A putative growth hormone receptor from rabbit liver and the growth hormone binding protein from rabbit serum have the same amino-terminal amino-acid sequence, indicating that the binding protein corresponds to the extracellular hormone-binding domain of the liver receptor. The complete amino-acid sequences derived from complementary DNA clones encoding the putative human and rabbit growth hormone receptors are not similar to other known proteins, demonstrating a new class of transmembrane receptors. 相似文献

Voltage-gated K+ (Kv) channels exhibit slow or C-type inactivation during continuous depolarization. A selective pharmacological agent targeting
C-type inactivation is hitherto lacking. Here, we report that 6β-acetoxy-7α-hydroxyroyleanone (AHR), a diterpenoid compound
isolated from Taiwania cryptomerioides, can selectively modify C-type inactivation of Kv1.2 channels. Extracellular, but not intracellular, AHR (50 μM) dramatically
accelerated the slow decay of Kv currents and left-shifted the steady-state inactivation curve. AHR blocked Kv currents with
an IC50 of 17.7 μM. AHR did not affect the kinetics and voltage-dependence of Kv1.2 channel activation. Channel block by AHR was
independent of intracellular K+ concentration. In addition, effect of AHR was much attenuated in a Kv1.2 V370G mutant defective in C-type inactivation. Therefore,
block of Kv1.2 channels by AHR did not appear to involve direct occlusion of the outer pore but depended on C-type inactivation.
AHR could thus be a probe targeting Kv channel C-type inactivation gate. 相似文献

Interaction of distinct domains in Mu transposase with Mu DNA ends and an internal transpositional enhancer 总被引:37,自引:0,他引:37
Bacteriophage Mu is the largest and most efficient transposable element known. The Mu transposase (A protein) of relative molecular mass 75,000 is a central component of the transposition machinery. We report here that the N-terminal region of Mu transposase contains two distinct DNA-binding domains, one which binds the two Mu DNA ends, and another which binds an internal operator region. This internal operator is required for the transposase-mediated synapsis and nicking of Mu ends in vitro, and stimulates transposition more than 100-fold in vivo. The orientation of the operator with respect to the ends is critical to its function, whereas its distance from the ends seems to be relatively unimportant. We propose that the operator enhances transposition by transiently interacting with the transposase and Mu DNA end(s) to form a complex in which synapsis of the ends occurs. 相似文献

This paper proposes a new approach to forecasting intermittent demand by considering the effects of external factors. We classify intermittent demand data into two parts—zero value and nonzero value—and fit nonzero values into a mixed zero-truncated Poisson model. All the parameters in this model are obtained by an EM algorithm, which regards external factors as independent variables of a logistic regression model and log-linear regression model. We then calculate the probability of occurrence of zero value at each period and predict demand occurrence by comparing it with critical value. When demand occurs, we use the weighted average of the mixed zero-truncated Poisson model as predicted nonzero demands, which are combined with predicted demand occurrences to form the final forecasting demand series. Two performance measures are developed to assess the forecasting methods. By presenting a case study of electric power material from the State Grid Shanghai Electric Power Company in China, we show that our approach provides greater accuracy in forecasting than the Poisson model, the hurdle shifted Poisson model, the hurdle Poisson model, and Croston's method. 相似文献
