Beethoven, a mouse model for dominant, progressive hearing loss DFNA36   总被引:9,自引:0,他引:9  
Despite recent progress in identifying genes underlying deafness, there are still relatively few mouse models of specific forms of human deafness. Here we describe the phenotype of the Beethoven (Bth) mouse mutant and a missense mutation in Tmc1 (transmembrane cochlear-expressed gene 1). Progressive hearing loss (DFNA36) and profound congenital deafness (DFNB7/B11) are caused by dominant and recessive mutations of the human ortholog, TMC1 (ref. 1), for which Bth and deafness (dn) are mouse models, respectively.  相似文献

During embryogenesis, multipotent progenitors within the single-layered surface epithelium differentiate to form the epidermis and its appendages. Here, we show that microRNAs (miRNAs) have an essential role in orchestrating these events. We cloned more than 100 miRNAs from skin and show that epidermis and hair follicles differentially express discrete miRNA families. To explore the functional significance of this finding, we conditionally targeted Dicer1 gene ablation in embryonic skin progenitors. Within the first week after loss of miRNA expression, cell fate specification and differentiation were not markedly impaired, and in the interfollicular epidermis, apoptosis was not markedly increased. Notably, however, developing hair germs evaginate rather than invaginate, thereby perturbing the epidermal organization. Here we characterize miRNAs in skin, the existence of which was hitherto unappreciated, and demonstrate their differential expression and importance in the morphogenesis of epithelial tissues within this vital organ.  相似文献

PALB2 was recently identified as a nuclear binding partner of BRCA2. Biallelic BRCA2 mutations cause Fanconi anemia subtype FA-D1 and predispose to childhood malignancies. We identified pathogenic mutations in PALB2 (also known as FANCN) in seven families affected with Fanconi anemia and cancer in early childhood, demonstrating that biallelic PALB2 mutations cause a new subtype of Fanconi anemia, FA-N, and, similar to biallelic BRCA2 mutations, confer a high risk of childhood cancer.  相似文献

Catabolism may give rise to toxic intermediates that compromise cell vitality, such as epoxide formation in the recently elucidated and apparently universal bacterial coenzyme A (CoA)-dependent degradation of phenylacetic acid. This compound is central to the catabolism of a variety of aromatics, such as phenylalanine, lignin-related compounds or environmental contaminants. The key phenylacetyl-CoA monooxygenase (epoxidase) of the pathway, PaaABCE, is also connected to the production of various primary and secondary metabolites, as well as to the virulence of certain pathogens. However, the enzyme complex has so far not been investigated in detail. Here we characterize the bacterial multicomponent monooxygenase PaaABCE that, surprisingly, not only transforms phenylacetyl-CoA into its ring-1,2-epoxide, but also mediates the NADPH-dependent removal of the epoxide oxygen, regenerating phenylacetyl-CoA with formation of water. We provide evidence for a catalytic di-iron centre that is probably the key to the unprecedented deoxygenation of an organic compound by an oxygenase. Presumably, the bifunctionality is vital to avoid toxic intracellular epoxide levels if the subsequent catabolic steps are impeded. Our data suggest that detoxification is assisted by two thioesterases (PaaI and PaaY) forming non-reactive breakdown products. Hence, PaaABCE may harbour an intrinsic escape mechanism from its own toxic product and represents the archetype of a bifunctional oxygenase/deoxygenase. Analogous reactions may possibly be catalysed by other di-iron epoxidases.  相似文献

Fanconi anemia is a rare recessive disorder characterized by genome instability, congenital malformations, progressive bone marrow failure and predisposition to hematologic malignancies and solid tumors. At the cellular level, hypersensitivity to DNA interstrand crosslinks is the defining feature in Fanconi anemia. Mutations in thirteen distinct Fanconi anemia genes have been shown to interfere with the DNA-replication-dependent repair of lesions involving crosslinked DNA at stalled replication forks. Depletion of SLX4, which interacts with multiple nucleases and has been recently identified as a Holliday junction resolvase, results in increased sensitivity of the cells to DNA crosslinking agents. Here we report the identification of biallelic SLX4 mutations in two individuals with typical clinical features of Fanconi anemia and show that the cellular defects in these individuals' cells are complemented by wildtype SLX4, demonstrating that biallelic mutations in SLX4 (renamed here as FANCP) cause a new subtype of Fanconi anemia, Fanconi anemia-P.  相似文献

The New Social Movement Approach and the Resource Mobilization Approach are the dominant approaches in social movement research. They focus either on macro-aspects and externalism or on micro-aspects and internalism. This paper suggests that the notion of self-organization is one way of taking into account both internal and external, structural- and action-based aspects of social movements and that it allows a dynamic concept of protest. The emergence of social movements is not determined, but a complex result of crisis, resource mobilization, cognitive mobilization, self-production—searching for singular laws of the emergence of movements is an expression of one-dimensional, linear, and deterministic thinking. Protest and social problems are non-linearly related. Social movements are part of the civil society system, by producing alternative topics and demands, they guarantee the dynamic of the political system. Existing system-theoretic approaches on social movements (Luhmann, Japp, Ahlemeyer, Hellmann) are rather uncritical and ignore the productive relationship between human actors and social structures in processes of social self-organization. Social movements are dynamic communication systems that permanently react to political and societal events with self-organized protest practices and protest communications that result in the emergence and differentiation (production and reproduction) of protest structures (events, oppositional topics, alternative values, regularized patterns of interaction and organization). The dynamic of social movements is based on the permanent emergence and mutual production of protest practices and protest structures. The self-organization of a social movement is a vivid process, it is based on the permanent movement and differentiation of actors and structures that communicate public protest, a social movement is only a movement, as long as it communicates protest and moves itself. In critical phases of protest new social systems of protest emerge whose form, content and effects are not determined, but dependent upon old structures, i.e., old structures enable and constrain new structures. The emergence of new protest issues, methods, identities, structures, and organizational forms starts as singular innovation, if it is widely imitated then it spreads within the protest system and transforms the system as a whole. In terms of Hegelian dialectics this means that novel qualities sublate the old structure of the total system, i.e., the system is transformed, reaches a higher level, incorporates old qualities, and creates new qualities. In critical phases protest can spontaneously and quickly spread and intensify itself. This reflects the idea of complexity thinking that small causes can spontaneously have large effects. The notion of self-organization as the idea of the networked, co-operative, synergetic production of emergent qualities and systems should be employed in order to arrive at a dynamic concept of protest. In order to reflect the increasing complexity of society and the emergence of a stratified knowledge society, a multidimensional model of class that is structurally coupled to the concept of social movements is suggested.

In the face of complex and uncertain issues, one important goal of public participation in resource management and research is to foster communication and the inclusion of non-expert knowledge—thus the effective flow of information between project organisers and stakeholders. We compare different methods (instruments, tools) that were employed in the German–Austrian ‘PartizipA’ project to structure information flows in participatory processes. Depending on their goals and context, more or less ‘formalised’ and ‘participatory’ methods were applied, the most important being guided interviews, focus groups, agent-based modelling, nutrient modelling, cognitive mapping and group model building as well as the development of a common document. Two regional case studies, both concerned with European-induced institutional change, are portrayed in which the specific participatory methods were embedded. The Austrian case study involved the analysis and modelling of agricultural land use in the region of St. Pölten against the background of the reform of the Common Agricultural Policy, while the implementation of recent European water policy was the issue in the German agricultural region north of Osnabrück. Presenting both cases in their regional context, the applied methods are first described according to the logic of the entire respective process. Subsequently, the specific methods are systematically analysed and compared according to their objective, context and degrees of participation and formalisation. Finally, we evaluate all methods regarding their effectiveness in terms of goal attainment and their potential generalisation, seeking to respond to the question of when a particular method might best be used.  相似文献
