辽宁本溪地区条带状铁矿(BIF)与其玄武质火山围岩之间的时空关联非常密切.对于玄武质火山围岩高场强元素(HFSE)的研究表明:wNb/wTa比值(7.00~19.93)表现出明显的分异,而wZr/wHf比值(33.46~38.28)则变化不大;从弓长岭到南芬、歪头山样品的wNb/wTa比值变化具有明显的循序性.这种高场强元素的迁移和分异特征反映出俯冲作用与盆地演化之间的关联.研究区玄武质火山围岩(wNb/wYb)N比值大于1(1.21~18.45,平均2.72),这进一步表明,其形成的构造背景为陆内弧后盆地提供了有利于BIF形成的条件. 相似文献

During embryogenesis, multipotent progenitors within the single-layered surface epithelium differentiate to form the epidermis and its appendages. Here, we show that microRNAs (miRNAs) have an essential role in orchestrating these events. We cloned more than 100 miRNAs from skin and show that epidermis and hair follicles differentially express discrete miRNA families. To explore the functional significance of this finding, we conditionally targeted Dicer1 gene ablation in embryonic skin progenitors. Within the first week after loss of miRNA expression, cell fate specification and differentiation were not markedly impaired, and in the interfollicular epidermis, apoptosis was not markedly increased. Notably, however, developing hair germs evaginate rather than invaginate, thereby perturbing the epidermal organization. Here we characterize miRNAs in skin, the existence of which was hitherto unappreciated, and demonstrate their differential expression and importance in the morphogenesis of epithelial tissues within this vital organ. 相似文献

During the 1960s and 1970s population geneticists pushed beyond models of single genes to grapple with the effect on evolution of multiple genes associated by linkage. The resulting models of multiple interacting loci suggested that blocks of genes, maybe even entire chromosomes or the genome itself, should be treated as a unit. In this context, Richard Lewontin wrote his famous 1974 book The Genetic Basis of Evolutionary Change, which concludes with an argument for considering the entire genome as the unit of selection as a result of linkage. Why did Lewontin and others devote so much intellectual energy to the “complications of linkage” in the 1960s and 1970s? We argue that this attention to linkage should be understood in the context of research on chromosomal inversions and co-adapted gene complexes that occupied mid-century evolutionary genetics. For Lewontin, the complications of linkage were an extension of this chromosomal focus expressed in the new language of models for linkage disequilibrium. 相似文献

Holliday proposed a four-way DNA junction as an intermediate in homologous recombination, and such Holliday junctions have since been identified as a central component in DNA recombination and repair. Phage T4 endonuclease VII (endo VII) was the first enzyme shown to resolve Holliday junctions into duplex DNAs by introducing symmetrical nicks in equivalent strands. Several Holliday junction resolvases have since been characterized, but an atomic structure of a resolvase complex with a Holliday junction remained elusive. Here we report the crystal structure of an inactive T4 endo VII(N62D) complexed with an immobile four-way junction with alternating arm lengths of 10 and 14 base pairs. The junction is a hybrid of the conventional square-planar and stacked-X conformation. Endo VII protrudes into the junction point from the minor groove side, opening it to a 14 A x 32 A parallelogram. This interaction interrupts the coaxial stacking, yet every base pair surrounding the junction remains intact. Additional interactions involve the positively charged protein and DNA phosphate backbones. Each scissile phosphate that is two base pairs from the crossover interacts with a Mg2+ ion in the active site. The similar overall shape and surface charge potential of the Holliday junction resolvases endo VII, RuvC, Ydc2, Hjc and RecU, despite having different folds, active site composition and DNA sequence preference, suggest a conserved binding mode for Holliday junctions. 相似文献

Nephrotic syndrome, a malfunction of the kidney glomerular filter, leads to proteinuria, edema and, in steroid-resistant nephrotic syndrome, end-stage kidney disease. Using positional cloning, we identified mutations in the phospholipase C epsilon gene (PLCE1) as causing early-onset nephrotic syndrome with end-stage kidney disease. Kidney histology of affected individuals showed diffuse mesangial sclerosis (DMS). Using immunofluorescence, we found PLCepsilon1 expression in developing and mature glomerular podocytes and showed that DMS represents an arrest of normal glomerular development. We identified IQ motif-containing GTPase-activating protein 1 as a new interaction partner of PLCepsilon1. Two siblings with a missense mutation in an exon encoding the PLCepsilon1 catalytic domain showed histology characteristic of focal segmental glomerulosclerosis. Notably, two other affected individuals responded to therapy, making this the first report of a molecular cause of nephrotic syndrome that may resolve after therapy. These findings, together with the zebrafish model of human nephrotic syndrome generated by plce1 knockdown, open new inroads into pathophysiology and treatment mechanisms of nephrotic syndrome. 相似文献

An anomopod branchiopod crustacean that displays many structural similarities to the extant genera Scapholeberis and Megafenestra (family Daphniidae), fortuitously preserved in amber of Early Cretaceous age, is described. Although most of its appendages have been lost, preservation of several structures is vastly better than in most ‘orthodox’ fossils of these delicate animals. The well-preserved carapace suggests that, like extant representatives of these genera, but in no other daphniid, the animal exploited the surface film, beneath which it suspended itself by means of the straight ventral margins of its functionally bivalved carapace, each of which was armed with a row of close-set setae. Unlike the postabdomen of all the many extant anomopods, which is unsegmented, that of the fossil species, which was well preserved, was segmented; its distal half, or rather more, being clearly divided into four segments. Its topographically ventral margin was armed with pairs of very long spines (relatively longer than in any extant species), and the paired, hook-like terminal spines, clearly homologous with the claws of living species, were modified in a way that has no exact parallel in extant forms. The distal segment shows signs of annulations and may have had slight independent mobility. It is suggested that the long ventral spines of the postabdomen facilitated levering the animal forward over soft substrata. Its minute size reflects the fact that it is a first instar. Moreover, it was possibly squeezed out of the brood pouch prematurely when its parent became trapped in resin. The difficulty of assigning a place to the fossil in the phylogeny of the anomopods, all of which, including fossil forms of greater antiquity, have an unsegmented postabdomen, is briefly noted. The possibility that this attribute represents a very ancient atavism is tentatively suggested. 相似文献

A large amount (about three solar masses) of cold (18 K) dust in the prototypical type II supernova remnant Cassiopeia A was recently reported. It was concluded that dust production in type II supernovae can explain how the large quantities (approximately 10(8) solar masses) of dust observed in the most distant quasars could have been produced within only 700 million years after the Big Bang. Foreground clouds of interstellar material, however, complicate the interpretation of the earlier submillimetre observations of Cas A. Here we report far-infrared and molecular line observations that demonstrate that most of the detected submillimetre emission originates from interstellar dust in a molecular cloud complex located in the line of sight between the Earth and Cas A, and is therefore not associated with the remnant. The argument that type II supernovae produce copious amounts of dust is not supported by the case of Cas A, which previously appeared to provide the best evidence for this possibility. 相似文献
