The magnesium ion, Mg2+, is essential for myriad biochemical processes and remains the only major biological ion whose transport mechanisms remain unknown. The CorA family of magnesium transporters is the primary Mg2+ uptake system of most prokaryotes and a functional homologue of the eukaryotic mitochondrial magnesium transporter. Here we determine crystal structures of the full-length Thermotoga maritima CorA in an apparent closed state and its isolated cytoplasmic domain at 3.9 A and 1.85 A resolution, respectively. The transporter is a funnel-shaped homopentamer with two transmembrane helices per monomer. The channel is formed by an inner group of five helices and putatively gated by bulky hydrophobic residues. The large cytoplasmic domain forms a funnel whose wide mouth points into the cell and whose walls are formed by five long helices that are extensions of the transmembrane helices. The cytoplasmic neck of the pore is surrounded, on the outside of the funnel, by a ring of highly conserved positively charged residues. Two negatively charged helices in the cytoplasmic domain extend back towards the membrane on the outside of the funnel and abut the ring of positive charge. An apparent Mg2+ ion was bound between monomers at a conserved site in the cytoplasmic domain, suggesting a mechanism to link gating of the pore to the intracellular concentration of Mg2+. 相似文献

The human genome holds an extraordinary trove of information about human development, physiology, medicine and evolution. Here we report the results of an international collaboration to produce and make freely available a draft sequence of the human genome. Many plants use day length as an environmental cue to ensure proper timing of the switch from vegetative to reproductive growth. Day-length sensing involves an interaction between the relative length of day and night, and endogenous rhythms that are controlled by the plant circadian clock. Thus, plants with defects in circadian regulation cannot properly regulate the timing of the floral transition. Here we describe the gene EARLY FLOWERING 4 (ELF4), which is involved in photoperiod perception and circadian regulation. ELF4 promotes clock accuracy and is required for sustained rhythms in the absence of daily light/dark cycles. elf4 mutants show attenuated expression of CIRCADIAN CLOCK ASSOCIATED 1 (CCA1), a gene that is thought to function as a central oscillator component. In addition, elf4 plants transiently show output rhythms with highly variable period lengths before becoming arrhythmic. Mutations in elf4 result in early flowering in non-inductive photoperiods, which is probably caused by elevated amounts of CONSTANS (CO), a gene that promotes floral induction. 相似文献

The CD4 glycoprotein is expressed on T-helper and cytotoxic lymphocytes which are restricted to class II major histocompatibility complex (MHC) antigens on target cells. Antibody inhibition studies imply that CD4 acts to increase the avidity of effector-target cell interactions. These observations have led to the speculation that CD4 binds to a monomorphic class II antigen determinant, thereby augmenting low affinity T-cell receptor-antigen interactions. However, no direct evidence has been presented indicating that CD4 and class II molecules interact. To address this issue, we have used a vector derived from simian virus 40 (SV40) to express a complementary DNA (cDNA) encoding the human CD4 glycoprotein. When CV1 cells expressing large amounts of the CD4 protein at the cell surface are incubated with human B cells bearing MHC-encoded class II molecules, they are bound tightly to the infected monolayer, whereas mutant B cells which lack class II molecules fail to bind. Furthermore, the binding reaction is specifically inhibited by anti-class II and anti-CD4 antibodies. Thus, the CD4 protein, even in the absence of T-cell receptor-antigen interactions, can interact directly with class II antigens to function as a cell surface adhesion molecule. 相似文献

The ability to create quantum degenerate gases has led to the realization of Bose-Einstein condensation of molecules, atom-atom entanglement and the accurate measurement of the Casimir force in atom-surface interactions. With a few exceptions, the achievement of quantum degeneracy relies on evaporative cooling of magnetically trapped atoms to ultracold temperatures. Magnetic traps confine atoms whose electronic magnetic moments are aligned anti-parallel to the magnetic field. This alignment must be preserved during the collisional thermalization of the atomic cloud. Quantum degeneracy has been reached in spherically symmetric, S-state atoms (atoms with zero internal orbital angular momentum). However, collisional relaxation of the atomic magnetic moments of non-S-state atoms (non-spherical atoms with non-zero internal orbital angular momentum) is thought to proceed rapidly. Here we demonstrate magnetic trapping of non-S-state rare-earth atoms, observing a suppression of the interaction anisotropy in collisions. The atoms behave effectively like S-state atoms because their unpaired electrons are shielded by two outer filled electronic shells that are spherically symmetric. Our results are promising for the creation of quantum degenerate gases with non-S-state atoms, and may facilitate the search for time variation of fundamental constants and the development of a quantum computer with highly magnetic atoms. 相似文献

The early stages in the history of biological investigation of the Great Salt Lake involved the identification and establishment of taxonomic relationships of the indigenous flora and fauna. A result of advancements in systematic biology is that many of the earlier names of organisms have been placed in synonomy. Recent interest in the lake has centered on biological productivity and interactions of components of the ecosystem. Accurate measurement of the lifetime of the neutron (which is unstable to beta decay) is important for understanding the weak nuclear force and the creation of matter during the Big Bang. Previous measurements of the neutron lifetime have mainly been limited by certain systematic errors; however, these could in principle be avoided by performing measurements on neutrons stored in a magnetic trap. Neutral-particle and charged-particle traps are widely used for studying both composite and elementary particles, because they allow long interaction times and isolation of particles from perturbing environments. Here we report the magnetic trapping of neutrons. The trapping region is filled with superfluid 4He, which is used to load neutrons into the trap and as a scintillator to detect their decay. Neutrons in the trap have a lifetime of 750(+330)(-200) seconds, mainly limited by their beta decay rather than trap losses. Our experiment verifies theoretical predictions regarding the loading process and magnetic trapping of neutrons. Further refinement of this method should lead to improved precision in the neutron lifetime measurement. 相似文献
