Bronchial asthma is a common inflammatory disease caused by the interaction of genetic and environmental factors. Through a genome-wide association study and a replication study consisting of a total of 7,171 individuals with adult asthma (cases) and 27,912 controls in the Japanese population, we identified five loci associated with susceptibility to adult asthma. In addition to the major histocompatibility complex and TSLP-WDR36 loci previously reported, we identified three additional loci: a USP38-GAB1 locus on chromosome 4q31 (combined P = 1.87 × 10(-12)), a locus on chromosome 10p14 (P = 1.79 × 10(-15)) and a gene-rich region on chromosome 12q13 (P = 2.33 × 10(-13)). We observed the most significant association with adult asthma at rs404860 in the major histocompatiblity complex region (P = 4.07 × 10(-23)), which is close to rs2070600, a SNP previously reported for association with FEV(1)/FVC in genome-wide association studies for lung function. Our findings offer a better understanding of the genetic contribution to asthma susceptibility. 相似文献

Summary 4-Methoxyphenylethylamine was found to be a specific substrate for type B monoamine oxidase (MAO) in rat brain mitochondria, whereas 3,4-dimethoxyphenylethylamine was common for both types of MAO. In human skin transplanted to the back of 3 strains of immuno-deficient mice the functin of the eccrine sweat glands of the human transplant was tested by topical intradermal application of pilocarine, adrenaline and atropine+pilocarpine. Sweat responses were observed in pre-selected fields of observation by means of video macroscope. The iodine strarch reaction served as an indicator for the appearance of seat sport and permitted the evaluation of areas wetted by sweat in the field of observation. Among 9 animals tested, the hybrids between the CB-17-scid mouse and the BALB/cA-nu mouse (BALB/cA-nu,scid) seemed to exhibit the most consistent seweating response to local pharmacological stimulation. According to histological examination, eccrine sweat glands were preserved in human skin trasplanted into the back skin of the BALB/cA-nu,scid mouse strain. the heterologous, human skin graft provides a novel model permitting, independent of the normal sweat gland innervation, the analysis of moecular receptors of sweat gland cells by which the actions of natural transmitters and pharmacological agents are transduced. 相似文献

Summary Physalaemin, which excites an identifiable molluscan giant neurone (the TAN, tonically autoactive neurone), lost the effect after the trypsin treatment. Unexpectedly, this peptide shows an inhibitory effect on the same neurone after chymotrypsin treatment. Deamino-dicarba-(d-d-)oxytocin and d-d-Arg-vasotocin, which excite another identifiable neurone (the PON, periodically oscillating neurone) continue to show the effect after chymotrypsin treatment (6 h). Summary Erythrocytes of vitamin E-deficient kids and chicks were hemolyzed at concentrations of Tween 20 above 1%. In vivo or in vitro uptake of vitamin E by the erythrocytes, or the addition of dithiothreitol or 2-mercaptoethanol, prevented the hemolysis with Tween 20. 相似文献
