The expression of the Escherichia coli DNA polymerases pol V (UmuD'2C complex) and pol IV (DinB) increases in response to DNA damage. The induction of pol V is accompanied by a substantial increase in mutations targeted at DNA template lesions in a process called SOS-induced error-prone repair. Here we show that the common DNA template lesions, TT (6-4) photoproducts, TT cis-syn photodimers and abasic sites, are efficiently bypassed within 30 seconds by pol V in the presence of activated RecA protein (RecA*), single-stranded binding protein (SSB) and pol III's processivity beta,gamma-complex. There is no detectable bypass by either pol IV or pol III on this time scale. A mutagenic 'signature' for pol V is its incorporation of guanine opposite the 3'-thymine of a TT (6-4) photoproduct, in agreement with mutational spectra. In contrast, pol III and pol IV incorporate adenine almost exclusively. When copying undamaged DNA, pol V exhibits low fidelity with error rates of around 10(-3) to 10(-4), with pol IV being 5- to 10-fold more accurate. The effects of RecA protein on pol V, and beta,gamma-complex on pol IV, cause a 15,000- and 3,000-fold increase in DNA synthesis efficiency, respectively. However, both polymerases exhibit low processivity, adding 6 to 8 nucleotides before dissociating. Lesion bypass by pol V does not require beta,gamma-complex in the presence of non-hydrolysable ATPgammaS, indicating that an intact RecA filament may be required for translesion synthesis.  相似文献

Ceramics based on Si(3)N(4) have been comprehensively studied and are widely used in structural applications. The development of an interlocking microstructure of elongated grains is vital to ensure that this family of ceramics have good damage tolerance. Until now this has been accomplished by heating the appropriate powder compacts to temperatures above 1,700 degrees C for extended periods. This procedure involves a necessary step of controlling the size and population of seeds added ex situ or formed in situ to ensure selective grain growth. Here we report the very fast (within minutes) in situ formation of a tough interlocking microstructure in Si(3)N(4)-based ceramics. The microstructures are obtained by a dynamic ripening mechanism, an anisotropic Ostwald ripening process that results from the rapid heating rate. The resulting microstructures are uniform and reproducible in terms of grain size distribution and mechanical properties, and are easily tailored by manipulating the kinetics. This process is very efficient and opens up new possibilities to optimize mechanical properties and cost-effectively manufacture ceramics.  相似文献

A robust H∞ approach to filtering for a class of uncertain linear systems with time-delayed measurement is investigated.The parameter uncertainties are described by real time-varying norm-bounded form.A methodology is developed for designing linear filters such that the filtering process remains robustly stable and the transfer function from the disturbance inputs to error state outputs meets the prescribed H∞-bound constraints.The filter can be obtained and parameterized by solving two Riccati equations based on the lemma of robust stability for control system with delay.Simulation example proves the method to be effective and feasible.  相似文献

Retroviral insertional mutagenesis in BXH2 and AKXD recombinant inbred mice induces a high incidence of myeloid or B- and T-cell leukaemia and the proviral integration sites in the leukaemias provide powerful genetic tags for disease gene identification. Some of the disease genes identified by proviral tagging are also associated with human disease, validating this approach for human disease gene identification. Although many leukaemia disease genes have been identified over the years, many more remain to be cloned. Here we describe an inverse PCR (IPCR) method for proviral tagging that makes use of automated DNA sequencing and the genetic tools provided by the Mouse Genome Project, which increases the throughput for disease gene identification. We also use this IPCR method to clone and analyse more than 400 proviral integration sites from AKXD and BXH2 leukaemias and, in the process, identify more than 90 candidate disease genes. Some of these genes function in pathways already implicated in leukaemia, whereas others are likely to define new disease pathways. Our studies underscore the power of the mouse as a tool for gene discovery and functional genomics.  相似文献

A novel process for boron enrichment and extraction from ludwigite based on iron nugget technology was proposed. The key steps of this novel process, which include boron and iron separation, crystallization of boron-rich slag, and elucidation of the boron extraction behavior of boron-rich slag by acid leaching, were performed at the laboratory. The results indicated that 95.7% of the total boron could be enriched into the slag phase, thereby forming a boron-rich slag during the iron and slag melting separation process. Suanite and kotoite were observed to be the boron-containing crystalline phases, and the boron extraction properties of the boron-rich slag depended on the amounts and grain sizes of these minerals. When the boron-rich slag was slowly cooled to 1100℃, the slag crystallized well and the efficiency of extraction of boron (EEB) of the slag was the highest observed in the present study. The boron extraction property of the slow-cooled boron-rich slag obtained in this study was much better than that of szaibelyite ore under the conditions of 80% of theoretical sulfuric acid amount, leaching time of 30 min, leaching temperature of 40℃, and liquid-to-solid ratio of 8 mL/g.  相似文献

In this paper, the diffusion behavior between MgO and Fe2O3 (the main iron oxide in pellets) is investigated using a diffusion couple method. In addition, the distribution regulation of MgO in MgO-bearing pellets is analyzed via pelletizing experiments. The results illustrate that MgO is prone to diffuse into Fe2O3 in the form of solid solution; the diffusion rate considered here is 13.64 μm·min-1. Most MgO content distributes in the iron phase instead of the slag phase. The MF phase {(Mg1-x Fex)O·Fe2O3, x ≤ 1} is generated in the MgO-bearing pellets. However, the distribution of MgO in the radial direction of the pellets is inconsistent. The solid solution portion of MgO in the MF phase is larger in the outer layer of the pellets than in the inner layer. In this work, the approximate chemical composition of the MF phase in the outer layer of the pellets is {(Mg0.35-0.77·Fe0.65-0.23) O·Fe2O3} and in the inner layer is {(Mg0.13-0.45·Fe0.87-0.55) O·Fe2O3}.  相似文献
