An efficient culture medium producing a bacterial elastase with high yields was developed further following preliminary studies by means of response surface method. Central composite design (CCD) and response surface methodology were applied to optimize the medium constituents. A central composite design was used to explain the combined effect of three medium constituents, viz, glucose, K(2)HPO(4), MgSO(4).7H(2)O. The strain produced more elastase in the completely optimized medium, as compared with the partially optimized medium. The fitted model of the second model, as per RSM, showed that glucose was 7.4 g/100 ml, casein 1.13 g/100 ml, corn steep flour 0.616 g/100 ml, K(2)HPO(4) 0.206 g/100 ml and MgSO(4).7H(2)O 0.034 g/100 ml. The fermentation kinetics of these two culture media in the flask experiments were analyzed. It was found that the highest elastase productivity occurred at 54 hours. Higher glucose concentration had inhibitory effect on elastase production. At the same time, we observed that the glucose consumption rate was slow in the completely optimized medium, which can explain the lag period of the highest elastase production. Some metal ions and surfactant additives also affected elastase production and cell growth.  相似文献

Electroantennogram (EAG) evaluation of selected compounds from wilted leaves of black poplar, Populus nigra, showed that phenyl acetaldehyde, methyl salicylate, (E)-2-hexenal elicited strong responses from male antennae of Helicoverpa armigera. When mixed with sex pheromone (Ph), some volatiles, e.g. phenyl acetaldehyde, benzyl alcohol, phenylethanol, methylsalicylate, linalool, benzaldehyde, (Z)-3-hexenol, (Z)-3-hexenylacetate, (Z)-6-nonenol, cineole, (E)-2-hexenal, and geraniol elicited stronger responses from male antennae than Ph alone. Wind tunnel bioassay demonstrated that various volatiles could either enhance or inhibit the effect of synthetic sex pheromone. (E)-2-hexenal, (Z)-3-hexenol and linalool in combination with Ph could not induce any male to land on source at all, whereas phenyl acetaldehyde, benzaldehyde, (Z)-6-nonenol and salicylaldehyde combined with Ph enhanced male response rates by 58.63%, 50.33%, 51.85% and 127.78%, respectively, compared to Ph alone. These results suggested that some volatiles shouldmodify sex pheromone caused behavior and that some of them could possibly be used as a tool for disrupting mating or for enhancing the effect of synthetic sex pheromone in the field.  相似文献

Land degradation is one of the severe environmental problems in China. In order to combat land degradation, a soil conservation program was introduced since 2000 to reduce soil erosion by converting slope-cultivated land into forestry and pasture. This paper represents the first systematic attempt to investigate the impact of the soil conservation program on land degradation in the loess plateau. The results indicate that the soil conservation program to convert slope fields into forest or pasture is an effective way to combat soil erosion. However, a subsidy that is higher than profit of land use activity of slope fields before their conversion into forest and pasture is needed to encourage farmers to join the conservation program. A policy measure to encourage and assist farmers to develop sedentary livestock by using crops produced from fields as well as fodder and forage grass from the converted slope fields might contribute to combat soil erosion. Increase in off-farm job opportunities may encourage households to reduce cultivation in slope fields. That implies a policy measure to encourage rural urbanization might contribute to combat soil erosion.  相似文献

Through the use of reliable AMS dating of high resolution (15–30 years) peat and the establishment of monsoon climate proxies sequence, we have been able to recognize several cold, dry events in the Tibetan Plateau during the Holocene. The more obvious ones occurred around 12800, 11300, 10200, 9580, 8900, 6400, 4400, 3700, 2800 and 1500 cal. aBP. These events correlate well with both ice rafting events recorded in high latitude North Atlantic Ocean sediment cores and cooling events in the low latitude SST. Spectral analysis indicates high frequency climate variation on centennial-millennial time scale during the Holocene. This further reflects Holocene climate instability and the existence of centennial-millenium scale rhythm in mid latitude areas as well.  相似文献

In infections, microbial components provide signals that alert the immune system to danger and promote the generation of immunity. In the absence of such signals, there is often no immune response or tolerance may develop. This has led to the concept that the immune system responds only to antigens perceived to be associated with a dangerous situation such as infection. Danger signals are thought to act by stimulating dendritic cells to mature so that they can present foreign antigens and stimulate T lymphocytes. Dying mammalian cells have also been found to release danger signals of unknown identity. Here we show that uric acid is a principal endogenous danger signal released from injured cells. Uric acid stimulates dendritic cell maturation and, when co-injected with antigen in vivo, significantly enhances the generation of responses from CD8+ T cells. Eliminating uric acid in vivo inhibits the immune response to antigens associated with injured cells, but not to antigens presented by activated dendritic cells. Our findings provide a molecular link between cell injury and immunity and have important implications for vaccines, autoimmunity and inflammation.  相似文献

Inclusion complex of Orange II with β-Cyclodextrin (β-CD) and the anti-photolysis effect under UV-light were investigated. The molar ratio of inclusion complex of β-Cyclodextrin and Orange II is 1∶1. The formation constantK = 1.236×103 L/mol was determined by the UV and Fluorescence spectra respectively, which was quite in accordance with the calculation with a modified Benesi-Hildbrand equation. The inclusion complex was characterized by the IR spectra and the molar ratio of inclusion complex is 1∶1 too. The formation constantK = 1.266×103 L/mol was determined by1H NMR analysis and was nearly the same by UV and fluorescence spectra. The photocatalytic decolorization rate of Orange II solutions containing β-CD and TiO2 was smaller by 51.9% than that of the Orange II solutions only containing TiO2, while in the case of direct photolysis of Orange II solutions, β-CD can lower the photolysis rate by 48.1% under UV-light. This result indicates β-CD can inhibit the photolysis and photocatalytic decolorization of Orange II under UV-light. The β-CD inclusion complex was found to be persistent to UV-light photolysis. 同位素纯硅片具有特殊的物理性质,在信息技术领域有着非常广阔的应用前景。为获得高丰度28Si,利用国产的高速气体离心机,选择SiF4作为分离的工作介质,对硅同位素进行了离心分离实验。通过单机分离实验,调整气体离心机的有关参数,摸索出适合级联分离的单机参数,同时得到气体离心机分离硅同位素的单位质量数的全分离系数γ0。利用由4台离心机组成的级联,经过6个阶段的分离,得到了28Si丰度达到99.5%的样品。实验结果表明,离心法分离硅同位素是完全可行的。  相似文献
