Mammalian cells have three ATP-dependent DNA ligases, which are required for DNA replication and repair. Homologues of ligase I (Lig1) and ligase IV (Lig4) are ubiquitous in Eukarya, whereas ligase III (Lig3), which has nuclear and mitochondrial forms, appears to be restricted to vertebrates. Lig3 is implicated in various DNA repair pathways with its partner protein Xrcc1 (ref. 1). Deletion of Lig3 results in early embryonic lethality in mice, as well as apparent cellular lethality, which has precluded definitive characterization of Lig3 function. Here we used pre-emptive complementation to determine the viability requirement for Lig3 in mammalian cells and its requirement in DNA repair. Various forms of Lig3 were introduced stably into mouse embryonic stem (mES) cells containing a conditional allele of Lig3 that could be deleted with Cre recombinase. With this approach, we find that the mitochondrial, but not nuclear, Lig3 is required for cellular viability. Although the catalytic function of Lig3 is required, the zinc finger (ZnF) and BRCA1 carboxy (C)-terminal-related (BRCT) domains of Lig3 are not. Remarkably, the viability requirement for Lig3 can be circumvented by targeting Lig1 to the mitochondria or expressing Chlorella virus DNA ligase, the minimal eukaryal nick-sealing enzyme, or Escherichia coli LigA, an NAD(+)-dependent ligase. Lig3-null cells are not sensitive to several DNA-damaging agents that sensitize Xrcc1-deficient cells. Our results establish a role for Lig3 in mitochondria, but distinguish it from its interacting protein Xrcc1.  相似文献

The stability and spontaneous decay of naturally occurring atomic nuclei have been much studied ever since Becquerel discovered natural radioactivity in 1896. In 1960, proton-rich nuclei with an odd or an even atomic number Z were predicted to decay through one- and two-proton radioactivity, respectively. The experimental observation of one-proton radioactivity was first reported in 1982, and two-proton radioactivity has now also been detected by experimentally studying the decay properties of 45Fe (refs 3, 4) and 54Zn (ref. 5). Here we report proton-proton correlations observed during the radioactive decay of a spinning long-lived state of the lightest known isotope of silver, 94Ag, which is known to undergo one-proton decay. We infer from these correlations that the long-lived state must also decay through simultaneous two-proton emission, making 94Ag the first nucleus to exhibit one- as well as two-proton radioactivity. We attribute the two-proton emission behaviour and the unexpectedly large probability for this decay mechanism to a very large deformation of the parent nucleus into a prolate (cigar-like) shape, which facilitates emission of protons either from the same or from opposite ends of the 'cigar'.  相似文献

Obesity increases the risk of cardiovascular disease and premature death. Adipose tissue releases a large number of bioactive mediators that influence not only body weight homeostasis but also insulin resistance - the core feature of type 2 diabetes - as well as alterations in lipids, blood pressure, coagulation, fibrinolysis and inflammation, leading to endothelial dysfunction and atherosclerosis. We are now beginning to understand the underlying mechanisms as well as the ways in which smoking and dyslipidaemia increase, and physical activity attenuates, the adverse effects of obesity on cardiovascular health.  相似文献

Although psychological differences appear to be a factor in political and social division, there are few process‐based accounts of the relationship between political orientation and personality. We present a complex adaptive systems model with biological differences at the micro level and sociocultural structures at the macro level, both of which exert a bidirectional influence on personality at the meso level. This provides the foundation for a process‐based model in which social interactions act as feedback loops that encourage the development of biases at the biological level, accentuating individual differences while driving collective adaptation to changing environments. Cyclical phases in adaptive processes are then associated with emergent political values. Therefore, political and social division is an inevitable feature of an adaptive society and results from interactions between individuals responding to stress. Using this model, we are able to account for both congruence and incongruence between personality and political orientation.  相似文献

Four activated carbon(AC) samples prepared from rice husk under different activation temperatures have been characterized by N2adsorption–desorption isotherms, thermogravimetric analysis(TGA–DTA), Fourier transform infrared spectroscopy(FTIR) and scanning electron microscopy(SEM). The specific surface area of AC sample reached 2681 m2 g 1under activation temperature of 800 1C. The AC samples were then tested as electrode material; the specific capacitance of the as-prepared activated carbon electrode was found to be 172.3 F g 1using cyclic voltammetry at a scan rate of 5 mV s 1and 198.4 F g 1at current density 1000 mA g 1in the charge/discharge mode.& 2014 Chinese Materials Research Society.
