Conformation of ribonuclease S-protein

Summary Ribonuclease S-protein exhibits a pH-dependent conformational transition between folded and unfolded states, and some unfolded S-protein persists up to pH 8. The histidine C2 proton resonance of the unfolded species was erroneously assigned by Bradbury et al. to histidine residue 119 of the folded species.     

Naturally occurring hemagglutinins in the hemolymph of the scorpionParuroctonus mesaensis Stahnke

Summary Lectins which bind sialic acid-containing receptors are present in the hemolymph of the scorpionParuroctonus mesaensis Stahnke. Glycoproteins like bovine submaxillary mucin, fetuin and human orosomucoid behave as strong inhibitors forParuroctonus lectins; desialylation of glycoproteins results in a drastic reduction of their inhibitory capabilities confirming that sialic acids are the terminal monosaccharides ofParuroctonus lectin receptors.We wish to thank Dr H. Exploring Community Collaborations: Social Network Analysis as a Reflective Tool for Public Health

Social network analysis is a potentially useful reflective tool that could be used by public health practitioners to assess the overall composition of their networks, strengthen collaborations with other community partners, and evaluate network function. The purpose of this paper is to report on public health practitioners’ experiences with social network analysis. Thirteen public health practitioners, each associated with their own advocacy-oriented community collaboration, participated in the study. Specifically, the intervention consisted of social network analysis output (social network analysis measures and sociograms) and a facilitated discussion to stimulate reflection on the respondents’ professional networks. Semi-structured interviews were conducted two weeks later to solicit participants’ reactions to the sociograms and the accompanying measures, and elicit thoughts about how social network analysis might have an impact on their work. Participants commented on ways in which social network analysis could be applied in practice, such as using sociograms to raise awareness of the nature of current networks, as a planning and evaluation tool, to identify gaps, and to assess the degree of sustainability of current networks.

The deoxyguanosine kinase gene is mutated in individuals with depleted hepatocerebral mitochondrial DNA.

Mitochondrial DNA (mtDNA)-depletion syndromes (MDS; OMIM 251880) are phenotypically heterogeneous, autosomal-recessive disorders characterized by tissue-specific reduction in mtDNA copy number. Affected individuals with the hepatocerebral form of MDS have early progressive liver failure and neurological abnormalities, hypoglycemia and increased lactate in body fluids. Affected tissues show both decreased activity of the mtDNA-encoded respiratory chain complexes (I, III, IV, V) and mtDNA depletion. We used homozygosity mapping in three kindreds of Druze origin to map the gene causing hepatocerebral MDS to a region of 6.1 cM on chromosome 2p13, between markers D2S291 and D2S2116. This interval encompasses the gene (DGUOK) encoding the mitochondrial deoxyguanosine kinase (dGK). We identified a single-nucleotide deletion (204delA) within the coding region of DGUOK that segregates with the disease in the three kindreds studied. Western-blot analysis did not detect dGK protein in the liver of affected individuals. The main supply of deoxyribonucleotides (dNTPs) for mtDNA synthesis comes from the salvage pathway initiated by dGK and thymidine kinase-2 (TK2). The association of mtDNA depletion with mutated DGUOK suggests that the salvage-pathway enzymes are involved in the maintenance of balanced mitochondrial dNTP pools.     

Elasticity of iron at the temperature of the Earth's inner core.

Seismological body-wave and free-oscillation studies of the Earth's solid inner core have revealed that compressional waves traverse the inner core faster along near-polar paths than in the equatorial plane. Studies have also documented local deviations from this first-order pattern of anisotropy on length scales ranging from 1 to 1,000 km (refs 3, 4). These observations, together with reports of the differential rotation of the inner core, have generated considerable interest in the physical state and dynamics of the inner core, and in the structure and elasticity of its main constituent, iron, at appropriate conditions of pressure and temperature. Here we report first-principles calculations of the structure and elasticity of dense hexagonal close-packed (h.c.p.) iron at high temperatures. We find that the axial ratio c/a of h.c.p. iron increases substantially with increasing temperature, reaching a value of nearly 1.7 at a temperature of 5,700 K, where aggregate bulk and shear moduli match those of the inner core. As a consequence of the increasing c/a ratio, we have found that the single-crystal longitudinal anisotropy of h.c.p. iron at high temperature has the opposite sense from that at low temperature. By combining our results with a simple model of polycrystalline texture in the inner core, in which basal planes are partially aligned with the rotation axis, we can account for seismological observations of inner-core anisotropy.
