective implementation of the ISD project and suggests major variables associated with them. Based on an inductive analysis of the data, the authors suggest some tentatively hypothesized action stages for future research and a broad conceptual model incorporating the process variables. The study of mammalian evolution depends greatly on understanding the evolution of teeth and the relationship of tooth shape to diet. Links between gross tooth shape, function and diet have been proposed since antiquity, stretching from Aristotle to Cuvier, Owen and Osborn. So far, however, the possibilities for exhaustive, quantitative comparisons between greatly different tooth shapes have been limited. Cat teeth and mouse teeth, for example, are fundamentally distinct in shape and structure as a result of independent evolutionary change over tens of millions of years. There is difficulty in establishing homology between their tooth components or in summarizing their tooth shapes, yet both carnivorans and rodents possess a comparable spectrum of dietary specializations from animals to plants. Here we introduce homology-free techniques to measure the phenotypic complexity of the three-dimensional shape of tooth crowns. In our geographic information systems (GIS) analysis of 441 teeth from 81 species of carnivorans and rodents, we show that the surface complexity of tooth crowns directly reflects the foods they consume. Moreover, the absolute values of dental complexity for individual dietary classes correspond between carnivorans and rodents, illustrating a high-level similarity between overall tooth shapes despite a lack of low-level similarity of specific tooth components. These results suggest that scale-independent forces have determined the high-level dental shape in lineages that are widely divergent in size, ecology and life history. This link between diet and phenotype will be useful for inferring the ecology of extinct species and illustrates the potential of fast-throughput, high-level analysis of the phenotype.  相似文献

An investigation to elucidate some facets of the biology and ecology of Paruroctonus boreus (Girard), in southeast Idaho, was conducted from 21 March to 13 October 1972. Three hundred and eighty - five observations were made on 202 scorpions captured in covered pitfalls and by ultraviolet light. The greatest activity occurred at 2130 hours. Surface activity was not observed when ambient temperature was below 10 C, and, as ambient temperature increased, an increase in surface activity was noted. Male scorpions frequently moved as much as 30 m between observations, while females seldom moved more than five m. Sex ratios varied considerably depending on time of year and method of sampling. Of 142 scoipions observed by blacklight, 92 were males, and 50 were females (1.84 males/female). Limited data suggest a mean litter size of 34 ± 11.  相似文献

Centrosomes are organelles involved in generating and organizing the interphase microtubule cytoskeleton, mitotic spindles and cilia. At the centrosome core are a pair of centrioles, structures that act as the duplicating elements of this organelle. Centrioles function to recruit and organize pericentriolar material which nucleates microtubules. While centrioles are relatively simple in construction, the mechanics of centriole biogenesis remain an important yet poorly understood process. More mysterious still are the regulatory mechanisms that oversee centriole assembly. The fidelity of centriole duplication is critical as defects in either the assembly or number of centrioles promote aneuploidy, primary microcephaly, birth defects, ciliopathies and tumorigenesis. In addition, some pathogens employ mechanisms to promote centriole overduplication to the detriment of the host cell. This review summarizes our current understanding of this important topic, highlighting the need for further study if new therapeutics are to be developed to treat diseases arising from defects of centrosome duplication.  相似文献

We studied nest success, burrow longevity, and rates of burrow reuse for a migratory population of Burrowing Owl ( Athene cunicularia ) in north central Oregon from 1995 to 1997. Nest success varied annually from 50% to 67%. Principal causes of nest failure were desertion (26%) and depredation by badgers ( Taxidea taxus ; 13%). Reuse of available nest and satellite burrows in subsequent years was 87% in 1996 and 57% in 1997. Reuse was highest at burrows in sandy soils, which may indicate that nest-site availability is a limiting factor in sandier soil types. Trampling by livestock resulted in the loss of 24% of all burrows between one season and the next, and natural erosion resulted in closure of 17%. Both causes of burrow failure occurred more frequently in soils with a sand component due to their friable nature. We recommend that habitat used by livestock be evaluated for use by Burrowing Owls, that occupied areas be managed to minimize destruction of burrows by livestock, and that predator-control efforts be revised to exclude mortality of badgers.  相似文献

We investigated interference and exploitative competition between endangered San Joaquin kit foxes ( Vulpes macrotis mutica ) and nonnative red foxes ( V. vulpes ). Seven kit foxes and 16 red foxes were radio-collared and tracked via radiotelemetry near Lost Hills, California. One kit fox was killed by a red fox. Home ranges of the 2 species did not overlap extensively. Although both species used similar habitats, they used different parcels of land. Kit foxes and red foxes primarily consumed rodents on the study site, and dietary overlap was considerable. Red foxes also may have been using dens formerly used by kit foxes. Thus, red foxes were engaging in both interference and exploitative competition with kit foxes, and red foxes constitute a potentially significant threat to kit foxes. Coyotes ( Canis latrans ) co-occur with kit foxes and may limit red fox abundance and distribution. Therefore, although they occasionally kill kit foxes, the presence of coyotes may benefit kit foxes by excluding red foxes.  相似文献
