Deafness and renal tubular acidosis in mice lacking the K-Cl co-transporter Kcc4

Hearing depends on a high K(+) concentration bathing the apical membranes of sensory hair cells. K(+) that has entered hair cells through apical mechanosensitive channels is transported to the stria vascularis for re-secretion into the scala media(). K(+) probably exits outer hair cells by KCNQ4 K(+) channels(), and is then transported by means of a gap junction system connecting supporting Deiters' cells and fibrocytes() back to the stria vascularis. We show here that mice lacking the K(+)/Cl(-) (K-Cl) co-transporter Kcc4 (coded for by Slc12a7) are deaf because their hair cells degenerate rapidly after the beginning of hearing. In the mature organ of Corti, Kcc4 is restricted to supporting cells of outer and inner hair cells. Our data suggest that Kcc4 is important for K(+) recycling() by siphoning K(+) ions after their exit from outer hair cells into supporting Deiters' cells, where K(+) enters the gap junction pathway. Similar to some human genetic syndromes(), deafness in Kcc4-deficient mice is associated with renal tubular acidosis. It probably results from an impairment of Cl(-) recycling across the basolateral membrane of acid-secreting alpha-intercalated cells of the distal nephron.     

Skewed superstatistical distributions from a Langevin and Fokker-Planck approach

The superstatistics concept is a useful statistical method to describe inhomogeneous complex systems for which a system parameter βfluctuatesonalargespatio-temporal scale. Inthispaper weanalyze ameasuredtimeseriesofwindspeed fluctuationsandextract the superstatistical distribution function f(β) directly from the data. We construct suitable Langevin and Fokker-Planck models with a position dependent β-field and show that they reduce to standard type of superstatistics in the overdamped limit.     

Initial sequencing and comparative analysis of the mouse genome

The sequence of the mouse genome is a key informational tool for understanding the contents of the human genome and a key experimental tool for biomedical research. Here, we report the results of an international collaboration to produce a high-quality draft sequence of the mouse genome. We also present an initial comparative analysis of the mouse and human genomes, describing some of the insights that can be gleaned from the two sequences. We discuss topics including the analysis of the evolutionary forces shaping the size, structure and sequence of the genomes; the conservation of large-scale synteny across most of the genomes; the much lower extent of sequence orthology covering less than half of the genomes; the proportions of the genomes under selection; the number of protein-coding genes; the expansion of gene families related to reproduction and immunity; the evolution of proteins; and the identification of intraspecies polymorphism.     

The effects of two neutral polymers on the geometry and deformability of the human erythrocyte

Summary Polyvinylpyrrolidone and dextran decrease cellular deformability. Changes in volume do not wholly account for the changes which imply a stiffening of the plasma membrane. The effects differ from those induced by charged macromolecules.The work was supported by the Medical Research Council of Canada.

继电保护管理系统中关键业务的应用开发离不开工作流技术的支持。针对于此,通过对系统的实际开发需求的分析,在传统的关系数据库的基础上,提出了一个适用于关键业务开发的基于关系数据库的轻量级工作流引擎的框架结构。深入讨论了采用关系数据库来设计工作流引擎的原因,并详细地给出了相关的设计原理以及具体的表示和实现方法。其原型已经应用到实际的继电保护管理系统中。实践证明,利用此工作流引擎可以显著地缩短业务的开发周期。     

Peat record reflecting Holocene climatic change in the Zoige Plateau and AMS radiocarbon dating

Through the use of reliable AMS dating of high resolution (15-30 years) peat and the establishment of monsoon climate proxies sequence, we have been able to recognize several cold, dry events in the Tibetan Plateau during the Holocene. The more obvious ones occurred around 12800, 11300, 10200, 9580, 8900, 6400, 4400, 3700, 2800 and 1500 cal. aBP. These events correlate well with both ice rafting events recorded in high latitude North Atlantic Ocean sediment cores and cooling events in the low latitude SST. Spectral analysis indicates high frequency climate variation on centennial-millennial time scale during the Holocene. This further reflects Holocene climate instability and the existence of centennial-millenium scale rhythm in mid latitude areas as well.     

Effects of mistletoe (Viscum album L.) extracts on cultured tumor cells

Summary Bacterially fermented mistletoe preparations (BFMP) were tested on rat hepatoma tissue culture (HTC) cells and human leukemia Molt 4 cells. A dose-dependent inhibition of the growth rate of the cells was observed. For both cell lines, cytostatic concentrations, expressed in weight of fresh plant, were 0.5 mg/ml culture medium for oak BFMP and 1 mg/ml for apple tree BFMP. However, the action of the two preparations was markedly different on each cell line. Non-viable HTC cells were not stained by trypan blue while non-viable Molt 4 cells were fully colored by this reagent. A lysis of cellular membranes of HTC cells was observed by electron microscopy. Furthermore, oak BFMP inhibited the growth of virus transformed 3T3-SV40 cells more than that of non-transformed 3T3 cells. In contrast to BFMP, non-fermented extracts and a purified mistletoe lectin showed a greater inhibition of the growth of Molt 4 cells than of HTC cells. Samples withdrawn at different times during fermentation gradually lost their inhibitory effect on the growth of Molt 4 cells while their action on HTC cells increased up to the 4th day of fermentation. These results are discussed in relation to the cytotoxic substances of mistletoe already characterized.The bacterially fermented mistletoe preparations, named BFMP in the text, were obtained from the Hiscia Institute, CH-4144 Arlesheim, Switzerland, under the name of Iscador. For oak BFMP, mistletoe was fromQuercus petraea Liebl. andQuercus robur L.; for apple tree BFMP fromMalus domestica Borkh.
