Uric acid is the end product of purine metabolism in humans and great apes, which have lost hepatic uricase activity, leading to uniquely high serum uric acid concentrations (200-500 microM) compared with other mammals (3-120 microM). About 70% of daily urate disposal occurs via the kidneys, and in 5-25% of the human population, impaired renal excretion leads to hyperuricemia. About 10% of people with hyperuricemia develop gout, an inflammatory arthritis that results from deposition of monosodium urate crystals in the joint. We have identified genetic variants within a transporter gene, SLC2A9, that explain 1.7-5.3% of the variance in serum uric acid concentrations, following a genome-wide association scan in a Croatian population sample. SLC2A9 variants were also associated with low fractional excretion of uric acid and/or gout in UK, Croatian and German population samples. SLC2A9 is a known fructose transporter, and we now show that it has strong uric acid transport activity in Xenopus laevis oocytes. 相似文献

Many video surveillance applications rely on efficient motion detection. However, the algorithms are usually costly since they compute a background model at every pixel of the frame. This paper shows that, in the case of a planar scene with a fixed calibrated camera, a set of pixels can be selected to compute the background model while ignoring the other pixels for accurate but less costly motion detection. The calibration is used to first define a volume of interest in the real world and to project the volume of interest onto the image, and to define a spatial adaptive subsampling of this region of interest with a subsampling density that depends on the camera distance. Indeed, farther objects need to be analyzed with more precision than closer objects. Tests on many video sequences have integrated this adaptive subsampling to various motion detection techniques. 相似文献

Currently, the shortest laser pulses that can be generated in the visible spectrum consist of fewer than two optical cycles (measured at the full-width at half-maximum of the pulse's envelope). The time variation of the electric field in such a pulse depends on the phase of the carrier frequency with respect to the envelope-the absolute phase. Because intense laser-matter interactions generally depend on the electric field of the pulse, the absolute phase is important for a number of nonlinear processes. But clear evidence of absolute-phase effects has yet to be detected experimentally, largely because of the difficulty of stabilizing the absolute phase in powerful laser pulses. Here we use a technique that does not require phase stabilization to demonstrate experimentally the influence of the absolute phase of a short laser pulse on the emission of photoelectrons. Atoms are ionized by a short laser pulse, and the photoelectrons are recorded with two opposing detectors in a plane perpendicular to the laser beam. We detect an anticorrelation in the shot-to-shot analysis of the electron yield. 相似文献

Conventional high-temperature reactions limit the control of coordination polyhedra in transition-metal oxides to those obtainable within the bounds of known coordination geometries for a given transition metal. For example, iron atoms are almost exclusively coordinated by three-dimensional polyhedra such as tetrahedra and octahedra. However, recent works have shown that binary metal hydrides act as reducing agents at low temperatures, allowing access to unprecedented structures. Here we show the reaction of a perovskite SrFeO3 with CaH2 to yield SrFeO2, a new compound bearing a square-planar oxygen coordination around Fe2+. SrFeO2 is isostructural with 'infinite layer' cupric oxides, and exhibits a magnetic order far above room temperature in spite of the two-dimensional structure, indicating strong in-layer magnetic interactions due to strong Fe d to O p hybridization. Surprisingly, SrFeO2 remains free from the structural instability that might well be expected at low temperatures owing to twofold orbital degeneracy in the Fe2+ ground state with D(4h) point symmetry. The reduction and the oxidation between SrFeO2 and SrFeO3 proceed via the brownmillerite-type intermediate SrFeO2.5, and start at the relatively low temperature of approximately 400 K, making the material appealing for a variety of applications, including oxygen ion conduction, oxygen gas absorption and catalysis. 相似文献

Human cytomegalovirus (HCMV) is a major pathogen in immunocompromised individuals. Here, non-toxic concentrations of the anti-cancer
kinase inhibitor sorafenib were shown to inhibit replication of different HCMV strains (including a ganciclovir-resistant
strain) in different cell types. In contrast to established anti-HCMV drugs, sorafenib inhibited HCMV major immediate early
promoter activity and HCMV immediate early antigen (IEA) expression. Sorafenib is known to inhibit Raf. Comparison of sorafenib
with the MEK inhibitor U0126 suggested that sorafenib inhibits HCMV IEA expression through inhibition of Raf but independently
of signaling through the Raf downstream kinase MEK 1/2. In concordance, siRNA-mediated depletion of Raf but not of MEK-reduced
IEA expression. In conclusion, sorafenib diminished HCMV replication in clinically relevant concentrations and inhibited HCMV
IEA expression, a pathophysiologically relevant event that is not affected by established anti-HCMV drugs. Moreover, we demonstrated

We identified association of restless legs syndrome (RLS) with PTPRD at 9p23-24 in 2,458 affected individuals and 4,749 controls from Germany, Austria, Czechia and Canada. Two independent SNPs in the 5' UTR of splice variants expressed predominantly in the central nervous system showed highly significant P values (rs4626664, P(nominal/lambda corrected) = 5.91 x 10(-10), odds ratio (OR) = 1.44; rs1975197, P(nominal/lambda corrected) = 5.81 x 10(-9), OR = 1.31). This work identifies PTPRD as the fourth genome-wide significant locus for RLS. 相似文献
