Infections localized to peripheral tissues such as the skin result in the priming of T-cell responses that act to control pathogens. Activated T cells undergo migrational imprinting within the draining lymph nodes, resulting in memory T cells that provide local and systemic protection. Combinations of migrating and resident memory T cells have been implicated in long-term peripheral immunity, especially at the surfaces that form pathogen entry points into the body. However, T-cell immunity consists of separate CD4(+) helper T cells and CD8(+) killer T cells, with distinct effector and memory programming requirements. Whether these subsets also differ in their ability to form a migrating pool involved in peripheral immunosurveillance or a separate resident population responsible for local infection control has not been explored. Here, using mice, we show key differences in the migration and tissue localization of memory CD4(+) and CD8(+) T cells following infection of the skin by herpes simplex virus. On resolution of infection, the skin contained two distinct virus-specific memory subsets; a slow-moving population of sequestered CD8(+) T cells that were resident in the epidermis and confined largely to the original site of infection, and a dynamic population of CD4(+) T cells that trafficked rapidly through the dermis as part of a wider recirculation pattern. Unique homing-molecule expression by recirculating CD4(+) T effector-memory cells mirrored their preferential skin-migratory capacity. Overall, these results identify a complexity in memory T-cell migration, illuminating previously unappreciated differences between the CD4(+) and CD8(+) subsets. 相似文献

Summary Microbial activity in mixed wastes can have an appreciable effect on the dissolution or precipitation of toxic metals and radionuclides. Fundamental information on microbial dissolution and stabilization (immobilization) of toxic metals and radionuclides, in particular actinides and fission products, in nuclear wastes under various microbial process conditions, e.g., aerobic, denitrifying, iron-reducing, fermentative, sulfate-reducing, and methanogenic conditions is very limited. Microbial transformations of typical waste components such as metal oxides, metal coprecipitates, naturally occurring minerals, and metal organic complexes are reviewed. Such information can be useful in the development of 1) predictive models on the fate and long-term transport of toxic metals and radionuclides from waste disposal sites, and 2) biotechnological applications of waste treatment leading to volume reduction and stabilization as wall as recovery and recycling of radionuclides and toxic metals. 相似文献

MutL homolog 3 (Mlh3) is a member of a family of proteins conserved during evolution and having dual roles in DNA mismatch repair and meiosis. The pathway in eukaryotes consists of the DNA-binding components, which are the homologs of the bacterial MutS protein (MSH 2 6), and the MutL homologs, which bind to the MutS homologs and are essential for the repair process. Three of the six homologs of MutS that function in these processes, Msh2, Msh3 and Msh6, are involved in the mismatch repair of mutations, frameshifts and replication errors, and two others, Msh4 and Msh5, have specific roles in meiosis. Of the four MutL homologs, Mlh1, Mlh3, Pms1 and Pms2, three are involved in mismatch repair and at least two, Pms2 and Mlh1, are essential for meiotic progression in both yeast and mice. To assess the role of Mlh3 in mammalian meiosis, we have generated and characterized Mlh3(-/-) mice. Here we show that Mlh3(-/-) mice are viable but sterile. Mlh3 is required for Mlh1 binding to meiotic chromosomes and localizes to meiotic chromosomes from the mid pachynema stage of prophase I. Mlh3(-/-) spermatocytes reach metaphase before succumbing to apoptosis, but oocytes fail to complete meiosis I after fertilization. Greenhouse gases and tropospheric sulphate aerosols--the main human influences on climate--have been shown to have had a detectable effect on surface air temperature, the temperature of the free troposphere and stratosphere and ocean temperature. Nevertheless, the question remains as to whether human influence is detectable in any variable other than temperature. Here we detect an influence of anthropogenic greenhouse gases and sulphate aerosols in observations of winter sea-level pressure (December to February), using combined simulations from four climate models. We find increases in sea-level pressure over the subtropical North Atlantic Ocean, southern Europe and North Africa, and decreases in the polar regions and the North Pacific Ocean, in response to human influence. Our analysis also indicates that the climate models substantially underestimate the magnitude of the sea-level pressure response. This discrepancy suggests that the upward trend in the North Atlantic Oscillation index (corresponding to strengthened westerlies in the North Atlantic region), as simulated in a number of global warming scenarios, may be too small, leading to an underestimation of the impacts of anthropogenic climate change on European climate. 相似文献
