Immobile survival of motoneuron (SMN) protein stored in Cajal bodies can be mobilized by protein interactions

Reduced levels of survival of motoneuron (SMN) protein lead to spinal muscular atrophy, but it is still unknown how SMN protects motoneurons in the spinal cord against degeneration. In the nucleus, SMN is associated with two types of nuclear bodies denoted as gems and Cajal bodies (CBs). The 23 kDa isoform of fibroblast growth factor-2 (FGF-2²³) is a nuclear protein that binds to SMN and destabilizes the SMN-Gemin2 complex. In the present study, we show that FGF-2²³ depletes SMN from CBs without affecting their general structure. FRAP analysis of SMN-EGFP in CBs demonstrated that the majority of SMN in CBs remained mobile and allowed quantification of fast, slow and immobile nuclear SMN populations. The potential for SMN release was confirmed by in vivo photoconversion of SMN-Dendra2, indicating that CBs concentrate immobile SMN that could have a specialized function in CBs. FGF-2²³ accelerated SMN release from CBs, accompanied by a conversion of immobile SMN into a mobile population. Furthermore, FGF-2²³ caused snRNP accumulation in CBs. We propose a model in which Cajal bodies store immobile SMN that can be mobilized by its nuclear interaction partner FGF-2²³, leading to U4 snRNP accumulation in CBs, indicating a role for immobile SMN in tri-snRNP assembly.     

Ablation of cyclase-associated protein 2 (CAP2) leads to cardiomyopathy

Cyclase-associated proteins are highly conserved proteins that have a role in the regulation of actin dynamics. Higher eukaryotes have two isoforms, CAP1 and CAP2. To study the in vivo function of CAP2, we generated mice in which the CAP2 gene was inactivated by a gene-trap approach. Mutant mice showed a decrease in body weight and had a decreased survival rate. Further, they developed a severe cardiac defect marked by dilated cardiomyopathy (DCM) associated with drastic reduction in basal heart rate and prolongations in atrial and ventricular conduction times. Moreover, CAP2-deficient myofibrils exhibited reduced cooperativity of calcium-regulated force development. At the microscopic level, we observed disarrayed sarcomeres with development of fibrosis. We analyzed CAP2’s role in actin assembly and found that it sequesters G-actin and efficiently fragments filaments. This activity resides completely in its WASP homology domain. Thus CAP2 is an essential component of the myocardial sarcomere and is essential for physiological functioning of the cardiac system, and a deficiency leads to DCM and various cardiac defects.     

History of fish hatchery development in the Great Basin states of Utah and Nevada

Fish hatchery systems in both Utah and Nevada are now an integral part of the fishery management scheme. Historic development of hatcheries, including the early stocking of exotics, is presented. Disease control and dry pelleted feed are discussed in historical perspective and present status.     

Fall diet of blue grouse in Oregon

The early fall diet of Oregon blue grouse ( Dendragapus obscurus pallidus ) from Wallowa County, Oregon, was determined from 145 crops obtained during 1981 and 1982. Of more than 50 plant and animal foods in the diet, short-horned grasshoppers ( Acrididae ), prickly lettuce ( Lactuca serriola ), yellow salsify ( Tragopogon dubius ), wild buck-wheat ( Eriogonum spp.), and snowberry ( Symphoricarpos albus ) occurred in 30% or more of the crops and collectively amounted to 68% of the diet by weight. Seven of the 12 most common foods were consumed differentially by the four sex and age classes of birds. Results indicated that blue grouse foraged in forest and grassland habitats.     

Buccal floor of reptiles, a summary

A general survey of the information presently available on the osteology and myology of the hyobranchial apparatus. Included in the survey are examples of the hyobranchial skeleton of the major groups of reptiles, including the Chelonia, Crocodilia, Rhynchocephalia, and Squamata. The myology treats the muscles directly associated with the hyoid as well as those associated with the functioning of the apparatus, but not arising or inserted directly on or from the hyoid. The innervation of the hyobranchial apparatus is reviewed and briefly discussed based on the information available in a few major studies. An attempt is made to cite all pertinent literature references, and in Tables 1 and 2 the references to basic areas are indicated. Twenty - nine plates and figures are included, some of which represent original research.     

Biomass of fifty conifer forests and nutrient exports associated with their harvest

Total biomass was approximately 1.5 times the biomass of normally merchantable boles. When compared with conventional bole harvest, the nutrient exports associated with harvest of all aboveground parts in these stands would apparently be at least three times higher for nitrogen, six times higher for phosphorus, four times higher for potassium, and three times higher for calcium.        

The effects of radiation from uranium mill tailings on Tradescantia

To assess the potential hazard from radioactive wastes, a plant with known radiosensitivity, Tradescantia clone 02, was grown in radioactive soil obtained from uranium mill tailings. The levels of radiation on these tailings varied from 0.03mR/hr (background) to a maximum of 3.00 mR/hr. Tradescantia grown in soil with a radiation level greater than 0.10 mR/hr evidenced significantly reduced reproductive integrity and fecundity, as measured by the number of stunted hairs per stamen, pollen viability, and numbers of somatic mutations. Based on these data, the radioactivity from uranium mill tailings has the potential to alter normal plant succession due to its detrimental effect on any species that is relatively radiosensitive.
