Ablation of the left cerebral cortex abrogates the production of thymic hormone, reduces the number of spleen T cells and impairs immunization with sheep erythrocytes. In addition, partial decortication inhibits the ability of sodium diethyldithiocarbamate (DTC) to increase the level of circulating thymic hormone, as well as the number of splenic T cells. Therefore, the cerebral cortex would display an important role to maintain body integrity and relations with the external environment, through its effects on the immune system.  相似文献

Summary Acid hydrolysis of Cuvier's organs alkoholic extract of sea- cucumber,Bohadschia vitiensis, led to many sapogenins. The first 3 isolated compounds had already been isolated from another sea-cucumber species/seychellogenin, 22,25-oxido-holothurinogenin and 24,25-dehydro holothurinogenin. The cerebral cortex develops through the coordinated generation of dozens of neuronal subtypes, but the mechanisms involved remain unclear. Here we show that mouse embryonic stem cells, cultured without any morphogen but in the presence of a sonic hedgehog inhibitor, recapitulate in vitro the major milestones of cortical development, leading to the sequential generation of a diverse repertoire of neurons that display most salient features of genuine cortical pyramidal neurons. When grafted into the cerebral cortex, these neurons develop patterns of axonal projections corresponding to a wide range of cortical layers, but also to highly specific cortical areas, in particular visual and limbic areas, thereby demonstrating that the identity of a cortical area can be specified without any influence from the brain. The discovery of intrinsic corticogenesis sheds new light on the mechanisms of neuronal specification, and opens new avenues for the modelling and treatment of brain diseases.  相似文献

Free fatty acids provide an important energy source as nutrients, and act as signalling molecules in various cellular processes. Several G-protein-coupled receptors have been identified as free-fatty-acid receptors important in physiology as well as in several diseases. GPR120 (also known as O3FAR1) functions as a receptor for unsaturated long-chain free fatty acids and has a critical role in various physiological homeostasis mechanisms such as adipogenesis, regulation of appetite and food preference. Here we show that GPR120-deficient mice fed a high-fat diet develop obesity, glucose intolerance and fatty liver with decreased adipocyte differentiation and lipogenesis and enhanced hepatic lipogenesis. Insulin resistance in such mice is associated with reduced insulin signalling and enhanced inflammation in adipose tissue. In human, we show that GPR120 expression in adipose tissue is significantly higher in obese individuals than in lean controls. GPR120 exon sequencing in obese subjects reveals a deleterious non-synonymous mutation (p.R270H) that inhibits GPR120 signalling activity. Furthermore, the p.R270H variant increases the risk of obesity in European populations. Overall, this study demonstrates that the lipid sensor GPR120 has a key role in sensing dietary fat and, therefore, in the control of energy balance in both humans and rodents.  相似文献

Climate modeling is closely tied, through its institutions and practices, to observations from satellites and to the field sciences. The validity, quality and scientific credibility of models are based on interaction between models and observation data. In the case of numerical modeling of climate and climate change, validation is not solely a scientific interest: the legitimacy of computer modeling, as a tool of knowledge, has been called into question in order to deny the reality of any anthropogenic climate change; model validations thereby bring political issues into play as well. There is no systematic protocol of validation: one never validates a model in general, but the capacity of a model to account for a defined climatic phenomenon or characteristic. From practices observed in the two research centers developing and using a climate model in France, this paper reviews different ways in which the researchers establish links between models and empirical data (which are not reduced to the latter validating the former) and convince themselves that their models are valid. The analysis of validation practices—relating to parametrization, modes of variability, climatic phenomena, etc.—allows us to highlight some elements of the epistemology of modeling.  相似文献
