Flowering plants possess a unique reproductive strategy, involving double fertilization by twin sperm cells. Unlike animal germ lines, the male germ cell lineage in plants only forms after meiosis and involves asymmetric division of haploid microspores, to produce a large, non-germline vegetative cell and a germ cell that undergoes one further division to produce the twin sperm cells. Although this switch in cell cycle control is critical for sperm cell production and delivery, the underlying molecular mechanisms are unknown. Here we identify a novel F-box protein of Arabidopsis thaliana, designated FBL17 (F-box-like 17), that enables this switch by targeting the degradation of cyclin-dependent kinase A;1 inhibitors specifically in male germ cells. We show that FBL17 is transiently expressed in the male germ line after asymmetric division and forms an SKP1-Cullin1-F-box protein (SCF) E3 ubiquitin ligase complex (SCF(FBL17)) that targets the cyclin-dependent kinase inhibitors KRP6 and KRP7 for proteasome-dependent degradation. Accordingly, the loss of FBL17 function leads to the stabilization of KRP6 and inhibition of germ cell cycle progression. Our results identify SCF(FBL17) as an essential male germ cell proliferation complex that promotes twin sperm cell production and double fertilization in flowering plants.  相似文献

Poly rG can form four-stranded helices. The Hoogsteen-paired quartets of G residues on which such structures depend are so stable that they will form in 5'-GMP solutions, provided that Na+ or K+ are present (see for example, refs 2-4). Telomeric DNA sequences, which are G-rich, adopt four-stranded antiparallel G-quartet conformations in vitro, and parallel tetramerization of G-rich sequences may be involved in meiosis. Here we show that RNAs containing short runs of Gs can also tetramerize. A 19-base oligonucleotide derived from the 5S RNA of Escherichia coli (strand III), 5'GCCGAUGGUAGUGUGGGGU3', forms a K(+)-stabilized tetrameric aggregate that depends on the G residues at its 3' end. This complex is so stable that it would be surprising if similar structures do not occur in nature.  相似文献

A novel cyclin encoded by a bcl1-linked candidate oncogene   总被引:145,自引:0,他引:145  
We have previously identified a candidate oncogene (PRAD1 or D11S287E) on chromosome 11q13 which is clonally rearranged with the parathyroid hormone locus in a subset of benign parathyroid tumours. We now report that a cloned human placental PRAD1 complementary DNA encodes a protein of 295 amino acids with sequence similarities to the cyclins. Cyclins can form a complex with and activate p34cdc2 protein kinase, thereby regulating progress through the cell cycle. PRAD 1 messenger RNA levels vary dramatically across the cell cycle in HeLa cells. Addition of the PRAD1 protein to interphase clam embryo lysates containing inactive p34cdc2 kinase and lacking endogenous cyclins allows it to be isolated using beads bearing p13suc1, a yeast protein that binds cdc2 and related kinases with high affinity and coprecipitates kinase-associated proteins. Addition of PRAD1 also induces phosphorylation of histone H1, a preferred substrate of cdc2. These data suggest that PRAD1 encodes a novel cyclin whose overexpression may play an important part in the development of various tumours with abnormalities in 11q13.  相似文献

Recently, a new type of Radio Frequency IDentification (RFID) system with mobile readers is introduced. In such a system, it is more desirable for mobile readers to identify tags without a back-end server, and thus it is frequently referred as a serverless mobile RFID system. In this paper, we formalize a serverless mobile RFID system model and propose a new encryption-based system that preserves the privacy of both tags and readers in the model. In addition, we define a new adversary model for the system model and show the security of the proposed system. Throughout comparisons between ours and the other alternatives, we show that our proposed system provides a stronger reader privacy and robustness against a reader forgery attack than the competitors.  相似文献

The application of protograph low density parity check(LDPC) codes involves the encoding complexity problem.Since the generator matrices are dense,and if the positions of 1 s are irregularity,the encoder needs to store every 1 of the generator matrices by using huge chip area.In order to solve this problem,we need to design the protograph LDPC codes with circular generator matrices.A theorem concerning the circulating property of generator matrices of nonsingular protograph LDPC codes is proposed.The circulating property of generator matrix of nonsingular protograph LDPC codes can be obtained from the corresponding quasi-cyclic parity check matrix.This paper gives a scheme of constructing protograph LDPC codes with circulating generator matrices,and it reveals that the fast encoding algorithm of protograph LDPC codes has lower encoding complexity under the condition of the proposed theorem.Simulation results in additive white Gaussian noise(AWGN) channels show that the bit error rate(BER) performance of the designed codes based on the proposed theorem is much better than that of GB20600 LDPC codes and Tanner LDPC codes.  相似文献

以证明在线口传内容对口传效果的影响及收信者的特性的作用为目的。通过已有的研究,把口传内容划分为商品信息和个人经验,把口传效果划分为口传受用和口传行为。以大学生为对象,对MP3购买活动和相关在线口传活动进行了268个问卷调查,并用SPSS 10.0统计法进行了分析。研究结果表明:第一,口传信息的受用度使商品资讯更高,口传行为使个人体验资讯更为受用。第二,商品资讯和个人体验对口传内容的效果(口传受用,口传行为)决定于收信者的产品知识,即对在线的口传内容和口传信息受用的关系有调节作用,而且,对口传行为的影响也有调节效果。最后,关联度变数如先前所设的口传内容和口传效果的关系,对它们的关系有调节作用。  相似文献

Entanglement is recognized as a key resource for quantum computation and quantum cryptography. For quantum metrology, the use of entangled states has been discussed and demonstrated as a means of improving the signal-to-noise ratio. In addition, entangled states have been used in experiments for efficient quantum state detection and for the measurement of scattering lengths. In quantum information processing, manipulation of individual quantum bits allows for the tailored design of specific states that are insensitive to the detrimental influences of an environment. Such 'decoherence-free subspaces' (ref. 10) protect quantum information and yield significantly enhanced coherence times. Here we use a decoherence-free subspace with specifically designed entangled states to demonstrate precision spectroscopy of a pair of trapped Ca+ ions; we obtain the electric quadrupole moment, which is of use for frequency standard applications. We find that entangled states are not only useful for enhancing the signal-to-noise ratio in frequency measurements--a suitably designed pair of atoms also allows clock measurements in the presence of strong technical noise. Our technique makes explicit use of non-locality as an entanglement property and provides an approach for 'designed' quantum metrology.  相似文献

Telomere dysfunction may result in chromosomal abnormalities, DNA damage responses, and even cancer. Early studies in lower organisms have helped to establish the crucial role of telomerase and telomeric proteins in maintaining telomere length and protecting telomere ends. In Oxytricha nova, telomere G-overhangs are protected by the TEBP-alpha/beta heterodimer. Human telomeres contain duplex telomeric repeats with 3' single-stranded G-overhangs, and may fold into a t-loop structure that helps to shield them from being recognized as DNA breaks. Additionally, the TEBP-alpha homologue, POT1, which binds telomeric single-stranded DNA (ssDNA), associates with multiple telomeric proteins (for example, TPP1, TIN2, TRF1, TRF2 and RAP1) to form the six-protein telosome/shelterin and other subcomplexes. These telomeric protein complexes in turn interact with diverse pathways to form the telomere interactome for telomere maintenance. However, the mechanisms by which the POT1-containing telosome communicates with telomerase to regulate telomeres remain to be elucidated. Here we demonstrate that TPP1 is a putative mammalian homologue of TEBP-beta and contains a predicted amino-terminal oligonucleotide/oligosaccharide binding (OB) fold. TPP1-POT1 association enhanced POT1 affinity for telomeric ssDNA. In addition, the TPP1 OB fold, as well as POT1-TPP1 binding, seemed critical for POT1-mediated telomere-length control and telomere-end protection in human cells. Disruption of POT1-TPP1 interaction by dominant negative TPP1 expression or RNA interference (RNAi) resulted in telomere-length alteration and DNA damage responses. Furthermore, we offer evidence that TPP1 associates with the telomerase in a TPP1-OB-fold-dependent manner, providing a physical link between telomerase and the telosome/shelterin complex. Our findings highlight the critical role of TPP1 in telomere maintenance, and support a yin-yang model in which TPP1 and POT1 function as a unit to protect human telomeres, by both positively and negatively regulating telomerase access to telomere DNA.  相似文献

As ubiquitous sensor networks (USN) technologies and its middleware are still at its early stages, the system of the USN relies on the middleware and applications. The past sensor networks are assumed to be designed for specific applications, having data communication protocols tightly coupled to applications. To avoid these problems, several kinds of USN middleware have been researched and developed. However, most middleware of USN are still restricted by its own infrastructure so far.
