A note on the use of protease inhibitors during chromatin fractionation on hydroxyapatite columns

Summary It was found that NaHSO₃ present in the eluents enabled full and reproducible recovery of chromosomal proteins from a hydroxyapatite column. Another protease inhibitor PMSF, did not have that effect.     

Antibody-induced formation of caps inToxoplasma gondii

Summary Trophozoites ofToxoplasma gondii from mouse peritoneal exudate move their surface membrane antigens towards one pole of the cell when incubated with antibodies. The phenomenon may be induced in up to 50% of incubated parasites. It is prvented by some metabolic inhibitors and low temperatures (0–4°C). These properties do not change in parasites subpassaged after repeated incubation with antibodies.We wish to thank Dr. J.lusarczyk of the National Institute of Hygiene for his help in preparation the photomicropgraphs.     

Glutaminase activity in rat skeletal muscle mitochondria

Summary Isolated rat skeletal muscle mitochondria took up about 40-ng-atoms O per min per mg protein, with glutamine as the only respiratory substrate. The mitochondria incubated in the presence of glutamine and KCN formed both ammonia and glutamate in equivalent amounts. The experiments reported here provide suggestive evidence that rat skeletal muscle mitochondria contain glutaminase (L-glutamine amidohydrolase EC 3.5.1.2.) activity.This work was supported by the Polish Academy of Sciences within the project II. 1, 2, 6.     

The reproductive strategies of clausiliid land snails from Northern Vietnam (Gastropoda: Stylommatophora)

Under laboratory conditions we scrutinized the reproduction and development of two clausiliid land snail species from northern Vietnam: Phaedusa paviei (Morlet, 1892) and Oospira vanbuensis (Bavay and Dautzenberg, 1899). These species, very similar in shell size and shape, feature different reproductive modes. The former is viviparous, giving birth to one or two neonates on a single occasion, with the number of developing embryos retained in the reproductive tract ranging from 1 to 11. The embryonic shells are 1.8–4.7 mm in height. The latter species is oviparous and produces eggs in clutches (four to six per clutch). Hatching occurs 18–21 days after egg-laying. Both species exhibit a similar growth pattern: the ultimate shell size is attained after 6.5–8.5 months and reproduction starts 5–7 months later. The distribution of viviparous reproduction among the Phaedusinae and its taxonomic importance are discussed.     

Lung cancer susceptibility locus at 5p15.33

We carried out a genome-wide association study of lung cancer (3,259 cases and 4,159 controls), followed by replication in 2,899 cases and 5,573 controls. Two uncorrelated disease markers at 5p15.33, rs402710 and rs2736100 were detected by the genome-wide data (P = 2 x 10(-7) and P = 4 x 10(-6)) and replicated by the independent study series (P = 7 x 10(-5) and P = 0.016). The susceptibility region contains two genes, TERT and CLPTM1L, suggesting that one or both may have a role in lung cancer etiology.     

KLHL3 mutations cause familial hyperkalemic hypertension by impairing ion transport in the distal nephron

Familial hyperkalemic hypertension (FHHt) is a Mendelian form of arterial hypertension that is partially explained by mutations in WNK1 and WNK4 that lead to increased activity of the Na(+)-Cl(-) cotransporter (NCC) in the distal nephron. Using combined linkage analysis and whole-exome sequencing in two families, we identified KLHL3 as a third gene responsible for FHHt. Direct sequencing of 43 other affected individuals revealed 11 additional missense mutations that were associated with heterogeneous phenotypes and diverse modes of inheritance. Polymorphisms at KLHL3 were not associated with blood pressure. The KLHL3 protein belongs to the BTB-BACK-kelch family of actin-binding proteins that recruit substrates for Cullin3-based ubiquitin ligase complexes. KLHL3 is coexpressed with NCC and downregulates NCC expression at the cell surface. Our study establishes a role for KLHL3 as a new member of the complex signaling pathway regulating ion homeostasis in the distal nephron and indirectly blood pressure.

Résumé Nous avons trouvé que le facteur Hageman (factor XII), contenu dans le plasma humain citraté, pauvre en plaquettes, est adsorbé presque sélectivement par le collagène. Il est également possible d'éluer ce facteur au pH alcalin.
