There is a clear case for drug treatments to be selected according to the characteristics of an individual patient, in order to improve efficacy and reduce the number and severity of adverse drug reactions. However, such personalization of drug treatments requires the ability to predict how different individuals will respond to a particular drug/dose combination. After initial optimism, there is increasing recognition of the limitations of the pharmacogenomic approach, which does not take account of important environmental influences on drug absorption, distribution, metabolism and excretion. For instance, a major factor underlying inter-individual variation in drug effects is variation in metabolic phenotype, which is influenced not only by genotype but also by environmental factors such as nutritional status, the gut microbiota, age, disease and the co- or pre-administration of other drugs. Thus, although genetic variation is clearly important, it seems unlikely that personalized drug therapy will be enabled for a wide range of major diseases using genomic knowledge alone. Here we describe an alternative and conceptually new 'pharmaco-metabonomic' approach to personalizing drug treatment, which uses a combination of pre-dose metabolite profiling and chemometrics to model and predict the responses of individual subjects. We provide proof-of-principle for this new approach, which is sensitive to both genetic and environmental influences, with a study of paracetamol (acetaminophen) administered to rats. Puzzlingly, the parent bodies of ordinary chondrites (the most abundant type of meteorites) do not seem to be abundant among asteroids. One possible explanation is that surfaces of the parent bodies become optically altered, to become the S-type asteroids which are abundant in the main asteroid belt. The process is called 'space weathering'-it makes the visible and near-infrared reflectance spectrum of a body darker and redder. A recent survey of small, near-Earth asteroids suggests that the surfaces of small S asteroids may have developing stages of space weathering. Here we report that a dark region on a small (550-metre) asteroid-25143 Itokawa-is significantly more space-weathered than a nearby bright region. Spectra of both regions are consistent with those of LL5-6 chondrites after continuum removal. A simple calculation suggests that the dark area has a shorter mean optical path length and about 0.04 per cent by volume more nanophase metallic iron particles than the bright area. This clearly shows that space-weathered materials accumulate on small asteroids, which are likely to be the parent bodies of LL chondrites. We conclude that, because LL meteorites are the least abundant of ordinary (H, L, and LL) chondrites, there must be many asteroids with ordinary-chondrite compositions in near-Earth orbits. 相似文献
In French mechanical treatises of the nineteenth century, Newton’s second law of motion was frequently derived from a relativity principle. The origin of this trend is found in ingenious arguments by Huygens and Laplace, with intermediate contributions by Euler and d’Alembert. The derivations initially relied on Galilean relativity and impulsive forces. After Bélanger’s Cours de mécanique of 1847, they employed continuous forces and a stronger relativity with respect to any commonly impressed motion. The name “principle of relative motions” and the very idea of using this principle as a constructive tool were born in this context. The consequences of Poincaré’s and Einstein’s awareness of this approach are analyzed. Lastly, the legitimacy and significance of a relativity-based derivation of Newton’s second law are briefly discussed in a more philosophical vein.
With the development of nanotechnology, many new optical phenomena in nanoscale have been demonstrated. Through the coupling of optical waves and collective oscillations of free electrons in metallic nanostructures, surface plasmon polaritons can be excited accompanying a strong near field enhancement that decays in a subwavelength scale, which have potential applications in the surface-enhanced Raman scattering, biosensor, optical communication, solar cells, and nonlinear optical frequency mixing. In the present article, we review the Green’s matrix method for solving the surface plasmon resonances and near field in arbitrarily shaped nanostructures and in binary metallic nanostructures. Using this method, we design the plasmonic nanostructures whose resonances are tunable from the visible to near-infrared, study the interplay of plasmon resonances, and propose a new way to control plasmonic resonances in binary metallic nanostructures. 相似文献
The ability to cross host barriers is an essential virulence determinant of invasive microbial pathogens. Listeria monocytogenes is a model microorganism that crosses human intestinal and placental barriers, and causes severe maternofetal infections by an unknown mechanism. Several studies have helped to characterize the bacterial invasion proteins InlA and InlB. However, their respective species specificity has complicated investigations on their in vivo role. Here we describe two novel and complementary animal models for human listeriosis: the gerbil, a natural host for L. monocytogenes, and a knock-in mouse line ubiquitously expressing humanized E-cadherin. Using these two models, we uncover the essential and interdependent roles of InlA and InlB in fetoplacental listeriosis, and thereby decipher the molecular mechanism underlying the ability of a microbe to target and cross the placental barrier. 相似文献
Today's surface ocean is saturated with respect to calcium carbonate, but increasing atmospheric carbon dioxide concentrations are reducing ocean pH and carbonate ion concentrations, and thus the level of calcium carbonate saturation. Experimental evidence suggests that if these trends continue, key marine organisms--such as corals and some plankton--will have difficulty maintaining their external calcium carbonate skeletons. Here we use 13 models of the ocean-carbon cycle to assess calcium carbonate saturation under the IS92a 'business-as-usual' scenario for future emissions of anthropogenic carbon dioxide. In our projections, Southern Ocean surface waters will begin to become undersaturated with respect to aragonite, a metastable form of calcium carbonate, by the year 2050. By 2100, this undersaturation could extend throughout the entire Southern Ocean and into the subarctic Pacific Ocean. When live pteropods were exposed to our predicted level of undersaturation during a two-day shipboard experiment, their aragonite shells showed notable dissolution. Our findings indicate that conditions detrimental to high-latitude ecosystems could develop within decades, not centuries as suggested previously. 相似文献
Fy blood group antigens are carried by the Duffy antigen receptor for chemokines (DARC), a red cells receptor for Plasmodium vivax broadly implicated in human health and diseases. Recombinant VHHs, or nanobodies, the smallest intact antigen binding fragment derivative from the heavy chain-only antibodies present in camelids, were prepared from a dromedary immunized against DARC N-terminal extracellular domain and selected for DARC binding. A described VHH, CA52, does recognize native DARC on cells. It inhibits P. vivax invasion of erythrocytes and displaces interleukin-8 bound to DARC. The targeted epitope overlaps the well-defined DARC Fy6 epitope. KD of CA52?CDARC equilibrium is sub-nanomolar, hence ideal to develop diagnostic or therapeutic compounds. Immunocapture by immobilized CA52 yielded highly purified DARC from engineered K562 cells. This first report on a VHH with specificity for a red blood cell protein exemplifies VHHs?? potentialities to target, to purify, and to modulate the function of cellular markers. 相似文献
