MYH9 is a major-effect risk gene for focal segmental glomerulosclerosis   总被引:1,自引:0,他引:1  
The increased burden of chronic kidney and end-stage kidney diseases (ESKD) in populations of African ancestry has been largely unexplained. To identify genetic variants predisposing to idiopathic and HIV-1-associated focal segmental glomerulosclerosis (FSGS), we carried out an admixture-mapping linkage-disequilibrium genome scan on 190 African American individuals with FSGS and 222 controls. We identified a chromosome 22 region with a genome-wide logarithm of the odds (lod) score of 9.2 and a peak lod of 12.4 centered on MYH9, a functional candidate gene expressed in kidney podocytes. Multiple MYH9 SNPs and haplotypes were recessively associated with FSGS, most strongly a haplotype spanning exons 14 through 23 (OR = 5.0, 95% CI = 3.5-7.1; P = 4 x 10(-23), n = 852). This association extended to hypertensive ESKD (OR = 2.2, 95% CI = 1.5-3.4; n = 433), but not type 2 diabetic ESKD (n = 476). Genetic variation at the MYH9 locus substantially explains the increased burden of FSGS and hypertensive ESKD among African Americans.  相似文献

Circuits based on organic semiconductors are being actively explored for flexible, transparent and low-cost electronic applications. But to realize such applications, the charge carrier mobilities of solution-processed organic semiconductors must be improved. For inorganic semiconductors, a general method of increasing charge carrier mobility is to introduce strain within the crystal lattice. Here we describe a solution-processing technique for organic semiconductors in which lattice strain is used to increase charge carrier mobilities by introducing greater electron orbital overlap between the component molecules. For organic semiconductors, the spacing between cofacially stacked, conjugated backbones (the π-π stacking distance) greatly influences electron orbital overlap and therefore mobility. Using our method to incrementally introduce lattice strain, we alter the π-π stacking distance of 6,13-bis(triisopropylsilylethynyl) pentacene (TIPS-pentacene) from 3.33?? to 3.08??. We believe that 3.08?? is the shortest π-π stacking distance that has been achieved in an organic semiconductor crystal lattice (although a π-π distance of 3.04?? has been achieved through intramolecular bonding). The positive charge carrier (hole) mobility in TIPS-pentacene transistors increased from 0.8?cm(2)?V(-1)?s(-1) for unstrained films to a high mobility of 4.6?cm(2)?V(-1)?s(-1) for a strained film. Using solution processing to modify molecular packing through lattice strain should aid the development of high-performance, low-cost organic semiconducting devices.  相似文献

This paper considers Kant’s conception of force and causality in his early pre-Critical writings, arguing that this conception is best understood by way of contrast with his immediate predecessors, such as Christian Wolff, Alexander Baumgarten, Georg Friedrich Meier, Martin Knutzen, and Christian August Crusius, and in terms of the scientific context of natural philosophy at the time. Accordingly, in the True estimation Kant conceives of force in terms of activity rather than in terms of specific effects, such as motion (as unnamed Wolffians had done). Kant’s explicit arguments in the Nova dilucidatio for physical influx (in the guise of the principle of succession) are directed primarily against the conception of grounds and existence held by Wolff, Baumgarten, and Meier, and only secondarily against Leibniz (by asserting the priority of bodies over mind rather than vice versa). Finally, Kant’s reconciliation of the infinite divisibility of space and the unity of monads in the Physical monadology is designed to respond to objections that could be raised naturally by Wolff and Baumgarten.  相似文献
