Role of Tip60 tumor suppressor in DNA repair pathway

To prevent the damage caused by DNA strand breaks, eukaryotic cells have evolved a series of highly conserved DNA repair mechanisms. The ubiquitously expressed acetyltransferase, Tip60, plays a central role in ATM (ataxia-telangiectasia mutated) activation which is involved in DNA repair. Recent work uncovered a new mechanism of ATM activation mediated by Tip60 and demonstrated that histone methylation, specifically, trimethylation of histone H3, is a key factor in the process. Here, we review the current understanding of how Tip60 is activated and how it activates ATM in response to DNA damage.     

Accurate localization and excision of genomic islands in four strains of <Emphasis Type=Italic>Pseudomonas aeruginosa</Emphasis> and <Emphasis Type=Italic>Pseudomonas fluorescens</Emphasis>

Mobile genomic islands (GIs) can be excised from the chromosome, then form a circular intermediate and be reintegrated into the chromosome by the GI internal integrase. Some mobile GIs can also be transferred into a new receptor cell by transformation, conjugation, or transduction. The action sites of the integrase are usually flanked direct repeats (DRs) of the GIs. Accurate localization of the flanking sequences is a precondition for determining the mobility of the GI. Mobile GIs are generally associated with transfer RNAs (tRNAs). Based on the correlation between flanking sequences and tRNA sequences, the flanking sequences of 11 putative mobile GIs in Pseudomonas aeruginosa PAO1, P. aeruginosa PA14, P. fluorescens Pf-5 and P. fluorescens Pf0-1 were identified. Among the 11 GIs, Pf0-1GI-1 is responsible for benzoate degradation. PAO1GI-1, Pf5GI-2, Pf5GI-3, and Pf5GI-4 were confirmed experimentally to be excised from a chromosome to form a circular intermediate. The action sites of the integrases are these GIs direct repeats. Due to distinct DRs, cutting sites for the internal integrase of PAO1GI-1, Pf5GI-2, Pf5GI-3 and Pf5GI-4 were determined outside the T-loop of the tRNA^Gly gene, outside the anticodon loop of the tRNA^Ser gene and tRNA^Lys gene, and at the asymmetric 3′-end of the tRNA^Leu gene, respectively. PAO1GI-1 and other mobile GIs may be transferred into many different strains that belong to different phyla because of the clear flanking sequences. This study describes basic information about the action sites of the integrases, assesses the mobility of GIs, and can help design and transfer mobile GIs to candidate strains.     

Emission inventories of primary particles and pollutant gases for China

Detailed high-resolution emission inventories of primary particles(PM2.5,BC and OC) and pollutant gases(SO2,NOx,NH3,CO and VOCs) for China in 2007 were constructed on the basis of the latest fuel consumption data,mostly at the county level,and from socio-economic statistics and data on fossil and biomass fuels obtained from government agencies.New emission factors reflecting local features were also used.The calculated emissions were 13.212 Mt PM2.5,1.4 Mt BC,2.946 Mt OC,31.584 Mt SO2,23.248 Mt NOx,16.017 Mt NH3,164.856 Mt CO and 35.464 Mt VOCs.The national and regional emissions were gridded with 0.5°× 0.5° resolution for use in air quality models.Larger emissions were found in eastern and central China than in western China.The emissions estimated here are roughly equal to those obtained in previous studies,but with different contributions from because of seasonal changes in residential heating and biomass combustion.Finally,uncertainties in inventories were analyzed.     

A Note on the Property of ψ-Function

About the accurate value of ψ ( x ) = Γ′ ( x ) / Γ ( x), we know only ψ (1)and ψ (1/ 2).In this paper, by calculating the accurate values of some series expansion of a function and applying some skills of complex analysis, we prove that the expression of the ψ-function admits a finite expression in elementary function for rational number z. As an application, some examples are given.     

开放式普通物理实验教学研究与实践

在理工科专业的普通物理课程的教学过程中,进行开放式物理实验教学能调动学生的学习积极性并培养学生的动手能力。针对大学普通物理课程的实验教学,首先对开放式物理实验的教学过程及其相关的各方面进行了讨论,然后针对Internet环境,论述了具有实验预约与实验教学管理等功能的开放式实验教学信息系统的实现。     

两性霉素B脂质卷小鼠口服后的药物组织分布研究

研究两性霉素B脂质卷小鼠口服后的组织分布行为. 将Balb/C小鼠分为2组,口服组灌胃给予10 mg·kg-1两性霉素B脂质卷,静脉注射组尾静脉给予1 mg·kg-1两性霉素B粉针注射溶液,于不同时间点处死小鼠,取心、肝、脾、肺、肾等组织和血浆,用液相-串联质谱法检测两性霉素B在组织和血浆中的药物质量分数.结果显示:脂质卷口服后组织中12 h药物质量分数最高,药物在肝、脾、肺中分布较多,心、肾、血浆中分布较少.粉针注射组药物质量分数在相同的时间点随着时间的延长组织中的质量分数呈下降趋势,其中肝组织中质量分数相对较高,脾、肾、肺、血浆次之,心中质量分数最低.2组12 h心和肺组织质量分数相当,而其他组织中各时间点粉针注射组质量分数均高于脂质卷口服组的质量分数.证实脂质卷可显著促进两性霉素B的口服吸收,药物在肝、脾、肺中分布较多.

自回转刀具切削SiC_w/LD_2铝基复合材料的切削力

针对自回转刀具切削碳化硅晶须增强铝基复合材料时的切削力特性问题,采用自回转刀具和圆形固定刀具切削SiC_w/LD_2铝基复合材料、45钢和铝合金,用Kistler测力系统检测和分析切削力.试验结果表明,自回转刀具与圆形固定刀具相比平均背吃刀抗力F_p减小了30%—60%,峰值减小了70%—80%;随着切削速度的增加自回转刀具的切削力呈快速减小的趋势,其切削力系数K_p1.圆形固定刀具的切削力亦呈减小的趋势,但切削力系数Kp1;随着进给量的增大,固定刀具的切削力增加剧烈,其切削力系数K_p1,而自回转刀具的切削力增幅很小,且切削力系数K_p由大于1转变为小于1,减小的幅度明显;自回转刀具是加工中低体积分数铝基复合材料时的高性能低成本刀具之一,宜采用高切削速度、大进给量的方式进行加工.

“决明山楂燕麦胶囊”的毒理学试验

目的观察决明山楂燕麦胶囊的急性及长期毒性反应,以确保用药安全性。方法急性毒性反应以10 g/kg(小鼠最大耐受量)的决明山楂燕麦给小鼠一次性灌胃,连续观察14 d,逐日记录毒性反应、死亡数,并作病理学检查。长期毒性反应以低剂量组1.6 g/kg、高剂量组2 g/kg给大鼠连续灌服8周,对大鼠进行一般状况、血液学指标、血液生化指标、主要脏器指数和组织病理学观察和检查。结果急性毒性实验未见小鼠出现毒性反应及死亡。长期毒性结果显示,低、高剂量组大鼠的一般状况、体质量、血液学指标、血液生化学指标及主要脏器系数与对照组比较无显著性差异;肉眼观察服药组心、肝、脾、肺、肾外观形态、颜色均无异常改变,光镜检查也未见各脏器组织结构和细胞形态异常。结论决明山楂燕麦对实验动物安全无毒,这可为临床应用提供有用信息。
