In order to maximize their use of light energy in photosynthesis, plants have molecules that act as light-harvesting antennae, which collect light quanta and deliver them to the reaction centres, where energy conversion into a chemical form takes place. The functioning of the antenna responds to the extreme changes in the intensity of sunlight encountered in nature. In shade, light is efficiently harvested in photosynthesis. However, in full sunlight, much of the energy absorbed is not needed and there are vitally important switches to specific antenna states, which safely dissipate the excess energy as heat. This is essential for plant survival, because it provides protection against the potential photo-damage of the photosynthetic membrane. But whereas the features that establish high photosynthetic efficiency have been highlighted, almost nothing is known about the molecular nature of the dissipative states. Recently, the atomic structure of the major plant light-harvesting antenna protein, LHCII, has been determined by X-ray crystallography. Here we demonstrate that this is the structure of a dissipative state of LHCII. We present a spectroscopic analysis of this crystal form, and identify the specific changes in configuration of its pigment population that give LHCII the intrinsic capability to regulate energy flow. This provides a molecular basis for understanding the control of photosynthetic light-harvesting.  相似文献

柴达木盆地三湖地区第四系浅层生物气一直被认为完全是由第四系腐殖型有机质生成的.最新研究结果表明:(1)第四系生物气藏探明储量与源岩条件之间不匹配;(2)高分辨率地震剖面特征表明三湖坳陷气田区域第四系发育了大量的气烟囱;(3)生物气组分特征表明有热成因气混入的可能性;(4)乙烷等重烃气碳同位素特征表明三湖坳陷第四系气藏有腐泥型生物气混入;(5)三湖坳陷上第三系狮子沟组发育偏腐泥型有机质.笔者认为第四系浅层生物气藏的形成机理比较复杂,包括“下生上储”和“自生自储”两种模式,第四系的浅生物气由上第三系狮子沟组生物气与第四系生物气混合而成.  相似文献

Many important agronomic traits in crop plants, including stress tolerance, are complex traits controlled by quantitative trait loci (QTLs). Isolation of these QTLs holds great promise to improve world agriculture but is a challenging task. We previously mapped a rice QTL, SKC1, that maintained K(+) homeostasis in the salt-tolerant variety under salt stress, consistent with the earlier finding that K(+) homeostasis is important in salt tolerance. To understand the molecular basis of this QTL, we isolated the SKC1 gene by map-based cloning and found that it encoded a member of HKT-type transporters. SKC1 is preferentially expressed in the parenchyma cells surrounding the xylem vessels. Voltage-clamp analysis showed that SKC1 protein functions as a Na(+)-selective transporter. Physiological analysis suggested that SKC1 is involved in regulating K(+)/Na(+) homeostasis under salt stress, providing a potential tool for improving salt tolerance in crops.  相似文献

Apoptosis is executed by caspases, which undergo proteolytic activation in response to cell death stimuli. The apoptotic protease-activating factor 1 (Apaf-1) controls caspase activation downstream of mitochondria. During apoptosis, Apaf-1 binds to cytochrome c and in the presence of ATP/dATP forms an apoptosome, leading to the recruitment and activation of the initiator caspase, caspase-9 (ref. 2). The mechanisms underlying Apaf-1 function are largely unknown. Here we report the 2.2-A crystal structure of an ADP-bound, WD40-deleted Apaf-1, which reveals the molecular mechanism by which Apaf-1 exists in an inactive state before ATP binding. The amino-terminal caspase recruitment domain packs against a three-layered alpha/beta fold, a short helical motif and a winged-helix domain, resulting in the burial of the caspase-9-binding interface. The deeply buried ADP molecule serves as an organizing centre to strengthen interactions between these four adjoining domains, thus locking Apaf-1 in an inactive conformation. Apaf-1 binds to and hydrolyses ATP/dATP and their analogues. The binding and hydrolysis of nucleotides seem to drive conformational changes that are essential for the formation of the apoptosome and the activation of caspase-9.  相似文献

This paper discusses deep crustal architecture of the Baiyun Sag of the Pearl River Mouth Basin, northern South China Sea based on velocity analysis, time-depth conversion and seismic interpretation of the deep seismic reflection profile DSRP-2002. The profile was acquired and processed to 14 S TWT by the China National Offshore Oil Corp. (CNOOC) in 2002. It extends across the Baiyun Sag of the Pearl River Mouth Basin, from the northern continental shelf of the SCS to the deepwater province. As the first deep seismic reflection profile in the Pearl River Mouth Basin, this profile reveals seismic phases from basement down to upper most mantle. The Moho surface appears in the profile as an undulating layer of varying thickness of 1-3 km. It is not a single reflector interface, but a velocity gradient or interconversion layer. The crust thins stepwisely from the shelf to the continental slope and the abyssal plain (from north to south), and also thins under depocenters. The crustal thickness is only 7 km in the depocenter of the main Baiyun Sag, which corresponds to a Moho upwelling mirroring the basement topography. In the lower slope and the ocean-continental transition zone of the southernmost portion of the profile, three sub-parallel, NW-dipping strong reflectors found at depths around 10--21 km are interpreted as indications of a subducted Mesozoic oceanic crust. Crustal faults exist in the northern and southern boundaries of the Baiyun Sag. The intense and persistent subsidence of the Baiyun Sag might be related to the long-term activity of the crustal faults.  相似文献

The expression, purification and spectra characterization of recombinant human neuroglobin (NGB) are reported. The pET3a plasmid with the gene of NGB was transformed to E. coli BL21 (DE3) plys cells and expressed in TB culture medium. The results indicated that the expression amount of NGB is about 10 percent of the total protein in cells. The NGB protein was purified by ammonium sulfate precipitation, DEAE-Sepharose anion exchange column, Hiload 16/60 superdex 75 size exclusion chromatography and a Hiprep 16/10 Q FF anion exchange column, and a red soluble protein was obtained which showed a single band in electrophoresis. Electrospray ionization mass spectrometry (ESI-MS) showed that its molecular weight is 16930.0 Da. UV-spectra indicated that the reduced NGB has a strong absorption peak at 425 nm, and two weak peaks at 531 and 559 nm, which can be assigned to γ, β and α bands of porphyrin, respectively, and the oxidized NGB has a strong absorption peak at 413 nm which corresponds to the transition of π electrons in the porphyrin ring. The fluorescence maximal excitation wavelength is at 281 nm and its maximal emission wavelength is at 338 nm. CD spectra indicated that its secondary structure is a typical α helix, and has a positive peak at 410 nm induced by heme, The NGB protein is stable when the pH is higher than 4.  相似文献
