Regulated apoptosis is essential for both the development and the subsequent maintenance of the immune system. Interleukins, including IL-2, IL-4, IL-7 and IL-15, heavily influence lymphocyte survival during the vulnerable stages of VDJ rearrangement and later in ensuring cellular homeostasis, but the genes specifically responsible for the development and maintenance of lymphocytes have not been identified. The antiapoptotic protein MCL-1 is an attractive candidate, as it is highly regulated, appears to enhance short-term survival and functions at an apical step in genotoxic deaths. However, Mcl-1 deficiency results in peri-implantation lethality. Here we show that mice conditional for Mcl-1 display a profound reduction in B and T lymphocytes when MCL-1 is removed. Deletion of Mcl-1 during early lymphocyte differentiation increased apoptosis and arrested the development at pro-B-cell and double-negative T-cell stages. Induced deletion of Mcl-1 in peripheral B- and T-cell populations resulted in their rapid loss. Moreover, IL-7 both induced and required MCL-1 to mediate lymphocyte survival. Thus, MCL-1, which selectively inhibits the proapoptotic protein BIM, is essential both early in lymphoid development and later on in the maintenance of mature lymphocytes.  相似文献

In an electric field, the flow of electrons in a solid produces an entropy current in addition to the familiar charge current. This is the Peltier effect, and it underlies all thermoelectric refrigerators. The increased interest in thermoelectric cooling applications has led to a search for more efficient Peltier materials and to renewed theoretical investigation into how electron-electron interaction may enhance the thermopower of materials such as the transition-metal oxides. An important factor in this enhancement is the electronic spin entropy, which is predicted to dominate the entropy current. However, the crucial evidence for the spin-entropy term, namely its complete suppression in a longitudinal magnetic field, has not been reported until now. Here we report evidence for such suppression in the layered oxide Na(x)Co2O4, from thermopower and magnetization measurements in both longitudinal and transverse magnetic fields. The strong dependence of thermopower on magnetic field provides a rare, unambiguous example of how strong electron-electron interaction effects can qualitatively alter electronic behaviour in a solid. We discuss the implications of our finding--that spin-entropy dominates the enhancement of thermopower in transition-metal oxides--for the search for better Peltier materials.  相似文献

A facile and green preparation of high surface area activated carbons with mixed microporosity and mesoporosity from durian shell waste is reported in this work. The pore structure and surface chemistry of the parent carbon were modified by the combination of ultrasonication and microwave irradiation techniques. The effects of temperature and time in the ultrasonication treatment and power output and time in the microwave irradiation were studied. The electrochemical performance of carbon electrodes for supercapacitors was tested by cyclic voltammeter (CV) and galvanostatic charge–discharge. The results show that the capacitive energy storage of electrodes is critically dependent on the microporosity and surface chemistry of activated carbons. The highest electrode capacitance in this work was 103.6 F/g that prepared from activated carbon modified at an ultrasonication temperature of 323.15 K for 10 min and microwave power output of 900 W for 10 min.  相似文献

Identification of complementary DNAs encoding the human glucocorticoid receptor predicts two protein forms, of 777 (alpha) and 742 (beta) amino acids, which differ at their carboxy termini. The proteins contain a cysteine/lysine/arginine-rich region which may define the DNA-binding domain. Pure radiolabelled glucocorticoid receptor, synthesized in vitro, is immunoreactive and possesses intrinsic steroid-binding activity characteristic of the native glucocorticoid receptor.  相似文献

Retinoic acid is known to have dramatic effects on vertebrate limb pattern in development and regeneration, supporting a model in which a gradient of retinoic acid serves as a morphogen to differentially supply positional information to a developing limb. The discovery of a retinoic acid receptor (RAR) and its homology to the steroid and thyroid hormone receptors provided a potential molecular mechanism for limb morphogenesis. One prediction of this model is that the receptor must be expressed in the developing and regenerating limb anlage. We investigated the expression of the RAR in the adult newt, Notophthalmus viridescens, whose amputated limbs are capable of regenerating and upon which retinoic acid can act to alter pattern. We report the cloning of cDNAs encoding a functional newt RAR and the localization of high and uniform levels of RAR mRNA specifically in the regenerating cells that control limb pattern. These results indicate that the morphogenic field is established through differential activation of pre-existing retinoic acid receptors rather than differential expression of the RAR gene.  相似文献
