Ankyrin binding to (Na+ + K+)ATPase and implications for the organization of membrane domains in polarized cells 总被引:8,自引:0,他引:8
The interaction between membrane proteins and cytoplasmic structural proteins is thought to be one mechanism for maintaining the spatial order of proteins within functional domains on the plasma membrane. Such interactions have been characterized extensively in the human erythrocyte, where a dense, cytoplasmic matrix of proteins comprised mainly of spectrin and actin, is attached through a linker protein, ankyrin, to the anion transporter (Band 3). In several nonerythroid cell types, including neurons, exocrine cells and polarized epithelial cells homologues of ankyrin and spectrin (fodrin) are localized in specific membrane domains. Although these results suggest a functional linkage between ankyrin and fodrin and integral membrane proteins in the maintenance of membrane domains in nonerythroid cells, there has been little direct evidence of specific molecular interactions. Using a direct biological and chemical approach, we show here that ankyrin binds to the ubiquitous (Na+ + K+)ATPase, which has an asymmetrical distribution in polarized cells. 相似文献

Titan is the only satellite in our Solar System with a dense atmosphere. The surface pressure is 1.5 bar (ref. 1) and, similar to the Earth, N2 is the main component of the atmosphere. Methane is the second most important component, but it is photodissociated on a timescale of 10(7) years (ref. 3). This short timescale has led to the suggestion that Titan may possess a surface or subsurface reservoir of hydrocarbons to replenish the atmosphere. Here we report near-infrared images of Titan obtained on 26 October 2004 by the Cassini spacecraft. The images show that a widespread methane ocean does not exist; subtle albedo variations instead suggest topographical variations, as would be expected for a more solid (perhaps icy) surface. We also find a circular structure approximately 30 km in diameter that does not resemble any features seen on other icy satellites. We propose that the structure is a dome formed by upwelling icy plumes that release methane into Titan's atmosphere. 相似文献

Efforts to identify gene variants associated with susceptibility to common diseases use three approaches: pedigree and affected sib-pair linkage studies and association studies of population samples. The different aims of these study designs reflect their derivation from biological versus epidemiological traditions. Similar principles regarding determination of the evidence levels required to consider the results statistically significant apply to both linkage and association studies, however. Such determination requires explicit attention to the prior probability of particular findings, as well as appropriate correction for multiple comparisons. For most common diseases, increasing the sample size in a study is a crucial step in achieving statistically significant genetic mapping results.
