CpG island methylator phenotype underlies sporadic microsatellite instability and is tightly associated with BRAF mutation in colorectal cancer

Aberrant DNA methylation of CpG islands has been widely observed in human colorectal tumors and is associated with gene silencing when it occurs in promoter areas. A subset of colorectal tumors has an exceptionally high frequency of methylation of some CpG islands, leading to the suggestion of a distinct trait referred to as 'CpG island methylator phenotype', or 'CIMP'. However, the existence of CIMP has been challenged. To resolve this continuing controversy, we conducted a systematic, stepwise screen of 195 CpG island methylation markers using MethyLight technology, involving 295 primary human colorectal tumors and 16,785 separate quantitative analyses. We found that CIMP-positive (CIMP+) tumors convincingly represent a distinct subset, encompassing almost all cases of tumors with BRAF mutation (odds ratio = 203). Sporadic cases of mismatch repair deficiency occur almost exclusively as a consequence of CIMP-associated methylation of MLH1 . We propose a robust new marker panel to classify CIMP+ tumors.     

Structural basis for iron piracy by pathogenic Neisseria

Neisseria are obligate human pathogens causing bacterial meningitis, septicaemia and gonorrhoea. Neisseria require iron for survival and can extract it directly from human transferrin for transport across the outer membrane. The transport system consists of TbpA, an integral outer membrane protein, and TbpB, a co-receptor attached to the cell surface; both proteins are potentially important vaccine and therapeutic targets. Two key questions driving Neisseria research are how human transferrin is specifically targeted, and how the bacteria liberate iron from transferrin at neutral pH. To address these questions, we solved crystal structures of the TbpA-transferrin complex and of the corresponding co-receptor TbpB. We characterized the TbpB-transferrin complex by small-angle X-ray scattering and the TbpA-TbpB-transferrin complex by electron microscopy. Our studies provide a rational basis for the specificity of TbpA for human transferrin, show how TbpA promotes iron release from transferrin, and elucidate how TbpB facilitates this process.

Allelopathic effects of bur buttercup tissue on germination and growth of various grasses and forbs in vitro and in soil

The allelopathic effects of bur buttercup ( Ranunculus testiculatus ) tissue on selected grasses and forbs varied according to the substratum for germination and growth. The in vitro effects of an aqueous extract of buttercup tissue on germination and root development of five grasses were strongly inhibitory in all cases.   However, in soil the effects of buttercup tissue on germination and growth of seven grasses and two dicotyledonous herbs were small to nonsignificant. Deleterious effects were less severe in fine - as opposed to coarse-textured soils. Under field conditions, the ability of seedlings of the grasses to compete with buttercup varied with the species.     

Equilibrium: the intracellular distribution of steroid receptors

Data are presented which suggest that there are unbound receptors for oestrogen in nuclei of the smooth muscle cells of the myometrium. A new model for the distribution of unbound receptors is proposed in which unbound receptor is in equilibrium, partitioned between nucleus and cytoplasm according to the free water content of these intracellular compartments.
