Specificity in the hydrolysis of N-acyl-L-phenylalanine 4-nitroanilides by chymotrypsin

Summary The affinity of N-acyl-L-phenylalanine 4-nitroanilides for chymotrypsin is enhanced as the hydrophobicity of non-amino acid residues in the P₂-position of the substrates increases, whereas k_cat remains nearly constant. On the other hand, if alanine or leucine is in the P₂-position k_cat increases with decreasing K_M.     

Paracelsin; characterization by NMR spectroscopy and circular dichroism,and hemolytic properties of a peptaibol antibiotic from the cellulolytically active mnoldTrichoderma reesei. Part B

Summary Paracelsin, a hemolytic and membrane active polypeptide antibiotic of the peptaibol class which is excreted by the moldTrichderma reesei, was obtained by a simplified and isolation procedure utilziing hydrophobic adsorber resin. Investigation by¹³C nuclear magnetic resonance spectroscopy and circular dichroism revealed considerable helical portions in solution, and the very recently accomplished sequence determination of paracelsin allows the discussion of the results with regard to the closely related analogues, alamethicin and suzukacillin. A selective cleavage of the peptide was achieved by careful treatment with various acids, and a buffer of pH 8.25 and of high ionic strength made possible the quantitative determination of the C-terminal phenylalaninol released by means of ion-exchange chromatography. The significance of the production of paracelsin and related mycotoxins of the peptaibol class, exhibiting various kinds of biological activity, is discussed with respect to the extensive effort being made towards biotechnological applications of species, strains and cellulolytically highly active mutants of the fungusTrichoderma.Presented in part at the 5th European Symposium on Animal, Plant and Microbial Toxins, Hannover, August 29–September 2, 1983 and a lecture given at Ciby-Geigy/Basel in March 1984.Acknowledgment. Model of the interaction between trypsin and alpha 2-macroglobulin of porcine serum]

A comparative study of the dissociation into subunits of Porcine alpha2 M, either native or bound to trypsin (Tn), has been carried out in order to determine the modifications of the alpha2 M structure due to the formation of the Tn-alpha2 M complex. Analytical ultra-centrifugation at pH 3.5 shows that the dissociation is smaller when alpha 2 M is bound to trypsin. Electrophoresis in 4% polyacrylamide gels, in presence of 0.1% SDS, of alpha2 M and Tn-alpha2 M incubated in 1% SDS leads to the same conclusion; the enzyme must stabilize the quaternay structure of alpha2 M. In presence of SDS + beta-mercaptoethanol, only a molecular weight (M.W.) 200,000 band is revealed in electrophoresis pattern of native alpha2 M. In the case of reduced Tn-alpha2 M, some other bands of M.W. 100,000, 50,000, 30,000 appear. When trypsin is inactivated by TLCK 100,000 M.W. band is present, accompanied by the 200,000 M.W. band whose intensity is function of the alpha2 M concentration. The 100,000 M.W. band appears therefore characteristic of the formation of the complex which must imply a proteolytic cleavage in the middle of the 100,000 polypeptidic chain of alpha2 M. A model of the complex is proposed in which the enzyme forms a proteic bridge between the two halves of the alpha2 M molecule.

Summary Ro 4-1778/1 (1-(p-Chlorphenethyl)-6,7-dimethoxy-2-methyl-1,2,3,4-tetrahydroisoquinoline) showed in pharmacological tests distinct analgetic effects similar to those of codeine as well as some spasmolytic activity resembling that of papaverine, whereas its side effects were absent or at least considerably less pronounced than those known for codeine.     

Visual spatial memory in desert ants,Cataglyphis bicolor (Hymenoptera: Formicidae)

Summary For navigation, desert ants apply piloting and dead-reckoning strategies based on terrestrial and celestial visual cues, respectively. Visual spatial memories, rather than general concepts derived from landmark constellations, are most probably used to define points on earth by nearby landmarks. The visual field of a specialized dorsal part of the ant's retina is mapped on to the celestial sphere to consider the possibility that similar mechanisms are used to define directions on earth by exploiting skylight patterns present at infinity.Supported by the Hescheler Foundation (Zurich) and the Swiss National Science Foundation grants Nos 3.529-0.75 and 3.313-0.78.

Summary Live gram-positive and gram-negative bacteria cause circular and chemotactic eccentric promotion of leucocyte emigration in a similar manner. In the case of gramnegative bacteria this effect corresponds to that of their polysaccharides, whereas in the case of gram-positive bacteria the polysaccharides are inactive.Thus, although gram-positive and gram-negative bacteria both have a similar effect on leucocyte emigration, the underlying mechanism of action is different.
