Voltage-gated potassium channels such as Shaker help to control electrical signalling in neurons by regulating the passage of K+ across cell membranes. Ion flow is controlled by a voltage-dependent gate at the intracellular side of the pore, formed by the crossing of four alpha-helices--the inner-pore helices. The prevailing model of gating is based on a comparison of the crystal structures of two bacterial channels--KcsA in a closed state and MthK in an open state--and proposes a hinge motion at a conserved glycine that splays the inner-pore helices wide open. We show here that two types of intersubunit metal bridge, involving cysteines placed near the bundle crossing, can occur simultaneously in the open state. These bridges provide constraints on the open Shaker channel structure, and on the degree of movement upon opening. We conclude that, unlike predictions from the structure of MthK, the inner-pore helices of Shaker probably maintain the KcsA-like bundle-crossing motif in the open state, with a bend in this region at the conserved proline motif (Pro-X-Pro) not found in the bacterial channels. A narrower opening of the bundle crossing in Shaker K+ channels may help to explain why Shaker has an approximately tenfold lower conductance than its bacterial relatives.  相似文献

Basidiomes of polypore fungi host many insects. Yet systematic information about insect assemblages from most fungal species is lacking. We studied the insect community associated with the wood-decaying fungus Inonotus radiatus (Sowerby: Fr.) P. Karst. (Hymenochaetales). More specifically, we studied the effect of successional stage and weight of basidiomes, as well as shore exposition (north or south), on species richness and composition, as well as occurrence and abundance of the most abundant fungivores. Basidiomes were collected from riparian forests at five lakes in Sweden. Insects were reared out from the basidiomes in the laboratory. A total of 5645 adult insect individuals of 117 taxa were obtained. Among these, 2782 specimens of 36 taxa use Inonotus radiatus basidiomes as breeding habitat. Eight species of parasitic wasps were new to Sweden. The most abundant fungivore was Ennearthron cornutum (Ciidae), which is a generalist breeding in many polypore species. Based on our material and literature, the melandryid beetles Abdera affinis and A. flexuosa seem to be specialists on the order Hymenochaetales. Other frequent fungivores were Dorcatoma dresdensis (Ptinidae), and the lepidopterans Archinemapogon yildizae, Nemapogon cloacellus and N. picarellus (Tineidae). The parasitoid community associated with the tineid moths was similar to the community associated with the other polypore species in the order Polyporales. In contrast, the parasitoids associated with Dorcatoma (and/or Abdera) were different from the other Polyporales species, suggesting that the fungal host species is more important for these parasitoid species than the beetle host species itself. The most abundant and frequent parasitoids were the braconids Diospilus dispar and Colastes fritzeni, which both parasitise Dorcatoma. Species richness was significantly smaller in fresh than in more decayed basidiomes, but species composition did not differ. There was no difference in species composition or richness between north and south shorelines.  相似文献

In this paper the astronomical explorations of the southern celestial sky by Dutch navigators at the end of the sixteenth century are investigated. It is shown that the main motivation for this scientific enterprise stemmed from Dutch cartographic tradition and interests, represented first and foremost by Petrus Plancius and the competing globemakers Hondius and Blaeu. It is shown, too, that at the time actually two surveys were carried out. We have investigated the results of the two surveys by analysing the data presented on celestial globes by Hondius and Blaeu, and in the star catalogue published by De Houtman in 1603. It appears that only after the first survey was it recognized that the stars of the well-known Southern Cross constellation could be matched with stars already recorded by Ptolemy.  相似文献

The interplay between discrete vibrational and electronic degrees of freedom directly influences the chemical and physical properties of molecular systems. This coupling is typically studied through optical methods such as fluorescence, absorption and Raman spectroscopy. Molecular electronic devices provide new opportunities for exploring vibration-electronic interactions at the single molecule level. For example, electrons injected from a scanning tunnelling microscope tip into a metal can excite vibrational excitations of a molecule situated in the gap between tip and metal. Here we show how current directly injected into a freely suspended individual single-wall carbon nanotube can be used to excite, detect and control a specific vibrational mode of the molecule. Electrons tunnelling inelastically into the nanotube cause a non-equilibrium occupation of the radial breathing mode, leading to both stimulated emission and absorption of phonons by successive electron tunnelling events. We exploit this effect to measure a phonon lifetime of the order of 10 ns, corresponding to a quality factor of well over 10,000 for this nanomechanical oscillator.  相似文献

Mitochondrial preproteins are imported by a multisubunit translocase of the outer membrane (TOM), including receptor proteins and a general import pore. The central receptor Tom22 binds preproteins through both its cytosolic domain and its intermembrane space domain and is stably associated with the channel protein Tom40 (refs 11-13). Here we report the unexpected observation that a yeast strain can survive without Tom22, although it is strongly reduced in growth and the import of mitochondrial proteins. Tom22 is a multifunctional protein that is required for the higher-level organization of the TOM machinery. In the absence of Tom22, the translocase dissociates into core complexes, representing the basic import units, but lacks a tight control of channel gating. The single membrane anchor of Tom22 is required for a stable interaction between the core complexes, whereas its cytosolic domain serves as docking point for the peripheral receptors Tom20 and Tom70. Thus a preprotein translocase can combine receptor functions with distinct organizing roles in a multidomain protein.  相似文献
