PTC124 targets genetic disorders caused by nonsense mutations

Nonsense mutations promote premature translational termination and cause anywhere from 5-70% of the individual cases of most inherited diseases. Studies on nonsense-mediated cystic fibrosis have indicated that boosting specific protein synthesis from <1% to as little as 5% of normal levels may greatly reduce the severity or eliminate the principal manifestations of disease. To address the need for a drug capable of suppressing premature termination, we identified PTC124-a new chemical entity that selectively induces ribosomal readthrough of premature but not normal termination codons. PTC124 activity, optimized using nonsense-containing reporters, promoted dystrophin production in primary muscle cells from humans and mdx mice expressing dystrophin nonsense alleles, and rescued striated muscle function in mdx mice within 2-8 weeks of drug exposure. PTC124 was well tolerated in animals at plasma exposures substantially in excess of those required for nonsense suppression. The selectivity of PTC124 for premature termination codons, its well characterized activity profile, oral bioavailability and pharmacological properties indicate that this drug may have broad clinical potential for the treatment of a large group of genetic disorders with limited or no therapeutic options.     

河北省御道口牧场气候生产潜力估算

御道口牧场位于河北省围场县北部坝上地区,是河北省畜牧业生产的重要产区.根据牧场多年降水和气温等气象观测资料,同时考虑地形因素和最小面积的限制,对御道口牧场的气候生产潜力进行了估算.得出牧场的气候生产潜力在3.75～5 t·hm-2·a-1之间,牧场内适合发展人工草地的平地和缓坡(＜5°)约占总土地面积的76.5%.     

高校化学实验室通风系统研究

针对高校化学实验室的特点,提出化学实验室通风系统、自动控制系统、BENTAX净化系统设计方案。通过对设计方案的研究,为类似工程实践提供可参考的思路和方法。     

长大公路隧道火灾安全疏散性能化设计与分析

基于性能化防火设计方法, 分析火灾条件下长大公路隧道内人 员疏散行为的特点,给出隧道内人 员疏散的安全判据,并结合某隧道 的结构及车流特征对人员疏散过程 进行了数值模拟研究,分析隧道在 正常通车和局部行车堵塞时,不同 火源位置、横通道间距与横通道宽 度对人员疏散时间及安全状态的影 响。结果表明,与建筑内人员疏散不 同,长大公路隧道内火灾的可用安 全疏散时间(ASET)与需要的安全疏 散时间(RSET)均为沿隧道长度方向 位置的函数。隧道内人员安全疏散 的时间曲线在各横通道疏散口出现 峰值,这主要是受到横通道通行能 力的限制,各疏散口人员集聚等待 所致。当隧道发生局部堵塞时,横 通道口需要的安全疏散时间显著增 加,危险性也急剧增大。隧道内人 员疏散的性能化防火设计应综合考 虑火源位置对人员载荷及疏散口位 置可用安全疏散时间的影响。该研 究为公路隧道火灾安全疏散设施的 优化设计提供依据。

Essential Independent Condition for Graphs to Be Hamiltonian

Let G be a graph of order n．For graph to be Hamiltonian,beginning with Dirac’s classic result in １９５２,Dirac’s theorem was followed by that of Ore in１９６０．In１９８４,Fan generalized Dirac’s theorem and Ore’s theorem as if G is a２-connected graph of order n and max｛d（u）,d（v）｝≥n/２for each pair of vertices u and v with d（u,v）＝２,then G is hamiltonian．In１９９１,Faudree et al proved that if G is a２-connected graph and,| N（u）∪N（v）|＋δ（G）≥n for each pair of nonadjacent vertices u,v∈V（G）,then G is hamiltonian．This paper generalizes the above onditions of Dirac,Ore,Fan and Faudree et al in the case of３-connected graph and proves that if G is a３-connected graph of order n and max｛｜N（x）∪ N（y）｜＋d（u）,｜N（w）∪N（z）｜＋d（v）｝≥n for every choice of６Essential independent vertices,then G is hamiltonian．     

大型公共项目融资风险与控制研究

在大型公共项目的运作过程中,融资已经成为解决资金缺口的主要手段。笔者在界定项目融资结构、项目资金流量及其内涵的基础上,通过建立定量模型,从融资比例、建设工期和项目收益率三个方面对大型公共项目运作中的融资风险进行了分析,并通过算例对该模型进行了验证。     

实时定量PCR检测B细胞恶性肿瘤中BCL11A基因的表达水平

目的:建立B细胞淋巴瘤/白血病BCL11A基因的定量检测方法,并分析其在B细胞恶性肿瘤中的表达水平。方法:利用实时定量RT-PCR分析B细胞淋巴瘤(18例)、B细胞性慢性淋巴细胞白血病(B-CLL,8例)、T细胞性急性淋巴细胞白血病(T-ALL,8例)和正常对照(15例)外周血单个核细胞(PBMC)中BCL11A基因的表达水平,以甘油醛-3-磷酸脱氢酶(GAPDH)基因作为内对照。结果:B-CLL组和B细胞淋巴瘤组患者PBMC中BCL11A表达水平均明显高于正常对照(P=0.000)和T-ALL组(P=0.000);T-ALL组和正常对照组BCL11A表达水平无显著差别(P=0.084);B-CLL组和B细胞淋巴瘤组BCL11A表达水平无显著差别(P=0.776)。在B细胞淋巴瘤不同的病例中,BCL11A表达水平有较大差异,其中最小值为0.04,最大值为9.70,中位值为1.00。结论:成功建立BCL11A基因的定量检测方法。
